The present book results from the cooperation of a bioehernist and morphologists: the revision and unified treatment of available data is the primary object of our work. A comprehensive review of all the available literature is therefore beyond the scope of this volume. lt is intended to be a manual to be used in the laboratory, with convenient guidelines for practical work. Plant microbodies have been treated by B. GERHARDT in Volume 5 of this series. The discovery of fatty acid ß-oxidation in animal peroxisomes has proved once more that plant and animal microbodies are members of the same family of organelles. lt provided new insights into the physiological meaning of these particles; our understanding of these "classical" cell organelies is undergoing continual alteration and development. De Duve's laboratory, peroxisomes - organelles which are present in nearly all eukaryotic cells - are still not fully understood. More than 40 peroxisomal enzymes catalyzing a variety of reactions have been characterized. Moreover, peroxisomes can be regarded as toxicological indicators: several compounds, including hypolipemic drugs, plasticizers or pesticides trigger their proliferation. This proliferation may lead to hepatocarcinogenesis in rodents. An account of mitochondriopathis is given in the chapter on the mitochondrion rather than in a separate one. The subject matter of this part of the volume shows quite clearly that the interdisciplinary approach to the study of organelles has shed considerable light on the nature of the mechanisms underlying the etiology and pathobiology of many of these disorders. As an example, mutations in the genes encoding integral membrane proteins are found to lead to disturbances in peroxisome assembly. It is also interesting and significant that mistargeting of protein is now thought to be another cause. It will be revealing to see whether mistargeting is the result of mutations in the genes encoding chaperones.The second part of the volume is concerned with the extracellular matrix. It sets out to show that a vast body of new knowledge of the extracellular matrix is available to us. Take for example the integrin family of cell adhesion receptors. It turns out that integrins play a key role not only in adhesion but also in coupling signals to the nucleus via the cytoskeleton. As for fibronectins, they seem to link the matrix with the cytoskeleton by interacting with integrins.Collagen molecules are dealt with in the last two chapters. The boundaries of collagen in disease are defined by drawing a clear line of demarcation between systemic connective tissue disorders (e.g., scleroderma), better known as autoimmune diseases, and the heritable, and the heritable diseases such as osteogenesis imperfect and the Marfan syndrome. This classification takes into account a second group of acquired disorders of collagen forming tissues in which regional fibrosis is the hallmark. Liver cirrhosis and pulmonary fibrosis are prime examples.The decision to place Volumes 2 and 3 before those dealing with cell chemistry was not easily made. It was based on the view that most students will have had an undergraduate course in biochemistry of cell biology or both courses, and that they could go to Volumes 4-7 in which the subject of cell chemistry is covered, and then return to Volumes 2 and 3.

Book excerpt: This book presents the proceedings of the meeting on "Lipid Storage Disorders" which took place in Toulouse, France, in September 14-18, 1987 and which was set up as a joint NATO Advanced Research Workshop and INSERM International Symposium. The meeting probably was the first truly international symposium devoted entirely to basic as well as applied aspects of lipid metabolism as related to the lipid storage disorders. Participants came from Europe, USA, Israel and Japan, and presented data on research in molecular biology and genetics, enzymology, cell biology as well as medical and epidemiological aspects of normal and pathological lipid metabolism. In the latter case, special attention was directed to the lysosomal B-glucosidase in relation to Gaucher disease and to sphin gomyelinase in relation to Niemann-Pick disease, and "two round" table discussions were devoted to the two respective items. But research on many other lipidoses was presented, as lectures or posters and avidly discussed. Other topics presented in special sessions were drug induced lipidoses and peroxisomal disorders. The meeting ended with a session devoted to medical aspects of disorders of lipid metabolism. The symposium which included about 50 lectures and close to 70 posters had an intense scientific character superimposed on a most pleasant and collegial atmosphere. Ample time was provided to personal discussions ; lodging of the participants in the students' residence located near the meeting place, permitted further contacts between them.
