Book excerpt: This thesis describes original research aimed at the development of highly efficient polymer functionalization strategies by introducing orthogonal chemistry within polymeric systems. The primary hypothesis of this thesis is that the use of click chemistries or noncovalent interactions can provide new and easy pathways towards the synthesis of highly functionalized polymers thereby addressing the shortcomings of traditional covalent functionalization approaches. To verify the hypothesis, the work presented in the following chapters of this thesis further explores previous methods of either covalent or noncovalent polymer functionalization described in Chapter 1. Chapters 2 and 3 present advanced methods of covalent polymer functionalization based on high-yielding and orthogonal click reactions: 1,3-dipolar cycloaddition, hydrazone formation, and maleimide-thiol coupling. All three click reactions employed can be orthogonal to one another and conversions can be quantitative, leading to the easy and rapid synthesis of highly functionalized polymers without interference among functional handles along the polymer backbones.

A conclusion is drawn inferring that upper limits in molecular design are inevitable, arising as a direct consequence of the predominance of covalent strategies in the field. To address these concerns, the?universal polymer backbone? (UPB) concept has been hypothesized. A UPB has been defined as any copolymer, side-chain functionalized with multiple recognition elements that are individually capable of forming strong, directional, and reversible non-covalent bonds. Non-covalent functionalization of these scaffolds can lead to the formation of a multitude of new polymer structures, each stemming from a single parent or?universal polymer backbone?. To prepare such a UPB, isomerically pure exo-norbornene esters containing either a PdII SCS pincer complex or a diaminopyridine residue were synthesized, polymerized, and copolymerized via ROMP. All polymerizations were living under mild reaction conditions. Kinetic studies showed that the kp values are highly dependent upon the isomeric purity but completely independent of the terminal recognition units. Non-covalent functionalization of these copolymers was accomplished via 1) directed self-assembly, 2) multi-step self- assembly, and 3) one-step orthogonal self-assembly. This system shows complete specificity of each recognition motif for its complementary unit with no observable changes in the association constant upon functionalization. To explore potential applications of this UPB concept, random terpolymers possessing high concentrations of pendant alkyl chains and small amounts of recognition units were synthesized. Non-covalent crosslinking using a directed functionalization strategy resulted in dramatic increases in solution viscosities for metal crosslinked polymers with only minor changes in viscosity for hydrogen bonding motifs. The crosslinked materials were further functionalized via self-assembly by employing the second recognition motif, which gave rise to functionalized materials with tailored crosslinks. This non-covalent crosslinking/functionalization strategy could allow for rapid and tunable materials synthesis by overcoming many difficulties inherent to the preparation of covalently crosslinked polymers. Finally, the current status of the UPB concept is reviewed and methodological extensions of the concept are suggested. Evaluation of how UPBs may be used to optimize materials and their potential use in fabricating unique electro-optical materials, sensors, and drug delivery vesicles are explored.

The discussion then focuses more explicitly on transition metal-based motifs and methodologies that are employed in polymer functionalization and continues with a more detailed overview of this field. The primary hypothesis that is addressed herein is that combining the versatility and strength of metal-ligand interactions with the efficiency and functional group tolerance of ROMP comprises a useful method of generating a variety of functionalized polymers and materials via side-chain metal coordination. Thus, the goal is to test this hypothesis by synthesizing functionalized polymers with a range of useful properties to demonstrate the relevance and importance of this methodology, by employing several different strategies to show the synthetic ease by which the materials can be realized. The strategies and methods discussed in the synthesis of side-chain functionalized polymers are divided into three subgroups: (1) pre-polymerization functionalization, in which all of the modifications take place on the monomer with polymerization as the last step, (2) post-polymerization functionalization, in which the polymer itself is subsequently modified, and (3) combinations of the first two strategies. It is shown that useful functional polymers and materials can be synthesized by any of the above strategies, and representative examples of each are given in both the introduction and in the body of work presented. Modes of functionalization are all based on transition metal coordination, and polymerizations are primarily carried out via ROMP. Metal coordination is shown to be a useful technique for functionalizing polymers, to creating supported emissive complexes, to modulating solution viscosity. Finally, conclusions are drawn regarding the various strategies presented herein, and potential future directions are discussed.

Chemo-enzymatic methods are an alternative strategy to increase the diversity of functional groups in polymeric materials. Specifically, enzyme-catalyzed polymer functionalization carried out under solventless conditions is a great advancement in the design of green processes for biomedical applications, where the toxicity of solvents and catalyst residues need to be considered. Enzymes offer several distinct advantages, including high efficiency, catalyst recyclability, and mild reaction conditions. This reseach aimed to precisely functionalized polymers using two methods: enzyme-catalyzed functionalization via polymerization and chemo-enzymatic functionalization of pre-made polymers for drug delivery. In the first method, well-defined poly(caprolactone)s were generated using alkyne-based initiating systems catalyzed by CALB. Propargyl alcohol and 4-dibenzocyclooctynol (DIBO) were shown to efficiently initiate the ring opening polymerization of e-caprolactone under metal free conditions and yielded polymers with Mn ~4 to 24 KDa and relatively narrow molecular mass distribution. In the second methodology, we present quantitative enzyme-catalyzed transesterification of vinyl esters and ethyl esters with poly(ethylene glycol)s (PEG)s that will serve as building blocks for dendrimer synthesis, followed by introducing a new process for the exclusive gamma-conjugation of folic acid. Specifically, fluorescein-acrylate was enzymatically conjugated with PEG. Additionally, halo-ester functionalized PEGs were successfully prepared by the transesterification of alkyl halo-esters with PEGs. 1H and 13C NMR spectroscopy, SEC and MALDI-ToF mass spectrometry confirmed the structure and purity of the products."--Website of ETD.

Introducing Sacrificial Synthesis, many of the limitations of ruthenium-catalyzed ROMP have been overcome. Here, the living ROMP polymer to be functionalized was turned into a diblock copolymer by polymerizing dioxepine monomers onto the desired first polymer block. The second block was then later removed to leave "half-a-dioxepin", i.e. exactly one hydroxyl group, at the chain-end. The efficiency of Sacrificial Synthesis is also studied. Thiol groups were also placed by a sacrificial strategy based on cyclic thioacetals. 2-Phenyl-1,3-dithiepin could be polymerized and subsequently cleaved by hydrogenation with Raney-Nickel. The presence of thiol groups on the chain end has been proven by chemical means (derivatization) and by coating gold-nanoparticles. The second strategy, vinyl lactone quenchingv is a termination reaction based on vinyl esters. After a metathesis step, an inactive Fischer-type carbene is formed. Such acyl carbenes are unstable and self-decompose to set an inactive ruthenium complex and the functional group free without changing the reaction conditions. The two compounds vinylene carbonate and 3H-furanone gave rise to the placement of aldehydes and carboxylic acids at the polymer chain ends without the necessity to perform any deprotection steps after the functionalization. The development of those two functionalization methods led to several applications. By reacting hydroxyl-functionalized ROMP-polymers with norbornene acid, macromonomers were formed which were subsequently polymerized to the respective graft-copolymers. Also, the derivatization of the same functionalized polymers with propargylic acid gave rise to alkyne-functionalized polymers which were conjugated with azides. Furthermore, "ugly stars", i.e. long-chain branched structures were synthesized by polycondensation of ABn-type macromonomers and telechelic polymers were accesse.
