Book excerpt: Physicochemical experimental techniques combined with the specificity of a biological recognition system have resulted in a variety of new analytical devices known as biosensors. Biosensors are under intensive development worldwide because they have many potential applications, e.g. in the fields of clinical diagnostics, food analysis, and environmental monitoring. Much effort is spent on the development of highly sensitive sensor platforms to study interactions on the molecular scale. In the first part, this thesis focuses on exploiting the biosensing application of nanoporous gold (NPG) membranes. NPG with randomly distributed nanopores (pore sizes less than 50 nm) will be discussed here. The NPG membrane shows unique plasmonic features, i.e. it supports both propagating and localized surface plasmon resonance modes (p SPR and l-SPR, respectively), both offering sensitive probing of the local refractive index variation on/in NPG. Surface refractive index sensors have an inherent advantage over fluorescence optical biosensors that require a chromophoric group or other luminescence label to transduce the binding event. In the second part, gold/silica composite inverse opals with macroporous structures were investigated with bio- or chemical sensing applications in mind. These samples combined the advantages of a larger available gold surface area with a regular and highly ordered grating structure. The signal of the plasmon was less noisy in these ordered substrate structures compared to the random pore structures of the NPG samples. In the third part of the thesis, surface plasmon resonance (SPR) spectroscopy was applied to probe the protein-protein interaction of the calcium binding protein centrin with the heterotrimeric G-protein transducin on a newly designed sensor platform. SPR spectroscopy was intended to elucidate how the binding of centrin to transducin is regulated towards understanding centrin functions in photoreceptor cells.

Book excerpt: Point-of-care (POC) devices are compact, mobile and fast detection platforms expected to advance early diagnosis, treatment monitoring and personalized healthcare, and revolutionize today's healthcare system, especially in remote areas. The need for POC devices strongly drives the development of novel biosensor technology. Building a small, fast, simple, and sensitive platform for biomolecule detection is a challenge that relies on the integration of multiple fields of expertise and engineering. Optical nanoresonators have shown great promise as label-free biosensors because of direct light coupling and sub-wavelength sensing modes. Metallic nanoresonators with localized surface plasmon resonances (LSPR) are already well studied and were proven a solid alternative to the commercialized surface plasmon resonance (SPR) sensors. More recently, dielectric nanoresonators have also gained traction due to the reduced losses and the ability to manipulate both the electric and magnetic components of the incident light. In this thesis, we advance the field of biosensing and use optical nanoresonators as operative platforms relevant for disease diagnosis and treatment monitoring. By combining different optimized optical nanoresonators, both metallic and dielectric, with state-of-the-art microfluidics and surface chemistry, we have developed and tested several detection platforms. We first focused on developing a microfluidic lab-on-chip device for multiplexed biosensing utilizing the LSPR of gold nanoresonator arrays. By simultaneously tracking the extinction of 32 sensor arrays, we demonstrated multiplexed quantitative detection of four breast cancer markers in human serum. We showed that with well-optimized immunoassays, a low limit of detection (LOD) can be reached, paving the way towards clinically-relevant POC devices. Additionally, we implemented silicon nanoresonators supporting Mie resonances into functional and clinically-relevant applications. By integrating several arrays of Si nanoresonators with state-of-the-art microfluidics, we demonstrated their ability to detect cancer markers in human serum with high sensitivity and high specificity. Furthermore, we showed that the fabrication of Si nanoresonator array using low cost and scalable projection lithography leads to sufficiently low limits of detection, while enabling cheaper and faster sensor production for future POC applications. We also investigated the respective role of electric and magnetic dipole resonances and showed that they are associated with two different transduction mechanisms: resonance redshift and extinction decrease. Our work advances the development of future point-of-care sensing platforms for fast and low cost health monitoring at the molecular scale.

From the very existential belief of the origins of the universe to its role in the evolution of life on earth, light has been inextricably woven into every aspect of our lives. I am grateful to Springer-Verlag and Thomas Scheper for this invitation to organize this volume that continues to expand the use of light to create next generation sensing applications. Indeed, the very act of expanding the frontiers of learning and knowledge are referred to in many languages and cultures as enlightenment. Early optical instruments relied largely on simple combinations of mirrors, prisms and lenses. With these simple devices, substantial progress was made in our understanding of the properties of light and of its interactions with matter. Things got more complicated with the evolution of optical instruments in labo- tory use. Early systems used bulky and expensive hardware to generate light, split it into the desired wavelengths and finally collect it for analysis. The discovery of the laser pushed the technology further, but did not do much to make its adoption more widespread as the lasers themselves were large and required substantial el- trical power to operate. The true revolution is just beginning. Advances in mic- electronics have resulted in the possibility of truly low-cost (using the consumer electronics industry as a parallel) devices that exploit optical measurement technology.
