Book excerpt: Potato virus Y (PVY) infects a wide host range mainly within the Solanaceae and is distributed worldwide. PVY is transmitted by more than 40 aphid species in a non persistent manner. Isolates of the PVY species are highly variable at biological, serological and molecular levels. Epidemiological studies have highlighted the emergence of distinct potato PVY variants able to induce necroses on potato tubers. Due to the lack of efficient resistance to PVY isolates inducing necrotic symptoms in cultivated varieties and the plant-to-plant transmission of isolates through the daughter tubers, PVY has become the most economically important virus for the potato industry. The review offers an overview of several decades of research on PVY but also focuses on the latest data obtained by expert on PVY worldwide on the biological characteristics of PVY, interactions between aphids-hosts, its evolution and management. Identified knowledge gaps to understand further PVY biology will be discussed.

Discusses the mechanisms of innate resistance in strains of tobacco, tomato, and cowpea; various aspects of induced resistance, including the characterization and roles of the pathogenesis-related proteins; antiviral substances and their comparison with interferon; and cross-protection between plant virus strains. Also presents several papers which evaluate the status of genetic engineering as it relates to breeding resistant plants. Among these are discussions of the potential use of plant viruses as gene vectors, gene coding for viral coat protein, satellite RNA, and antisense RNA, and practical issues such as the durability of resistant crop plants in the field.

PVY is composed of various strains some with a recombinant genome such as PVYNTN, PVYNE-11 and some with no recombinant genome such as PVYEu-N and PVYO. Pathotype severity is directly correlated to the type of strain that infects the potato and the type of resistance genes, known as hypersensitive genes (HR), that the specific cultivar possesses to counteract the virus infection. This dissertation will focus on three different projects that took place in different regions of North America and will explore and expand on the previously mentioned concepts. 1) Biological and molecular characterization of hypersensitive reaction genes in two popular Mexican potato cultivars Alfa and Mondial using various strains of potato virus Y. 2) Genetic diversity of the NE-11 strain potato virus Y. 3) Full genome sequencing of three potato virus Y isolates found in the commercial field and a wild solanum in the state of Jalisco Mexico.

At the same time, others are concerned that the technology is not reaching its potential to improve human health and the environment because of stringent regulations and reduced public funding to develop products offering more benefits to society. While the debate about these and other questions related to the genetic engineering techniques of the first 20 years goes on, emerging genetic-engineering technologies are adding new complexities to the conversation. Genetically Engineered Crops builds on previous related Academies reports published between 1987 and 2010 by undertaking a retrospective examination of the purported positive and adverse effects of GE crops and to anticipate what emerging genetic-engineering technologies hold for the future.
