Glutathione-S-transferases (GSTs) play a role in the placenta by protecting the fetus from toxicants and oxidative stress. A soluble form of GST, GST pi (GSTP1), accounts for a significant amount of placental GST enzyme activity. Previous studies indicate that the Ile105Val polymorphism in GSTP1 affects susceptibility to pathologic conditions of pregnancy such as preeclampsia and recurrent pregnancy loss. GSTP1 also detoxifies environmental carcinogens such as polyaromatic hydrocarbons (PAHs). This study investigates the relationship between placental levels of GST pi, enzyme activity, and genetic polymorphisms of biotransformation enzymes CYP1A1 exon 7, GSTP1, GSTM1; and PAH-DNA adduct measurements. Maternal venous and umbilical cord blood and placental tissue were collected from 54 consenting obstetric patients from Tijuana, Baja California (BC), Mexico (maternal age mean 25.8, range 16--39 years). Placental GST pi levels were measured via Western blots; GST activity was measured against CDNB; and genetic polymorphisms were assessed by PCR and restriction enzyme digestion. Relative adduct levels were obtained by 32P-postlabeling. A 30-fold variation in enzyme activity was found and activity ranged from 21--623 nmol/min/ml. Activity and levels of GSTPi were modestly correlated (r = 0.359, p = 0.13). No difference was found in placental GSTPi activity associated with the GSTP1 Ile105/Ile105 genotype vs. the Val105/Val105 genotype. An increased activity was however observed in placental supernatant from GSTP1 Ile105 positive mother/baby pairs that were concordant for GST mu negative genotype vs. pairs with GST mu positive genotype, though numbers were very small (n = 6 vs. n = 31, p = 0.024); median PAH-DNA adduct measurements were 10.4 RAL x 109 nucleotides. Placental variability in levels and activity of the protective enzyme GST pi can be substantial between Hispanic women. GST pi levels and genetic variation at GSTP1 ile105val does not appear to explain variability in placental enzyme activity and PAH-DNA adduct measurements. Further studies are necessary to elucidate the impact of placental biotransformation enzymes on pregnancies which are more at risk from endogenous and environmental toxicants.

These landmark changes in policy have impacted cannabis use patterns and perceived levels of risk. However, despite this changing landscape, evidence regarding the short- and long-term health effects of cannabis use remains elusive. While a myriad of studies have examined cannabis use in all its various forms, often these research conclusions are not appropriately synthesized, translated for, or communicated to policy makers, health care providers, state health officials, or other stakeholders who have been charged with influencing and enacting policies, procedures, and laws related to cannabis use. Treatment differs widely depending on the type: chronic hypertension, albuminuric hypertension, and hypertensive crises during pregnancy. One paper suggests that to prove any environmental exposure to a particular substance affecting pregnancy, the exact timing of exposure must be established, large samples are necessary, possibly on a national or international scale. The paper cites as example the four-year documentation period of the 50% to 80% incidence of malformations due to thalidomide. General medicine practitioners, obstetricians, gynecologists, and researchers dealing with pharmacology, pharmocokinetics, toxicology, or embryology will find the collection valuable.
