Book excerpt: During May 21-June 1 1990, the eleventh course of the International School of Pure and Applied Biostructure, a NATO Advanced Study Institute, was held at the Ettore Majorana Center for Scientific Culture in Erice, Italy, co-sponsored by the Italian Ministry of Universities and of Scientific and Technological Research, the North Atlantic Treaty Organization, the Italian National Research Council, the Sicilian Regional Government and Technobiochip. The subject of the course was "Molecular Basis of Human Cancer" with participants selected worldwide from 15 different countries. The purpose of the course was to address, in a tutorial and structural fashion, the molecular basis of human cancer, including the mechanism of signal transduction in mammalian cells, the genetic mechanism of malignant transformation in man, growth factors, hormone receptors, cell membrane and cytoskeleton, and DNA high order structure. The course had this as its major objective and the resulting book reflects it. The participants were exposed to a critical evaluation of current knowledge about cancer and to some of the key problems that remain as stumbling blocks to our eventual understanding of this important biological and medical problem. Through the media of formal and informal lectures, workshops, symposia and informal discussions, a select group of interested young and senior scientists were acquainted with many of the aspects of human cancer.

Understanding ethanol action presents a special and functional diver challenge because of its molecular simplicity sity. In fact, the ability for alcohol to disrupt cellular function is at tributed to its cellular injury without regard to an apparent specific mechanism of action. Nevertheless, the key to an effective treatment to this problem is through research into understanding the mechan isms underlying how ethanol interacts with cells and membranes. This book is the result of a cooperative effort among scientists from many nations who met in a symposium in Taipei, Taiwan, ROC, July 1988. The focus of this book is on experimental ap proaches to better understand the molecular mechanisms of ethanol on the biological system. These recent advances in the examination of the alcohol effects on cellular function are divided into four sec tions. The first section addresses specific actions of ethanol on the central nervous system. The second section is directed to the use of cell cuI tures in ethanol research and the usefulness of cell cultures in examination of the effects of ethanol in vitro.

In most cases, these interactions involve and are driven by proteins, which are the main conductors of any life process within the organism. The physical nature of these interactions is still not well known. This book presents an entirely new approach to analysis of biomolecular in teractions, in particular protein-protein and protein-DNA interactions, based on the assumption that these interactions are electromagnetic in nature. This new ap proach is the basis of the Resonant Recognition Model (RRM), which was devel oped over the last 15 years. Certain periodicities within the distribution of energies of delocalised electrons along a protein molecule are crucial to the protein's biological function, i.e. inter action with its target. If protein conductivity were introduced, then charges mov ing through the protein backbone might produce electromagnetic irradiation or ab sorption with spectral characteristics corresponding to energy distribution along the protein. The RRM is capable of calculating these spectral characteristics, which we hypothesized would be in the range of the infrared and visible light. These characteristics were confirmed with frequency characteristics obtained ex perimentally for certain light-induced biological processes.

Nutrients in Cancer Prevention and Treatment also contains several studies on the role of diet and vitamins in cancer treat ment. There are very few books that have reviewed laboratory and clinical studies and the role of vitamins in cancer treatment. There are preliminary data suggesting that daily supplementa tion with high doses of certain vitamins in combination with conventional therapeutic agents may enhance their growth inhibitory effects on tumor cells, and may protect normal tissues against some of their toxic effects. This book is unique in the sense that several articles have discussed the mechanisms of action of individual vitamins on cellular and molecular parameters. It is very exciting to note that some of the vitamins inhibit protein kinase C activity, increase the production of certain growth factors, and modulate the expression of a number of oncogenes. These studies, at least in part, offer rationales for the cancer protective effects of vitamins.

Book excerpt: During the last 10 years, the role of specific nutrients in cancer prevention and cancer treatment has been the subject of intense basic, preclinical, and clinical research. At present, the major focus of nutri tional oncology is on the mechanisms of carcinogenesis and their modification by nutrients and on cancer prevention studies in animals and humans. Some human epidemiological studies have confirmed the hypothesis, developed on animals, that there is an inverse relation ship between the intake and/or level of !3-carotene, vitamin A vita min E, or vitamin C and the risk of cancer, whereas others have shown no such relationship. This is not unexpected, since the protective effect of individual nutrients may be too small to be detected by epidemiological methodologies in which a single vitamin or mineral is considered as one variable. Conclusive evidence regarding the role of nutrients in human cancer prevention will come from a well designed human intervention study using one or more nutrients in a population that has a high risk of developing cancer. The involvement of specific nutrients in the regulation of protooncogene expression has just begun. Also, some of the results of human intervention trials are beginning to yield interesting results. A large number of interna tional scientists from various disciplines, including cell biology, mo lecular biology, nutritional oncology, epidemiology, and public health, reviewed and discussed their most recent findings. More recent investigations on this virus have focused on the identity of the viral genes responsible for the different disease mani festations observed following viral infection. It is hoped that by thorough investigation of this virus, clues to how cancer develops from a normal cell will surface. In addition, many of the gene products are now being exploited for the development of new and more sensitive tests for the diagnosis and clinical management of individu als with EBV -associated diseases. Thus, studies on this virus continue to provide new information of importance to our understanding of the malignant process. In an effort to attract both basic and clinical scientists to the same meeting for purposes of scientific exchange and fostering a closer interaction between these individuals, a series of international symposia was initiated in 1984. The first meeting was held in Loutraki, Greece, and was attended by approximately 100 participants. The second international symposium was held in St. Petersburg, Florida, in 1986, and was attended by approximately twice as many partici pants as attended the Loutraki meeting.

Role of activated and suppressor oncogenes in neoplastic trans formation. The workshop was organized by J. S. Rhim and A. Dritschilo (cochairmen), G. Jay, J. little, M. McCormick, R Tennant, and R R Weischelbaum. There were 32 speakers, 30 poster presentations, and about 190 participants.
