Book excerpt: Since the discovery of cisplatin as a potent anticancer agent, transition metal complexes have played a significant role in the development of new anticancer compounds. The anticancer potential of cobalt complexes has been extensively studied over the last three decades, and much time has been devoted to understanding their mechanisms of action. After understanding the Co(III)/Co(II) reduction activation mechanism, several Co(III) complexes of bioactive anticancer agents have been investigated as potential hypoxia activated prodrugs. Furthermore, after the initial use of nitrogen mustard, several bioactive compounds and their analogues have been investigated as ligands to develop hypoxia activated Co(III) complexes. This thesis describes syntheses and characterisation of Co(III) complexes that have been designed using structural analogues of bioactive anticancer compounds. Chapter 1 gives an overview of the prodrug strategy in cancer therapy, prodrug design using a trigger and an effector unit, and different mechanisms for prodrug activation. Several examples of Co(III) complexes are given, which have been investigated as potential prodrugs to target tumour hypoxia. There are three different parts to this research. Chapters 2, 3 and 4 describe the syntheses and characterisation of Co(III) complexes designed using ligands, which are structural analogues of bioactive anticancer compounds. Chapter 2 describes syntheses of dipeptide analogues of bortezomib, a proteasome inhibitor that is currently being used for the treatment for multiple myeloma. The Co(III)-dipeptide complexes were also analysed for their possible use as hypoxia activated prodrugs in Chapter 2. Chapter 3 includes the continuation to the previous investigation by Ware et al. on Co(III)-cyclen complex using 8-hydroxyquinoline (8HQ) as a model cytotoxin. Efforts were made to establish a synthetic route to introduce a porphyrin derivative into prodrug design, with the aim of improving tumour cell targeting properties of the prodrug complex. Chapter 4 describes syntheses of two polypyridyl carboxamide ligands, PDPA and MID(AMP)2, and syntheses of their Co(III) complexes. PDPA and MID(AMP)2, are structural analogues of PMAH, Py3PH2, which are carboxamide ligands containing a similar metal-binding domain as bleomycins. The experimental procedures are detailed in Chapter 5.

Book excerpt: This thesis describes the synthesis and characterization of a number of polyborate compounds containing transition metal complex cations; most of these compounds contain 6-membered B3O3 boroxole rings within their structures.A total of twenty-six transition metal complex polyborate salts are reported: one contains the triborate monoanion, [B3O3(OH)4]-, three contain the tetraborate dianion [B4O5(OH)4]2-, twelve contain the pentaborate anion, [B5O6(OH)4]-, three contain the hexaborate dianion, [B6O7(OH)6]2- and five contain heptaborate anions (three of which are [B7O9(OH)5]2-, and two are [B7O9(OH)6]3-). Additionally, two compounds contain two isolated polyborate anion species (octaborate dianion [B8O10(OH)6]2- with pentaborate anion [B5O6(OH)4]-, and triborate anion [B3O3(OH)4]- with [B5O6(OH)4]-). The crystal structures of fifteen salts containing these polyborate anions are reported: [Co(en)3][B5O6(OH)4][B8O10(OH)6]∙5H2O (6), [Co(NH3)6]2[B4O5(OH)4]3∙11H2O (9), s-fac-[Co(dien)2][B7O9(OH)6]∙9H2O (11), [Co(diNOsar)]2[B3O3(OH)4]Cl5∙4.75H2O (12), [Cu(en)2][B5O6(OH)4]2∙2H2O (20), [Cu(pn)2{B5O6(OH)4}][B5O6(OH)4]∙4H2O (22), [Cu(TMEDA){B6O7(OH)6}]∙6H2O (23), [Cu(dac)2(H2O)2][Cu(dac)2][B7O9(OH)5]2∙4H2O (24), [Cu(N,N-dmen)2(H2O)][B5O6(OH)4]2·3H2O (27), [Cu(N,N-dmen){B6O7(OH)6}]·4H2O (28), [Ni(en){B6O7(OH)6}(H2O)2]∙H2O (37), [Ni(AEN)][B5O6(OH)4]∙H2O (38), [Ni(dac)2(H2O-)2][Ni(dac)2][B7O9(OH)5]2∙4H2O (39), [Ni(hn)2][B5O6(OH)4]2 (40), and s-fac-[Ni(dien)2][B5O6(OH)4]2 (41). All the synthesized compounds reported were characterized using spectroscopic (IR, multi-element NMR) and analytical (melting point, elemental analysis, magnetic susceptibility, thermal analysis, powder X-ray diffraction) techniques.The solid-state structures of transition metal complex polyborate salts all display multiple cation-anion H-bond interactions and these undoubtedly play a major role in the energetics of engineering these structures. For example, in compound [Co(NH3)6]2[B4O5(OH)4]3∙11H2O (9), fourteen of the 18 amino hydrogen atoms of the [Co(NH3)6]3+ cation are involved in secondary coordination to anions via H-bonds.Two unique polyborate anions, [B8O10(OH)6)]2- and [B7O9(OH)6]3- have been prepared and characterized in [Co(en)3][B5O6(OH)4][B8O10(OH)6]∙5H2O (6) and s-fac-[Co(dien)2][B7O9(OH)6]∙9H2O (11), respectively.viiThe reaction of transition metal complex cations with boric acid in different ratiosproduced polyborate compounds with different polyborate anions e.g. reaction of [Cu(en)2]2+with boric acid in 1:10 and 1:5 ratios produce polyborate compounds with pentaborate(1-) andtetraborate(2-) anions, respectively.New polyborate anion structural architectures are identified in this thesis: (i) thepentaborate(1-) anion in [Cu(pn)2{B5O6(OH)4}][B5O6(OH)4]∙4H2O (22) acts as a monodentateligand coordinated to the Cu(II) cation, (ii) the hexaborate(2-) anion in[Cu(TMEDA){B6O7(OH)6}]∙6H2O (23) and [Cu(N,N-dmen){B6O7(OH)6}]·4H2O (28)coordinates as a tridentate ligand with the Cu(II) cations, and (iii) the hexaborate(2-) anion in[Ni(en){B6O7(OH)6}(H2O)2]∙H2O (37) coordinates as a bidentate ligand with the Ni(II) cation.This is the first time that such coordination mode (i) and (iii) have been observed.

Unusual multinuclear Fe(II) and Co(II) complexes with the basic dinuclear unit M2X2[[superscript{tBu}]NON] (X= Br, M= Fe (1), Co (2); X= Cl, M= Fe (3), Co (4)), ([[superscript{tBu}]NON]2−= [Me3CN(SiMe2)]2O2−) were synthesized and characterized. The reduction reaction of 3 with KC8 and addition of CO and dmpe (Me2P(CH2)2PMe2) generated {Fe2[[superscript{tBu}]NON]}2 and, with dmpe also FeCl2(dmpe)2. However, the addition of 1,3-bis-(2,4,6-trimethylphenyl)imidazol-2-ylidene (NHC) carbene to 3 resulted in the new {[NHC]FeCl2}2. Alkyl for halide metathesis via the reaction of LiCH2SiMe3 in 1-4 generated rare high-spin alkyl/halide {[[superscript{tBu}]NON]M2X(CH2SiMe3)2}2 (8-11) complexes and unusual high-spin dialkyl iron and cobalt complexes of the form {M2(CH2SiMe3)2[[superscript{tBu}]NON]} (M = Fe (6) and Co (7)); reaction with MeLi to form the dimethyl analogue was not successful, while addition of one equivalent of methyl reagent per dinuclear unit in 3 and 4 resulted in {Fe2Br(Me)[[superscript{tBu}]NON]}2 and {Co2Cl(Me)[[superscript{tBu}]NON]}2. Neither 6 nor 7 act as polymerization catalysts for ethylene; addition of B(C6F5)3 as a cocatalyst was found to further hinder any activity of 6 and 7 by the formation of {Co2(C6F5)2[[superscript{tBu}]NON]} and {Fe2Cl(C6F5)[[superscript{tBu}]NON]}2. Upon exposing 6 to excess CO, a rare dicarbamoyl trinuclear iron complex was obtained, which results from CO binding, isocarbonyl binding and Fe-C and Fe-N CO insertions. Attempts towards obtaining high oxidation state iron and cobalt complexes showed that the [NON] ligand did not support such systems. Addition of benzyl bromide to {Fe[[superscript{Me3Ph}]NON]}2 resulted in {FeBr[[superscript{Me3Ph}]NON]}2 and reaction of {FeCl[[superscript{tBu}]NON]}2 with dmpe led to a rare tetrahedral iron(III) complex {FeCl[[superscript{tBu}]NON]}2([mu]-Me2PCH2CH2PMe2). To overcome potential difficulties with limited Si-N bond stabilities, carbon-backbone diamido {M[[superscript{iPr}]NN'N]}2 and {M[[superscript{iPr}]NO'N]}2 (M= Fe, Co) [MeN((CH2)2N−iPr)2] ([[superscript{iPr}]NO'N]2−) and [O((CH2)2N−iPr)2] ([[superscript{iPr}]NN'N]2−) were synthesized, characterized and compared with similar diamido complexes.
