Book excerpt: Abstract: The experimental technique of mass spectrometric tracer pulse chromatography was used to investigate the uptake of RPLC eluents and a series of test analytes by a C (18subscript- ) bonded packing over the full element composition range. The experiments were carried out with binary eluents consisting of aqueous mixtures of acetonitrile, methanol, or tetrahydrofuran, as well as ternary eluents composed of acetonitrile, methanol, and water. The primary experimental data obtained for eluent components were in the form of excess volumes. The excess volume data were then used to determine the absolute volume of each eluent component in the stationary phase as a function of eluent composition. The absolute sorption isotherms of the eluent components were indirectly estimated by a combination of techniques specific to a limited eluent composition range. Regression analysis of the non-stationary inflection point of the excess isotherms provided an estimate of the volume of eluent sorbed by the stationary phase for organic-rich eluents. Three different assumptions were used and evaluated for the water-rich eluent compositions. The calculated void volume data were compared with the retention volume of thiourea, uracil, and acetone, solutes commonly used to estimate the volume of the mobile phase. The experimental results showed that water and thiourea did provide an accurate measure of the mobile phase volume but only over a limited range of eluent composition. Deuterated water was chosen as the unretained dead time marker for water rich eluents. With data for the water-rich eluents, the regression results from excess isotherm data was used to estimate the absolute volume of eluent sorbed by the stationary phase over the full eluent composition range. Retention factors for a series of test solutes were calculated with the thermodynamic and kinetic void volumes in order to determine the influence of immobilized eluent. It was hypothesized the retention factors of the test analytes would increase as the absolute amount of organic component in the eluent increased. However, retention factors continually decreased. Therefore, the effect of the uptake of eluent on the retention of test solutes appeared to be minimal for this particular set of test analytes. The experimental technique of mass spectrometric tracer pulse chromatography was shown to be a valid method for the investigation of the uptake of eluent by a C (subscript- ) bonded stationary phase, even with ternary eluent systems.

Different reagents, such as alkylammonium salts, surfactants (below and above the critical micelle concentration), perfluorinated carboxylate anions, chaotropic ions, and ionic liquids, are considered. The potential of metal chelation for the determination of metal ions and organic compounds is also summarized.
