Book In Vitro Evaluation of Therapeutic Laser Treatment on Equine Tendon Fibroblasts

Download or read book In Vitro Evaluation of Therapeutic Laser Treatment on Equine Tendon Fibroblasts written by Kirsty A. Husby and published by . This book was released on 2016 with total page 113 pages. Available in PDF, EPUB and Kindle. Book excerpt: Previous investigations have failed to show benefits of low level laser therapy (LLLT) for tendon healing in horses, contrary to reports in other species. We hypothesized that delivery of LLLT at manufacturer-recommended energy fluence through equine cadaver skin to equine tendon-derived fibroblasts in vitro would increase cell proliferation, metabolic activity and migration. Fibroblasts from grossly normal front superficial digital flexor tendons of four horses euthanatized for reasons unrelated to the study were cultured in monolayer. Cells were treated through one of three equine cadaver skin samples (light, medium, or dark colored) at manufacturer-recommended energy fluence. Untreated cells served as controls. At 24 hours after treatment, cell proliferation was determined via manual cell counts and cellular metabolic rate was assessed via resazurin sodium salt reduction assay. Cell migration was determined by scratch assay and by transwell migration assay. Data was analyzed by repeated measures ANOVA with p

Book Evaluation and Optimization of a Three dimensional Construct Model for Equine Superficial Digital Flexor Tendon

Download or read book Evaluation and Optimization of a Three dimensional Construct Model for Equine Superficial Digital Flexor Tendon written by Monica Yvette Pechanec and published by . This book was released on 2017 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: Musculoskeletal injuries in equine athletes represent one of the leading causes for sport-induced and career-ending injuries due to slow or incomplete tendon repair creating clinical challenges in functional repair and effective rehabilitation. As ideal strategies for tendon repair are being developed, testing these treatments begins with models. Three-dimensional (3D) tendon constructs provide an in vitro model highly representative of in vivo tendon. For some model species, protocols for cell seeding have been established, yet this is not the case for horse cells. Using a mouse 3D construct protocol with horse superficial digital flexor tendon (SDFT) cells led us to consider the need for protocol optimization for the horse cells. High cellularity in a construct could be advantageous due to the greater amount of cellular machinery for collagen production; however, more cells may produce spatial constraints thereby limiting area for collagen deposition. A balance between cell number and spatial constraint is necessary. Additionally, seeding technique – either by spreading the matrix or localizing – may affect mechanical or material properties due to differences in fibrinogen processing, collagen production and alignment. Biochemical, collagen content, and fibril ultrastructure organization analysis conducted on varying cell seeding concentrations (100,000, 300,000, and 500,000) showed that although not significant 300,000 cells per construct produced a more functionally and structurally reproducible construct. Assessing biomechanical and collagen content analysis for two seeding methods – local and spread – the localized method proved to be statistically stronger and more representative of in vivo tendon. Collectively, our findings show evidence that although cell seeding number does not ultimately affect the construct, the cell seeding method for three-dimensional constructs does affect the overall structure and function of the model tendon.

Book Acoustoelastographic Analysis of Tendon Ultrasound Images and Its Application to Management of Equine Tendon Strain Injury

Download or read book Acoustoelastographic Analysis of Tendon Ultrasound Images and Its Application to Management of Equine Tendon Strain Injury written by Sabrina Helen Brounts and published by . This book was released on 2018 with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: Superficial digital flexor tendon injury is common in athletic horses. Recovery requires balancing rest and controlled exercise, resulting in high risk of reinjury. It is difficult to determine the level of work the damaged tissue can sustain based on serial ultrasound examination alone. Because small changes in sonographic appearance are associated with large changes in biomechanical strength, it is difficult to accurately determine the degree of loading an injured tendon can sustain. Acoustoelastography (AEG), is a new ultrasound-based tissue property evaluation technique for tendons. AEG determines stiffness gradient, the rate of change of normalized stiffness as a function of strain, by analyzing the changes in echo intensity observed in cine loops captured from gradually deforming tendon. Further, stiffness generally correlates with strength, so stiffness changes provide a useful functional metric. The central hypothesis of this dissertation is that AEG is able to detect differences in stiffness between normal and injured equine SDFT in-vitro and in-vivo. The studies presented in this dissertation demonstrate that the elastic strain limit, in the equine SDFT is ~6%. AEG can detect changes in biomechanical properties in-vitro once damage has occurred. AEG can be applied in-vivo to measure stiffness in SDFT's in clinically normal horses. AEG measurement by a novice operators is as reliable as measurement by an experienced operator. However, sedation of the horse does affect AEG; therefore, the sedation regimen needs to be consistent during serial examination of a patient. Finally, we demonstrated that AEG is able to detect the loss in stiffness in equine SDFT associated with injury with 91.9% sensitivity and 81.6% specificity. In addition, AEG is also able to identify increases in stiffness during healing. This dissertation suggests that AEG is a promising new imaging technology that can be used for the non-invasive evaluation of equine SDFT injury. Use of AEG can improve detection, monitoring and treatment of tendon injury and may ultimately shorten healing time. The findings of this dissertation will be used as the bases for future research and clinical studies to refine the technology and to investigate the use of AEG in other tendons.

Book The Soft Hard Tissue Junction

Download or read book The Soft Hard Tissue Junction written by Neil D. Broom and published by Cambridge University Press. This book was released on 2018-11-29 with total page 810 pages. Available in PDF, EPUB and Kindle. Book excerpt: Richly illustrated throughout with actual tissue images, this innovative book shows that the soft-hard tissue junction is best understood in a biomechanical context. The authors describe their pioneering experimental methods, providing an essential structure-function framework for computational modelling, and thereby encouraging the development of more realistic, predictive models of this important tissue junction. Covering the three main musculoskeletal junctions of cartilage-bone, disc-vertebra, and ligament/tendon-bone, the relevant soft tissues are examined with respect to both their own inherent structure and their mode of integration with the hard tissue. The soft-hard tissue interface is explored with a focus on structural damage resulting from overloading, and its associated pathologies. Adopting a multiscale approach, ranging in structural resolution from the macro to fibril levels, this is a must-have guide to the field and an ideal resource for researchers seeking new and creative approaches for studying the joint and spine tissues.
