Book excerpt: Plant RNA– and DNA-viruses have small genomes and with this limited coding capacity exhibit a strong dependency on host cellular processes and factors to complete their viral life cycle. Various interactions of viral proteins or nucleic acids with host components (proteins, nucleic acids, carbohydrates, lipids and metabolites) evolved, which are essential for a successful systemic spread of viruses within the plant. For example, in plants, transport of endogenous macromolecules like proteins and nucleic acids occurs in a highly selective and regulated manner and viruses exploit these specifically controlled trafficking pathways. Research on plant virus movement is located at the interface of molecular plant virology and plant cell biology. The proposed book project aims to give an overview on the current state of this research and to highlight novel insights into the dynamic interplay between plant viruses and host cells. The book is intended for researchers in plant biology and virology and especially written for those who aim to understand cell biology of virus-plant interactions. ​

Further, the book highlights the capabilities of the rhizosphere and endosphere, which help manage ecosystem responses to climate change, nutrient cycling and sequestration of carbon; and discusses their application to the development and management of renewable energy sources. In their natural environments, plants are surrounded by a tremendous number of microorganisms. Some microbes directly interact with plants in a mutually beneficial fashion, while others colonize plants solely for their own advantage. In addition, microbes can indirectly affect plants by drastically altering their environments. Understanding the complex nature of the plant-microbe interface (PMI) can pave the way for novel strategies to improve plant productivity in an eco-friendly manner. The PMI approach focuses on understanding the physical, molecular, and chemical interactions between organisms in order to determine their functional roles in biological, physical, chemical and environmental systems. Although several metabolites from plants and microbes have now been fully characterized, their roles in chemical interactions between these associates remain poorly understood, and require further investigation.

Knowledge has accumulated on the mecha nisms of the microbial infection of plants, which can lead to either disease or resistance. The mechanisms developed by plants to interact with microbes, whether viruses, bacteria, or fungi, involve events that can lead to symbiotic association or to disease or tumor formation. Cell death caused by pathogen infection has been of great interest for many years because of its association with plant resistance. There appear to be two types of plant cell death associated with pathogen infection, a rapid hypersensitive cell death localized at the site of infection during an incompatible interaction between a resistant plant and an avirulent pathogen, and a slow, normosensitive plant cell death that spreads beyond the site of infection during some compatible interactions involving a susceptible plant and a virulent, necrogenic pathogen. Plants possess a number of defense mechanisms against infection, such as (i) production of phytoalexin, (ii) formation of hydrolases, (iii) accumulation of hydroxyproline-rich glycoprotein and lignin deposition, (iv) production of pathogen-related proteins, (v) produc tion of oligosaccharides, jasmonic acid, and various other phenolic substances, and (vi) production of toxin-metabolizing enzymes. Based on these observations, insertion of a single suitable gene in a particular plant has yielded promising results in imparting resistance against specific infection or disease. It appears that a signal received after microbe infection triggers different signal transduction pathways.

In addition, omics techniques will make it possible to create a complete and complex vision using big data technologies, spurring new strategies to achieve an agriculture with a greater degree of integration of the environment, and greater efficiency of production with reduced risk to human.
