In the meantime many researchers have put enormous effort into improving methods for monoclonal antibody production. The proce dures have become more and more standardized and by this have more and more lost the character of magic secrets. Hinrich Peters and Horst Baumgarten, who had followed this good tradition already in the previous edition, written in German, suc ceeded in making laboratory tricks teachable. They had contributed their own experiences in cell culture and immunology, and were able to engage a number of experienced authors to contribute to the work. They were all willing to follow the general concept of this book, which contains a brief theoretical background for the methods described and presents the procedures in a highly organized structure. So the book has retained its shape as a "cook-book", which I especially like.

New techniques have been regularly intro duced and the sensitivity of older techniques greatly improved upon. Developments in the field of genetic engineering in particular have con tributed a wide range of new techniques. In Volume 1, published in 1983, we introduced the reader to a selection of the more advanced analytical and preparative techniques which we considered to be frequently used by research workers in the field of molecular biology. In choosing techniques for Volume 1 we obviously had to be selective and were unable to cover as broad a spectrum of techniques as we would have liked. However, the pro duction of Volume 2 has allowed us to develop the theme initiated in Volume 1 and also expand to include a wider range of subject areas. As with Volume 1, the majority of chapters relate to nucleic acid method ology, but we have also covered immunological methodology and protein 1. Obviously, we purification techniques that were not included in Volume see Volume 2 as simply a continuation of Volume 1. As with Volume 1, a knowledge of certain basic biochemical techniques and terminology has been assumed. However, since many areas of molecular biology are developing at a formidable rate and constantly generating new termin ology, a glossary of terms has been included.

For ex am pie, the root-knot nematodes belonging to the genus Meloidogyne are worldwide in their distribution and attack almost every type of crop, causing considerable losses of yield and affecting quality of produce. The cyst nematodes within the genera Globodera and Heterodera constitute a major group of plant pathogens in many countries throughout the world, suppressing yields of potato, sugar beet, soybean and cereals. Several nematodes such as longidorids and trichodorids are implicated in the transmission of numerous plant viruses. Many others cause constraints to agricultural production either locally or on large areas. However, despite their economic importance (they account for worldwide crop reduction in excess of 10%), plant parasitic nematodes are still poorly understood, because most of them are obligate parasites of roots. Environmental concerns over the agricultural use of pesticides demand the development of alternative measures to control them. To achieve environmentally sound control, knowledge of the basic biology of nematodes must be expanded. Important research areas include understanding the molecular bases for pathogenicity, the molecular mechanisms of the host parasite interactions and the genetic bases for population fluctuations. The workshop has, for the first time, brought together an international group of researchers using molecular approaches to study plant parasitic nematodes and their host responses.

The size and general nature of insects present special problems to the study of ento mology. For example, many commercially available instruments are geared to measure in grams, while the forces commonly encountered in studying insects are in the milligram range. Therefore, techniques developed in the study of insects or in those fields concerned with the control of insect pests are often unique. Methods for measuring things are common to all sciences. Advances some times depend more on how something was done than on what was measured; indeed a given field often progresses from one technique to another as new methods are discovered, developed, and modified. Just as often, some of these techniques find their way into the classroom when the problems involved have been sufficiently ironed out to permit students to master the manipulations in a few laboratory periods. Many specialized techniques are confined to one specific research labora tory. Although methods may be considered commonplace where they are used, in another context even the simplest procedures may save considerable time. It is the purpose of this series (1) to report new developments in method ology, (2) to reveal sources of groups who have dealt with and solved particular entomological problems, and (3) to describe experiments which may be appli cable for use in biology laboratory courses.

Many of these advances have been que to monoclonal antibody technology with its ability to identify antigens on tumour cells with a precision never before available. Monoclonal antibodies have the ability to identify subsets of human T-Iymphocytes and aid in diagnosis of both immunodeficiency disorders such as AIDS, or autoimmune diseases, and they can be adapted as drug targeting agents. Additionally, however, major advances have been made in identifying immunomodulating agents, and the last year has seen two such agents, IL-2 and OKT3, made available commercially for such intervention. Furthermore, another immuno modulating technique, bone marrow transplantation, ha~ now been established as a curative modality in leukaemia treatment. A central issue in tumour immunology is whether human tumours express antigens which can be termed tumour-specific. This has important implications for both tumour immunity as well as drug targeting. This issue is considered in detail by R. A. Robins in the introductory chapter in which the expression of antigens in human tumours is compared with that in experimentally induced animal tumours. This controversial issue is also considered in later chapters by Bast in breast carcinoma, Riimke and de Vries in melanoma, V{mky in lung cancer, Armitage in colorectal cancer, and Paulie and Perlmann in bladder carcinoma.

Book excerpt: The American Anti-Vivisection Society (AAVS) petitioned the National Institutes of Health (NIH) on April 23, 1997, to prohibit the use of animals in the production of mAb. On September 18, 1997, NIH declined to prohibit the use of mice in mAb production, stating that "the ascites method of mAb production is scientifically appropriate for some research projects and cannot be replaced." On March 26, 1998, AAVS submitted a second petition, stating that "NIH failed to provide valid scientific reasons for not supporting a proposed ban." The office of the NIH director asked the National Research Council to conduct a study of methods of producing mAb. In response to that request, the Research Council appointed the Committee on Methods of Producing Monoclonal Antibodies, to act on behalf of the Institute for Laboratory Animal Research of the Commission on Life Sciences, to conduct the study. The 11 expert members of the committee had extensive experience in biomedical research, laboratory animal medicine, animal welfare, pain research, and patient advocacy (Appendix B). The committee was asked to determine whether there was a scientific necessity for the mouse ascites method; if so, whether the method caused pain or distress; and, if so, what could be done to minimize the pain or distress. The committee was also asked to comment on available in vitro methods; to suggest what acceptable scientific rationale, if any, there was for using the mouse ascites method; and to identify regulatory requirements for the continued use of the mouse ascites method. The committee held an open data-gathering meeting during which its members summarized data bearing on those questions. A 1-day workshop (Appendix A) was attended by 34 participants, 14 of whom made formal presentations. A second meeting was held to finalize the report. The present report was written on the basis of information in the literature and information presented at the meeting and the workshop.

Book excerpt: Soon after the first description of monoclonal antibodies in 1976, there was enormous interest in the clinical application of antibodies, especially in the context of cancer. Antibodies appeared to offer the “magic bullet” that would allow the specific destruction of neoplastic cells. H- ever, many years’ effort resulted in very few cases of successful immu- therapy with antibodies. As a result there was a major backlash against antibody therapy, and the field lost a considerable amount of popularity. Fashion, in science as well as in other things, tends to be cyclical. Antibody-based therapy is once again attracting scientists and clinicians. There are several reasons for the renewed optimism; certainly the expe- ence of the last two decades has provided a wealth of information about problems associated with antibody therapy, and possible solutions to these problems. Recombinant antibody engineering has rejuvenated the field, allowing both the modification of antibodies to improve their in vivo pr- erties and the isolation of novel antibody molecules by such techniques as phage display. The results of recent clinical trials have demonstrated unequivocally the benefit of antibody therapy in a number of settings, and, finally, more careful consideration has been taken of the types of disease best treated using this approach.
