Its symptoms may mimic those of a number of other unrelated infections such as influenza meningitis hepatitis dengue or viral haemorrhagic fevers. It is important to distinguish leptospirosis from these diseases. For this reason new diagnostic methods have been developed in recent years. In humid tropical and subtropical areas where most developing countries are located leptospirosis poses a greater health problem than in areas with a temperate climate. Because leptospirosis is easily overlooked and consequently underreported in many parts of the world it is necessary to increase awareness and knowledge of the disease as a public health threat. The aim of these guidelines is to assist in this process. The target groups to which these guidelines are directed consist of health workers clinicians laboratory technicians microbiologists public health workers veterinarians and biologists with an interest in zoonoses having no specialized knowledge of leptospirosis but who wish to be generally informed about the microorganism concerned and the disease that it may cause. This is not a handbook and avoids technical details but the interested reader can find further information in the annexes and the general bibliography. These guidelines are concerned essentially with human leptospirosis.

Masson & Cie, requested me to write this book. A classification of these works based on their particular orientation and carrying the excess inherent in all that is schematic would lead to the definition of three types of books. Some are handbooks or treatises on "microscopy". They are tacitly limited to the biological applications of the microscope, all of which are reviewed, and include in the same volume the techniques of histology proper along with those of bacteriology, botany and embryology. Such a work, on the scale of an encyclopaedic treatise contributed by specialists from a number of discip lines and meant to be consulted as a dictionary, may have a certain value if it is really exhaustive and periodically revised. But it is no longer possible on the scale of a handbook. A single person, however gifted, cannot have acquired sufficient mastery over all the techniques of the above disciplines to be able to speak from personal experience.

Book excerpt: The mucins (mucus glycoproteins) have long been a complex corner of glycoprotein biology. While dramatic advances in the separation, structural an- ysis, biosynthesis, and degradation have marked the progress in general glycop- tein understanding, the mucins have lagged behind. The reasons for this lack of progress have always been clear and are only now being resolved. The mucins are very large molecules; they are difficult to separate from other molecules present in mucosal secretions or membranes; they are often degraded owing to natural protective functions or to isolation methodology and their peptide and oligos- charide structures are varied and complex. Understanding these molecules has demanded progress in several major areas. Isolation techniques that protect the intact mucins and allow dissociation from other adsorbed but discrete molecules needed to be developed and accepted by all researchers in the field. Improved methods for the study of very large molecules with regard to their aggregation and polymerization were also needed. Structural analysis of the peptide domains and the multitude of oligosaccharide chains was required for smaller sample sizes, for multiple samples, and in shorter time. In view of these problems it is perhaps not surprising that the mucins have remained a dilemma, of obvious biological importance and interest, but very difficult to analyze.
