Book excerpt: This book constitutes the proceedings of a NATO Advanced Research Workshop held at Chateau de Bonas (France) from 10-15 July 1990 on the Midbrain Periaqueductal Gray Matter (PAG). The aim of this meeting was to review and integrate our knowledge about the functional, anatomical and neuro chemical organization of the PAG. The PAG has been the subject of many investi gations during the last decade usually on different topics (e.g., pain modulation, defensive and sexual behavior) and generally there has been little interchange between the different research areas. The main purpose of this meeting was to bring together, for the first time, scientists who have worked on the PAG from different perspectives. This book does not pretend to present an exhaustive review of the data collected during the last 20 years of research on the PAG. The contributors to this book have been selected because their data provide key elements in the search to understand both the organization of the PAG and the role of this structure in the integration of behavior. Book excerpt: Serotonin (5-hydroxytryptamine, often cited as 5-HT) is one of the major excitatory neurotransmitter, and the serotonergic system is one of the best studied and understood transmitter systems. It is crucially involved in the organization of virtually all behaviours and in the regulation of emotion and mood. Alterations in the serotonergic system, induced by e.g. learning or pathological processes, underlie behavioural plasticity and changes in mood, which can finally results in abnormal behaviour and psychiatric conditions. Not surprisingly, the serotonergic system and its functional components appear to be targets for a multitude of pharmacological treatments - examples of very successful drugs targeting the serotoninergic system include Prozac and Zoloft. Book excerpt: In 1970 I gave up the chairmanship of the Department of Pharmacology at Stanford University Schoel of Medicine to devote full time to basic and clinical research on problems of drug addiction. In 1971 I developed the method of radioligand binding that led to the important characterization of opioid receptors in several laboratories. The extraordinary specificity of these receptors for morphine and related opiates suggested the likelihood that there were naturally occurring morphine-like molecules in the brain and other tissues. The systematic search for these molecules culminated in 1979 in the discovery, by my group, ofthe dynorphin peptides-one of the three families of opioid peptides, the first of which (the enkephalin family) had been discovered in Aberdeen, Scotland, in 1975. I also became involved in clinical research on the pharmacologic treatment of heroin addicts, for which I established the first large methadone mainte nance treatment program in California. My basic and clinical research experience convinced me that an institution encompassing laboratory research, studies on normal human volunteers, and treatment research, under a single roof, could expedite progress in understanding the drug addictions. That concept was transformed into reality by the founding, in 1974, of the Addiction Research Foundation of Palo Alto, California. The funds for construction of a laboratory were provided by a generous grant from the Drug Abuse Council (a consortium of several foundations), the president of which was Thomas L. Bryant.

This series of symposia began with a meeting in Naples in 1953 and it was particularly gratifying to welcome two members of that original gathering, Berta Scharrer and Ellen Thomsen, to the 10th Symposium. The acceptance of the invitation to meet in Bristol gave particular pleasure to the Local Organising Committee because it was the first time that a former venue was revisited - the 3rd Symposium was organised in Bristol by Hans Heller in 1961 and we were very glad that his widow, Josephine, was able to be the Guest-of-Honour at the banquet of our 10th symposium. Neurosecretion has diversified considerably since the first meeting in 1953. From the outset, the local committee decided to focus the meeting by organising it under the title "Cellular aspects of the production and release of neuropeptides. " Having sought advice from the International Committee for Symposia on Neurosecretion, we invited 28 scientists from throughout the world to present their work on aspects ranging from the organisation of the gene through to the electrical properties of peptide-secreting neurones. The progress of studies in neurosecretion owes much to the comparative approach and we attempted to reflect this in the programme of the tenth symposium by inviting the speakers, to illustrate the various aspects of neuropeptide secretion, from among scientists working with systems in many parts of the animal kingdom.

It is possible for controlled movements of muscles to be executed only if sensory information is integrated into complex neural circuits at various hierarchical levels. The chapters in this volume deal with basic and clinical aspects of integrative processing of sensory and motor activities. New findings emphasize the important influence of somatosensory activity such as tactile, proprioceptive, noxious cutaneous, and articular input on motor output. Furthermore, recordings of evoked potentials as well as unit recordings indicate that sensory and cortical activities are highly interrelated. Control of muscles by motoneurons is exerted both electrically and chemically. Disturbed muscle-motoneuron interaction is reflected in ultrastructural motoneuron morphology and may be of importance in the pathogenesis of motoneuron disease. Long loop reflex testing under various pathological conditions provides insight into disturbed sensory motor circuitry in humans. Electrophysiological recording as well as neurochemical and im munohistochemical studies elucidate the neural circuitry of basal ganglia and their neural connections, thus providing improved therapeutic concepts. The role of the thalamus and thalamocortical connections in sensory motor processing is of particular interest, because motor disturbances such as tremor or dystonia can be effectively relieved by stereotaxic interventions at the subthalamic or thalamic level.

Book excerpt: The presence of sophisticated auditory processing in mammals has permitted perhaps the most significant evolutionary development in humans: that of language. An understanding of the neural basis of hearing is thus a starting point for elucidating the mechanisms that are essential to human communication. The cochlear nucleus is the first region of the brain to receive input from the inner ear and is therefore the earliest stage in the central nervous system at which auditory signals are processed for distribution to higher centers. Clarifying its role in the central auditory pathway is crucial to our knowledge of how the brain deals with complex stimuli such as speech, and is also essential for understanding the central effects of peripheral sensorineural hearing loss caused by, for example, aging, ototoxic drugs, and noise. Ambitious new developments to assist people with total sensorineural deafness, including both cochlear and cochleus nuclear implants, require a detailed knowledge of the neural signals received by the brainstem and how these are processed. Recently, many new data have been obtained on the structure and function of the cochlear nucleus utilizing combinations of anatomical, physiological, pharmacological and molecular biological procedures. Approaches such as intracellular dye-filling of physiologically identified neurons, localization of classical neurotransmitters, peptides, receptors and special proteins, or gene expression have opened the door to novel morphofunctional correlations.
