Book excerpt: Agrobacterium is a plant pathogen which causes the “crown-gall” disease, a neoplastic growth that results from the transfer of a well-defined DNA segment (“transferred DNA”, or “T-DNA”) from the bacterial Ti (tumor-inducing) plasmid to the host cell, its integration into the host genome, and the expression of oncogenes contained on the T-DNA. The molecular machinery, needed for T-DNA generation and transport into the host cell and encoded by a series of chromosomal (chv) and Ti-plasmid virulence (vir) genes, has been the subject of numerous studies over the past several decades. Today, Agrobacterium is the tool of choice for plant genetic engineering with an ever expanding host range that includes many commercially important crops, flowers, and tree species. Furthermore, its recent application for the genetic transformation of non-plant species, from yeast to cultivated mushrooms and even to human cells, promises this bacterium a unique place in the future of biotechnological applications. Book excerpt: The broad host range pathogenic bacterium Agrobacterium tumefaciens has been widely studied as a model system to understand horizontal gene flow, secretion of effector proteins into host cells, and plant-pathogen interactions. Agrobacterium-mediated plant transformation also is the major method for generating transgenic plants for research and biotechnology purposes. Agrobacterium species have the natural ability to conduct interkingdom genetic transfer from bacteria to eukaryotes, including most plant species, yeast, fungi, and even animal cells. In nature, A. tumefaciens causes crown gall disease resulting from expression in plants of auxin and cytokinin biosynthesis genes encoded by the transferred (T-) DNA. Gene transfer from A. tumefaciens to host cells requires virulence (vir) genes that reside on the resident tumor-inducing (Ti) plasmid. In addition to T-DNA, several Virulence (Vir) effector proteins are also translocated to host cells through a bacterial type IV secretion system. These proteins aid in T-DNA trafficking through the host cell cytoplasm, nuclear targeting, and T-DNA integration. Genes within native T-DNAs can be replaced by any gene of interest, making Agrobacterium species important tools for plant research and genetic engineering. In this research topic, we provided updated information on several important areas of Agrobacterium biology and its use for biotechnology purposes.

Book excerpt: This White Paper sets out the future relationships Defence seeks with reservists and their families, their employers and society. Under the Future Reserves 2020 programme the Government has committed an additional £1.8 billion over ten year. The route from selection, is being simplified, making it easier for reservists to get to the level of training required. There will be improved alignment of pay and benefits with regulars. Reservists' total remuneration will be increased through the provision of a paid annual leave entitlement. From April 2015, when the new Armed Forces pension scheme is introduced, reservists will accrue pension entitlements for time spent on training as well as when mobilised. Welfare support will also be delivered to regulars and reservists alike, and their families, according to the impact of military service and their need. Recruitment of Army Reserve officers and those leaving the Regular Army will be encouraged to join the Reserves through bonuses of up to £5,000. A National Relationship Management scheme will be established to strengthen relationships with employer organisations and the largest employers from both the public and private sector and there will be provision of extra financial support to small and medium sized employers. New legislation will be introduced to enable mobilisation for the full range of tasks which our Armed Forces may be asked to undertake. The Territorial Army will also be renamed the 'Army Reserve' to reflect the significant changes in its role and its integration into the Whole Force.

The value of nonproducing acreage is more simply ascertained. This work is concerned with these topics, with less emphasis on the subject of estimating the oil reserves. Most of the recorded study relating to the subject has been directed toward the estimation of the recoverable oil and gas, called the oil and gas reserves, and has had its inception in tax situations and the accom panying accounting matters. The derivation of the worth of these reserves, expressed in dollars, has received less attention. However, the dollar values of oil properties have been of concern to the leaders in the industry since its beginning.
