The aims of the book are twofold. First, it explains the mechanisms of gigaseal formation using the latest discoveries. Second, it provides practical techniques for frequent formation of high resistance seals. The formation of a high-resistance electrical seal, also known as a gigaseal, between a cell membrane and a glass micropipette tip is essential in patch-clamp experiments. Even though four decades have passed since the introduction of the patch-clamping technique by Neher and Sakmann, gigaseal formation remains an obstacle in developing the high-throughput ion channel screening systems required by the pharmaceutical industry. Here the authors share their latest methods for achieving gigaseal formation and describe techniques that are highly desirable at both research and industrial levels. Nanotechnology has been found to be a powerful tool for studying and modifying glass micropipettes and in tackling the problem of gigaseal formation.

Book excerpt: This PhD project is set out to study the mechanism of gigaseal formation and to provide techniques which enable the frequent formation of high resistance seals. Such techniques are highly desirable at both research and industrial levels. The research can be divided into three main sections. In the first section the most important factors in seal formation were identified. Then a group of novel approaches have been developed to alter these factors and achieve better sealing conditions. In the second section the effect of roughness, hydrophilicity and tip size on gigaseal formation has been studied. Patch clamp experiments were carried out using conventional and treated pipettes. In the third section of this research, glass micropipettes were characterized using various measurement techniques. The proposed techniques in this thesis not only explain the gigaseal formation in more detail, but also provide techniques to increase both the seal resistance and probability of seal formation. FIB polished pipettes improved the seal resistance significantly and chemically treated pipettes resulted in the formation of seals for more than 80% of trials. Therefore the research aims have been successfully met.

Book excerpt: The patch-clamp technique is considered the gold standard to assess ion channel function across cell membranes. The technique's low throughput and highly labor-intensive nature are its main drawbacks. Emerging microelectromechanical systems (MEMS) chip-based technologies are beginning to make large-scale screens of ion channels possible. However, extremely low gigaseal formation makes it uniquely challenging to achieve the conversion from the traditional technique to chip-based patch clamp technology. A novel chip-based patch clamp system is developed by modifying surface chemistry surrounding the chip aperture with a special material. Two methods are presented to synthesize these special materials to increase electrical resistance so as to improve the chance of gigaseal formation. One method, which uses an organometallic hybrid film, mimics the chemical composition of traditional patch clamp glass and incorporates special metal elements into a silicon dioxide matrix while another method directly deposits the traditional patch clamp glass on chip surface. Chemical composition is analyzed by Energy Disperse X-ray technique and surface roughness is measured at different reflowing temperatures. The modification of surface chemistry provides a potential possibility for the chip-based patch clamp system to achieve high frequency of successful high impedance seals, which make it possible to realize high throughput and automation of this technology application in drug screening for pharmaceutical companies.
