Book excerpt: Regulation of gene expression is important to cellular growth and differentiation of eukaryotes, as it plays key roles in determining the eventual expression levels and cellular localizations of a large number of proteins. RNA-binding proteins (RBPs) with conserved RNA-binding domains (RBDs) are key molecules in controlling gene expression. These conserved RBDs can specifically interact with target RNAs through specific sequences and structures to regulate mRNA stability, translation efficiency and subcellular localization. One of the well-studied RBPs is the Puf family RBPs. Members of the Puf family share a highly conserved Puf RBD, which typically consists of eight imperfect tandem repeats of ~36 amino acids (aa). Here I report the studies of two Puf members, PfPuf3 in Plasmodium falciparum and its ortholog PyPuf3 in Plasmodium yoelii. Both genes could not be disrupted, suggesting that they may be essential for the intraerythrocytic developmental cycle (IDC). A time-course study of both PfPuf3 mRNA and protein indicated that PfPuf3 was expressed during the entire IDC, with peak expression in early trophozoites. Cellular fractionation demonstrated that PfPuf3 preferentially partitioned to the nuclear than the cytoplasmic fractions. Interestingly, tagging of the endogenous of PfPuf3 and PyPuf3 with the green fluorescent protein showed that both proteins were limited to the nucleus, which is in contrast to the cytoplasmic localization of PfPuf1 and PfPuf2. Further, we found PfPuf3 co-localized with a well-known nucleolus maker PfNop1, demonstrating that PfPuf3 is a nucleolar protein. Using a parasite line with PfPuf3-tagged with PTP (ProtC-TEV-ProtA), we were able to affinity purify the PfPuf3 protein complex and the associated RNAs. Proteomic analysis of the PfPuf3 complex identified 32 proteins that are associated with the 60S ribosome subunit, suggesting that PfPuf3 might be involved in ribosomal biogenesis. RNA sequencing analysis of the purified PfPuf3 complex revealed significant enrichment of the 28S rRNA and ITS2 (internal transcribed spacer 2), implying that PfPuf3 may recognize and bind to these rRNA sequences. Taken together, these results demonstrated nucleolar localization of PfPuf3 and suggested an essential function of PfPuf3 in ribosomal biogenesis.

Book excerpt: Malaria remains a devastating disease with respect to both mortality and the constraints it places on the economic development of the countries in which it is endemic. Our laboratory is seeking new antimalarial targets, by characterising the protein kinases of the most lethal human malaria parasite, Plasmodium falciparum. As central components of many diverse signalling pathways, protein kinases are crucial for the control of proliferation and differentiation in other eukaryotes; we hypothesise that they play similar roles in P. falciparum. The life cycle of P. falciparum is complex, consisting of a series of tightly controlled stages of division and differentiation. In the related apicomplexan parasite Toxoplasma gondii, stress stimuli have been implicated in an important differentiation step, from rapidly dividing tachyzoites, to quiescent bradyzoites (which enable immune evasion). Evidence suggests that stress may also contribute to an essential differentiation stage, gametocytogenesis, in P. falciparum. In yeast and metazoans, part of the stress response is mediated through phosphorylation of eukaryotic initiation factor 2alpha (eIF2alpha), which results in selective translation of mRNAs encoding stress response proteins. Post-transcriptional control of gene expression is suspected to play an important role in P. falciparum. Importantly, the Goldberg laboratory recently demonstrated that similarly, in P. falciparum the eIF2alpha orthologue is phosphorylated in response to starvation. Here we identify the P. falciparum orthologue of the translation initiation factor eIF2alpha and provide bioinformatic evidence for the presence of three eIF2alpha kinases in P. falciparum; PfeIK1, PfeIK2 and PfPK4, only one of which (PfPK4) has been described previously (Mohrle et al., 1997). We show that one of the novel eIF2alpha kinases, PfeIK1, is able to phosphorylate P. falciparum eIF2alpha in vitro. In addition, initial experiments support previous observations that PfPK4 is indeed an active protein kinase (Mohrle et al., 1997). We present evidence that PfPK4 is essential for asexual growth, which precludes straightforward reverse genetics studies aiming to determine its possible role in gametocytogenesis. In contrast, transgenic parasites allowed us to show that neither PfeIK1 nor PfeIK2 are required for asexual growth, or sexual development of the parasite in the mosquito vector. However, preliminary evidence (requiring confirmation) may indicate that parasites lacking PfeIK1 over-express PfPK4, which would suggest that PfeIK1 may play an important function in the parasite. This study strongly suggests that a mechanism for versatile regulation of translation by several kinases with a similar catalytic domain, but distinct regulatory domains, is conserved in P. falciparum.

Book excerpt: Immunofluorescence assays showed that this protein is exported from the Plasmodium parasite into the host erythrocyte cytosol which is consistent with studies on other Plasmodium proteins that also have the Pexel motif. To determine the enzymatic activity of FIKK11, we overexpressed the recombinant protein in E. coli and then purified it. However, no protein kinase activity was detected using several commonly used protein kinase substrates including histone H1, myelin basic protein, or dephosphorylated casein. We also did not detect any kinase activity of the native enzyme using pull-down assays of the Plasmodium falciparum cell extract against those same substrates. In addition, kinase substrate peptide array analysis of FIKK11 showed no evidence of protein kinase activity either for FIKK11. Interestingly, however, we were able to detect some kinase activity using the recombinant protein alone with no substrate. The lack of the glycine triad within subdomain I of these FIKK kinases as compared with most traditional eukaryotic protein kinases may explain why we were unable to find any interactions between FIKK11 and other commonly protein kinase substrates. Of interest was the observation that the protein reproducibly exhibited what appeared to be an autophosphorylation activity when using the standard protein kinase assay. Further analyses, however, showed that FIKK11 actually possesses diacylglycerol kinase activity utilizing 1-Stearoyl-2-arachidonoyl-sn-glycerol as a substrate. This is the first evidence of diacylglycerol kinase activity in Plasmodium falciparum. Because FIKK11 is exported into the host cell and is localized on the erythrocyte membrane, its enzymatic activity may potentially have relevance in the pathophysiology of the disease.
