Book Oxidative Folding of Proteins

Download or read book Oxidative Folding of Proteins written by Matthias J Feige and published by Royal Society of Chemistry. This book was released on 2018-07-30 with total page 450 pages. Available in PDF, EPUB and Kindle. Book excerpt: The formation of disulphide bonds is probably the most influential modification of proteins. These bonds are unique among post-translational modifications of proteins as they can covalently link cysteine residues far apart in the primary sequence of a protein. This has the potential to convey stability to otherwise marginally stable structures of proteins. However, the reactivity of cysteines comes at a price: the potential to form incorrect disulphide bonds, interfere with folding, or even cause aggregation. An elaborate set of cellular machinery exists to catalyze and guide this process: facilitating bond formation, inhibiting unwanted pairings and scrutinizing the outcomes. Only in recent years has it become clear how intimately connected this cellular machinery is with protein folding helpers, organellar redox balance and cellular homeostasis as a whole. This book comprehensively covers the basic principles of disulphide bond formation in proteins and describes the enzymes involved in the correct oxidative folding of cysteine-containing proteins. The biotechnological and pharmaceutical relevance of proteins, their variants and synthetic replicates is continuously increasing. Consequently this book is an invaluable resource for protein chemists involved in realted research and production.

Book Protein Folding

    Book Details:
  • Author : Charis Ghelis
  • Publisher : Academic Press
  • Release : 2012-12-02
  • ISBN : 0323140920
  • Pages : 580 pages

Download or read book Protein Folding written by Charis Ghelis and published by Academic Press. This book was released on 2012-12-02 with total page 580 pages. Available in PDF, EPUB and Kindle. Book excerpt: Protein Folding aims to collect the most important information in the field of protein folding and probes the main principles that govern formation of the three-dimensional structure of a protein from a nascent polypeptide chain, as well as how the functional properties appear. This text is organized into three sections and consists of 15 chapters. After an introductory chapter where the main problems of protein folding are considered at the cellular level in the context of protein biosynthesis, the discussion turns to the conformation of native globular proteins. Definitions and rules of nomenclature are given, including the structural organization of globular proteins deduced from X-ray crystallographic data. Folding mechanisms are tentatively deduced from the observation of invariants in the architecture of folded proteins. The next chapters focus on the energetics of protein conformation and structure, indicating the principles of thermodynamic stability of the native structure, along with theoretical computation studies of protein folding, structure prediction, and folding simulation. The reader is also introduced to various experimental approaches; the reversibility of the unfolding-folding process; equilibrium and kinetic studies; and detection and characterization of intermediates in protein folding. This text concludes with a chapter dealing with problems specific to oligomeric proteins. This book is intended for research scientists, specialists, biochemists, and students of biochemistry and biology.

  • Author : Natalya K. Nagradova
  • Publisher : Nova Publishers
  • Release : 2008
  • ISBN : 9781604563894
  • Pages : 254 pages

Download or read book Foldases written by Natalya K. Nagradova and published by Nova Publishers. This book was released on 2008 with total page 254 pages. Available in PDF, EPUB and Kindle. Book excerpt: A great deal of talent has been called upon over the past decade to try and understand the structure and the function of different types of foldases; as a result, a remarkable insight into how these enzymes work has been gained. Recently the main focus of research has begun to shift to unveiling the precise role of these folding catalysts in many different processes in which they are involved inside the cell. It is becoming increasingly clear that the cellular functions of foldases go far beyond accelerating the folding of proteins. This is undoubtedly an area where one can expect significant research efforts in the next few years. The present book summarises the current state of knowledge as to the structural and functional properties of four major types of foldases listed above. In structuring the material the author has sought to maintain a balance between historical development and current understanding, while also trying to look to the future and anticipate where the field is likely to move.

Book Mechanisms of Protein Folding

Download or read book Mechanisms of Protein Folding written by Roger H. Pain and published by Oxford University Press, USA. This book was released on 2000 with total page 486 pages. Available in PDF, EPUB and Kindle. Book excerpt: The process by which newly synthesized polypeptide chains become their final 3-dimensional protein forms is clearly explained, providing students and researchers of various backgrounds with both a conceptual and technical understanding of the subject. The new Second Edition incorporates the significant improvements in the field such as advances in interpreting observed kinetic data, the development of technology to observe fast folding reactions, the molten globule state, and the vital role of chaperone proteins in protein folding. The emphasis on experimental approaches has been maintained but this edition does so within the explicit context of simulations and energy surfaces. New discoveries of the central importance of protein folding and unfolding reactions in biology and medicine (including mutation and 'misfolding') are carefully explored. Three case studies elucidate the difficulties of studying protein folding in vivo.

Book Foldases Catalyzing the Formation and Isomerization of Disulfide Bonds in Proteins

Download or read book Foldases Catalyzing the Formation and Isomerization of Disulfide Bonds in Proteins written by Natalya K. Nagradova and published by Nova Science Publishers. This book was released on 2009-01-01 with total page 75 pages. Available in PDF, EPUB and Kindle. Book excerpt: One of the rate-limiting steps in the folding pathways of many secretory proteins is the formation of correct disulfide bonds between cysteine residues. In eukaryotes, both disulfide bond formation and isomerisation which shuffles incorrectly formed disulfides are catalysed by protein disulfide isomerase (PDI), whereas in bacteria these two reactions are catalysed by separate enzymes. Both in eukaryotic and prokaryotic cells the oxidation and isomerisation steps proceed exclusively in extracytoplasmic environments (the lumen of the eukaryotic endoplasmic reticulum and the Gram-negative bacterial periplasmic space). The family of foldases under discussion is characterised by a conserved "thioredoxin fold" and a common active site motif: Cys-X-X-Cys. The process of disulfide bond formation relies on thiol-disulfide exchange between oxidised and reduced cysteine pairs in the catalyst and substrate protein. Two separate pathways involved in disulfide bond formation and isomerisation have been characterised both in eukaryotes and in bacteria. In the oxidative pathway, oxidizing equivalents flow from oxygen to a membrane protein (Ero1p in eukaryotes or DsbB in bacteria), and then to a folding protein containing reduced cysteines via PDI (in eukaryotes) or via DsbA (in bacteria). In the isomerisation pathway, DsbC (bacterial protein disulfide isomerase) or PDI (in eukaryotes) interacts with substrate proteins that contain non-native disulfide bonds, allowing these bonds to rearrange to their native pairings. Reducing equivalents which are necessary to maintain DsbC in a reduced form, able to attack misfolded disulfides, are transferred from the cytoplasm with the aid of the cytoplasmic membrane protein DsbD. In eukaryotes, reduced glutathione is the main source of reducing equivalents for PDI. A dual role of PDI as an oxidase and an isomerase is facilitated by its complex domain architecture.
