Book excerpt: The sixth annual research conference of the American Institute for Cancer Research was held August 31 and September 1, 1995, at the Loews L'Enfant Plaza Hotel in Washing ton, DC. In view of the promising leads in the diet/nutrition and cancer research field, the conference was devoted to "Dietary Phytochemicals in Cancer Prevention and Treatment. " The number of sessions was increased over that in previous conferences in order to accommodate the topics of interest. The conference overview, entitled "Plants and Cancer: Food, Fiber, and Phytochemicals," provided a framework for the following sessions. In addition, the attendees were reminded that for several decades epidemiologists have noted a lower risk of lung, esophageal, stomach, and colon cancer in populations consuming diets high in fruits and vegetables. However, isolation and ingestion of individual protective factors are not the preferred action since the complexity of the food and the matrix in which nutritional factors are embedded are important. The individual sessions then provided more insight as to why eating fruits and vegetables is associated with a lower risk of cancer. The first of these sessions was on "Isothiocyanates" that induce both the Phase I and Phase II enzymes that increase detoxification and conjugation reactions, thus causing more rapid removal of any xenobiotic or carcinogen. Thus, less carcinogen is available for interaction with DNA or other critical cellular macromolecules.

The network gathers 70 partners from academia, corresponding to a total of 29 countries. The three main scientific goals are: Identify the beneficial food components released in the gut during digestion; Support the effect of beneficial food components on human health; Promote harmonization of currently used digestion models Infogest meetings highlighted the need for a publication that would provide researchers with an insight into the advantages and disadvantages associated with the use of respective in vitro and ex vivo assays to evaluate the effects of foods and food bioactives on health. Such assays are particularly important in situations where a large number of foods/bioactives need to be screened rapidly and in a cost effective manner in order to ultimately identify lead foods/bioactives that can be the subject of in vivo assays. The book is an asset to researchers wishing to study the health benefits of their foods and food bioactives of interest and highlights which in vitro/ex vivo assays are of greatest relevance to their goals, what sort of outputs/data can be generated and, as noted above, highlight the strengths and weaknesses of the various assays. It is also an important resource for undergraduate students in the ‘food and health’ arena.

An overview of normal and abnormal colonic development emphasized that although the typical human colon undergoes 1013 cell divisions by age 60, with the asso ciated possibilities for error, relatively few colon tumors develop. Since dietary modu lation leads to extremely small changes in colonic cells over a long period, animal models are useful to time, observe, and delineate the events associated with colon cancer. In the development colon cancer, the inactivation of the adenomatous polyposis coli (Apc) gene is one of the earliest known events. Normally Apc downregulates the cellular protein beta-catenin, but this is lost during cancer development. Beta-catenin may itself be an oncogene; it has a short half-life, but it is stabilized by binding to is more prevalent in the cell nucleus, the gene shuttles caherin. Colorectal cancer is one of the leading causes of cancer-related death in industrialized countries. It has been recognized to be the consequence of a dynamic process leading from hyperproliferative epithelium through different classes of adenomas to invasive carcinoma. This adenoma-carcinoma sequence has been characterized on a molecular basis. Modern molecular biology has also helped to clarify the clustering of colorectal cancer within families, a phenomenon that has been known to clinicians for a long time. Thus, the pathogenesis of the two distinct familial colon cancer syndromes FAP (familial adenomatous polyposis) and HNPCC (hereditary non-polyposis colorectal cancer) is increasingly being understood. Thereby, an identification of affected people has become possible before the disease has manifested. There is also convincing evidence that the pathogenesis of sporadic colonic cancer is modulated by environmental, mainly nutritional, factors. Carcinogens seem to be far less important than the components of the `normal' human diet. It is likely that the interplay between protective and noxious dietary compounds determines the progression of the adenoma-carcinoma sequence. Additionally, a broad spectrum of drugs has been shown to affect colonic tumorigenesis, which provides the rationale for chemoprevention strategies. These issues set the scene for discussions on how genetic and environmental factors may interact in the pathogenesis of colonic cancer, contributing fresh ideas to the prevention of this most prevalent malignancy in the industrialized world.
