Book excerpt: Modern molecular -omics tools (metagenomics, metaproteomics etc.) have greatly contributed to the rapid advancement of our understanding of microbial diversity and function in the world’s oceans. These tools are now increasingly applied to host-associated environments to describe the symbiotic microbiome and obtain a holistic view of marine host-microbial interactions. Whilst all eukaryotic hosts are likely to benefit from their microbial associates, marine sessile eukaryotes, including macroalgae, seagrasses and various invertebrates (sponges, acidians, corals, hydroids etc), rely in particular on the function of their microbiome. For example, marine sessile eukaryotes are under constant grazing, colonization and fouling pressure from the millions of micro- and macroorganisms in the surrounding seawater. Host-associated microorganisms have been shown to produce secondary metabolites as defense molecules against unwanted colonization or pathogens, thus having an important function in host health and survival. Similarly microbial symbionts of sessile eukaryotes are often essential players in local nutrient cycling thus benefiting both the host and the surrounding ecosystem. Various research fields have contributed to generating knowledge of host-associated systems, including microbiology, biotechnology, molecular biology, ecology, evolution and biotechnology. Through a focus on model marine sessile host systems we believe that new insight into the interactions between host and microbial symbionts will be obtained and important areas of future research will be identified. This research topic includes original research, review and opinion articles that bring together the knowledge from different aspects of biology and highlight advances in our understanding of the diversity and function of the microbiomes on marine sessile hosts.

Sydney rock oysters (Saccostrea glomerata) are among the most important native Australian oysters. In 1990, a selective breeding program for faster growth rates was implemented for S. glomerata. This has since expanded to include breeding for resistance to winter mortality and QX disease. Selectively-bred oysters are currently farmed in estuaries containing wild oyster populations, providing the opportunity for interbreeding that could alter the genetic variability and structure of the wild populations. Here, we use next-generation genotype by-sequencing to investigate the genetic structures of wild populations and the selectively bred B2 line of S. glomerata. These data are used to test for genetic introgression between the populations at two sites in the Georges River, an estuary in Sydney where selectively bred oysters have been farmed since 1990. Strong genetic partitioning was identified between wild and selectively-bred populations and there was no evidence of sustained gene flow in the form of introgression. Contrary to our expectations, we found significantly higher levels of genetic diversity and heterozygosity in the selectively-bred population than the wild population. These results potentially reflect the impact of population bottlenecks and are relevant to the resilience of this species to environmental change.

Book excerpt: The foundation of quantitative genetics theory was developed during the last century and facilitated many successful breeding programs for cultivated plants and t- restrial livestock. The results have been almost universally impressive, and today nearly all agricultural production utilises genetically improved seed and animals. The aquaculture industry can learn a great deal from these experiences, because the basic theory behind selective breeding is the same for all species. The ?rst published selection experiments in aquaculture started in 1920 s to improve disease resistance in ?sh, but it was not before the 1970 s that the ?rst family based breeding program was initiated for Atlantic salmon in Norway by AKVAFORSK. Unfortunately, the subsequent implementation of selective breeding on a wider scale in aquaculture has been slow, and despite the dramatic gains that have been demonstrated in a number of species, less than 10% of world aquaculture production is currently based on improved stocks. For the long-term sustainability of aquaculture production, there is an urgent need to develop and implement e- cient breeding programs for all species under commercial production. The ability for aquaculture to successfully meet the demands of an ever increasing human p- ulation, will rely on genetically improved stocks that utilise feed, water and land resources in an ef?cient way. Technological advances like genome sequences of aquaculture species, and advanced molecular methods means that there are new and exciting prospects for building on these well-established methods into the future.
