Book excerpt: This book is a tutorial designed to instruct the reader in use and application of immunochemical methods of analysis for environmental contaminants. A brief introduction describes basic principles and the advantages and disadvantages of the technology, and gives a listing of references which supply more detail. Preparation of the laboratory for use of this technology and the general scientific considerations prior to using the technology are discussed. Detailed step-wise procedures are given for analysis of selected analytes, triazine herbicides, carbaryl, paraquat, and p-nitrophenols, etc. In addition to the specific immunoassay methods, a series of support techniques necessary to perform immunochemical methods are described. This book provides specific instruction for certain analytes, but also serves to familiarize the novice reader with many generic concepts needed to successfully utilize immunochemistry technology including: applications, sampling, sample preparation, extraction, cleanup, quality assurance, methods development and optimization, data handling and troubleshooting. It is not necessary for the reader to actually perform the immunoassays given in this user's guide to obtain familiarity with these concepts. The guide is written so that the information presented can be applied to other immunoassays not given here. Thus, the strength of the guide is its universal applicability to immunoassay methods.

The development of methods, especially of the rather expensive physico-chemical procedures, has experienced considerable progress. However, to lower instrumental costs and personnel being involved, remedial measures are used and required. The highly selective and sensitive immunochemical methods are such an important supplement. They can be used to cope with quantitative (enzyme immunoessays) or qualitative problems (immunolocalization of adsorbed residues in soil and plant). The book introduces validated assays for pesticides and their metabolites that can be applied routinely. An extension to immunofluorescent methods further improves the sensitivity. The significance of the test development has been directly proven by screening programs including the use of model ecosystems.

It is the product of a workshop entitled "Immunochemical Approaches to Coastal, Estuarine and Oceanographic Questions" held at the University of Southern Maine, Portland, Oct. 5-7, 1986. Funding from many sources made this workshop possible, and is gratefully acknowledged. The reader will note great variability in both the quality and scope of the papers which we have divided 'into four sections: I. Background material on the immune system (borrowed from a recent NIH publication) II. Background material related to aquatic sciences III. Techniques IV. Applications Readers familiar with terminology are directed to start with the original contributions related to aquatic sciences, page 21. Some of the contributions are abstracts only. Others are preliminary, for which we are grateful to the authors for their willingness to share recent sometimes tentative findings with a broader community. Some offer novel and tempting insights, but as they exist, lack adequate standards and controls. Others are thorough and represent major scientific contributions. The promise rests in generating and working with reagents of great specificity enabling orders of magnitude improvements in the sensitivity of our measurements. Authors also address the problems and limitations. We thank the numerous authors and reviewers. In support of every successful endeavor is someone of unusual savvy. For us, this has been David A.
