Available in PDF, EPUB and Kindle. Book excerpt: Abstract: Reduced attentional preference for faces and symptoms of social anxiety are common in autism spectrum disorders (ASDs). The neuropeptide oxytocin triggers anxiolytic functions and enhances eye gaze, facial emotion recognition and neural correlates of face processing in ASD. Here we investigated whether a single dose of oxytocin increases attention to faces in ASD. As a secondary question, we explored the influence of social anxiety on these effects. We tested for oxytocin's effects on attention to neutral faces as compared to houses in a sample of 29 autistic individuals and 30 control participants using a dot-probe paradigm with two different presentation times (100 or 500 ms). A single dose of 24 IU oxytocin was administered in a randomized, double-blind placebo-controlled, cross-over design. Under placebo, ASD individuals paid less attention to faces presented for 500 ms than did controls. Oxytocin administration increased the allocation of attention toward faces in ASD to a level observed in controls. Secondary analyses revealed that these oxytocin effects primarily occurred in ASD individuals with high levels of social anxiety who were characterized by attentional avoidance of faces under placebo. Our results confirm a positive influence of intranasal oxytocin on social attention processes in ASD. Further, they suggest that oxytocin may in particular restore the attentional preference for facial information in ASD individuals with high social anxiety. We conclude that oxytocin's anxiolytic properties may partially account for its positive effects on socio-cognitive functioning in ASD, such as enhanced eye gaze and facial emotion recognition

Book excerpt: Emotion processing involves attending to and recognising emotional stimuli, as well as experiencing a feeling of emotion and expressing an emotional reaction. Age-related differences in the processing of emotions are apparent when comparing older (over 60 years) and young (18-30years) participants. Oxytocin has been shown to increase emotion recognition and some measures that are associated with emotional responses in young adults. The findings are not consistent though, and the effects of oxytocin are often moderated by internal and external factors, such as baseline social proficiency. There is reason to think that oxytocin could improve emotion processing in older adults. This was examined in two studies testing emotion recognition and emotion experience. In the first study, 68 older and 68 young adults were randomly allocated to receive oxytocin nasal spray (20IU) or placebo using a double-blind design. Forty-five minutes after receiving the spray, participants completed a range of tasks to assess accuracy in emotion recognition, including a basic emotion recognition task, the Reading the Mind in the Eyes Test, and an emotion-matching task. During the basic emotion recognition task and the RMET, participants wore an eye-tracking device.

Book excerpt: Intranasal oxytocin (OT) can enhance emotion recognition, perhaps by promoting increased attention to social cues. Some studies indicate that individuals with difficulties processing social information, including those with psychopathology, show more pronounced effects in response to OT. As such, there is interest in the potential therapeutic use of OT in populations with deficits in social cognition. The present study examined the effects of intranasal OT on the processing of facial features and selective attention to emotional facial expressions, as well as whether individual differences in depressive symptom severity predict sensitivity to intranasal OT. In a double-blind placebo-controlled within-subject design, eye tracking was used to measure attention to facial features in an emotional expression appraisal task, and attention to emotional expressions in a free-viewing task with a quadrant of multiple faces. OT facilitated the processing of positive cues, enhancing the maintenance of attention to the mouth region of happy faces and to happy faces within a quadrant, although the latter effect fell short of conventional statistical significance. Further, persons with depressive symptoms, and particularly males, were sensitive to OT's effects. For males only, OT, relative to placebo, increased attentional focus to the mouth region of all faces. Individuals with depressive symptoms showed less attentional focus on angry (males only) and sad facial expressions, and more attention to happy faces (particularly for males). Results indicate increased sensitivity to OT in males and persons at risk for depression, with OT administration promoting a positive bias in selective attention to social stimuli.

From a human perspective, it is crucial to understand more precisely who can benefit from potential oxytocin-related treatments, which outcome measures will best represent their effects, how they should be administered, and what brain mechanisms are likely involved in mediating their effects. This type of “precision medicine” approach is in line with the research domain criteria defined by the U.S. National Institute of Mental Health.
