Book excerpt: Feeding conjugated linoleic acids (CLA) improves reproductive performance in dairy cows; however, the molecular mechanisms by which CLA improves reproduction are not well understood. Therefore, we evaluated whether the CLA isomers, trans-10, cis-12 CLA and cis-9, trans-11 CLA altered synthesis of steroidogenic hormones in bovine luteal cells by measuring concentrations of progesterone, PGE2, and PGF2[alpha] in conditioned medium and expression of genes involved in their synthesis. Confluent luteal cells from each of 4 cows were cultured in 0 [mu]M (control) or 0.1 [mu]M solutions of trans-10, cis-12 CLA and cis-9, trans-11 CLA in varying ratios (1:0, 0:1, 1:1, 2:1, 1:2, 5:1, 1:5, 9:1, or 1:9) for 48 h in the presence and absence of 1 [mu]M of the adenylate cyclase activator forskolin. Independent of CLA isomer and ratio, CLA decreased, compared to control, hormone concentrations of prostaglandin F2[alpha] (62.6 ± 10.5 vs. 50.4 ± 9.9 pg/mL; P[subscript Overall] = 0.003) and, in the absence of forskolin, prostaglandin E2 (61.2 ± 11.3 vs. 36.1 ± 10.1 pg/mL; P

Both of these processes impact the viability of the corpus luteum, which is required to maintain early pregnancy. In cattle and other species, progesterone supports gestation, regulates embryogenesis and works in concert with estrogen to maintain the reproductive tract. Many investigators have sought to understand the processes that control regression of the corpus luteum. Two prostaglandins, PGF2a and PGE2, have been implicated in the functioning of this transient gland. It is accepted by most researchers that the primary luteolysin responsible for functional and structural demise of the corpus luteum is PGF2a. However, there is ambiguity between reported effects of PGF2a and PGE2 on whole cells and on animals in vivo. Therefore, the specific effects of these prostaglandins on oxygen consumption and progesterone secretion in mitochondria isolated from bovine corpora lutea are the focus of this dissertation. A decrease in oxygen consumption and a concomitant decrease in ATP may contribute to luteolysis. These in vitro studies using isolated mitochondria may help to resolve these questions. The first objective was to determine the influence of PGF2a and PGE2 on the oxygen consumption of mitochondria isolated from bovine corpora lutea. The second objective was to determine the effect of both of these prostaglandins on mitochondrial progesterone production. This is the first time that the direct effects of PGF2a on mitochondrial cellular respiration and steroidogenesis have been studied in the corpora lutea of any species. These results will expand information in the area of ovarian physiology and may clarify the roles of PGF2a and PGE2 in corpus luteum function.

The organizers of this symposium recognized the need to focus male reproductive understanding on contraceptive development. The ultimate objective was and still is to produce a variety of safe and effective male contraceptives similar to that accomplished in the female. Speakers were invited to review the state of the art in several areas related to male contra ception, reproductive toxicity and reproductive biology. The abstracts of these sessions were published as a special issue of Archives of Andrology (Vol. 9, No.1, August, 1982). Subsequently, this volume was assembled from key papers presented at the Symposium.
