Book Effect of Selective COX 2 Inhibitors on Hepatic Progenitor Cells and the Pathologies of Experimental Hepatocarcinogenesis

Download or read book Effect of Selective COX 2 Inhibitors on Hepatic Progenitor Cells and the Pathologies of Experimental Hepatocarcinogenesis written by Richard Davies and published by . This book was released on 2007 with total page pages. Available in PDF, EPUB and Kindle. Book excerpt: [Truncated abstract] Hepatocellular carcinoma (HCC) is the major malignancy complicating chronic liver disease. New therapies for the prevention of HCC are required due to the limited success and high tumour recurrence rates of existing treatments. Emerging evidence suggests that HCC arise from the transformation of adult liver progenitor cells (LPCs), which have the capacity to differentiate into hepatocytes and biliary cells during liver regeneration. LPC activation precedes neoplasia in experimental hepatocarcinogenesis. LPCs share antigenic epitopes with HCCs, including α-fetoprotein (AFP) and M2- pyruvate kinase (M2PK). In animal models of hepatocarcinogenesis, attenuation of the LPC response reduces the incidence of HCC following prolonged liver injury via a tumour necrosis factor (TNF) dependent mechanism. As TNF is a pro-inflammatory cytokine, these data suggest that anti-inflammatory agents may be effective in inhibiting LPC activation and hepatocarcinogenesis. Cyclo-oxygenase-2 (COX-2) is an inducible enzyme that mediates the production of many prostaglandins during inflammation and carcinogenesis. Recent investigations show that the administration of selective COX-2 inhibitors (SC2Is) may reduce the incidence of a variety of tumours including breast, colon and skin. The broad aim of this thesis was to conduct a series of detailed studies on the effects of a SC2I on LPC activation and the hepatic pathologies associated with hepatocarcinogenesis in order to test the hypothesis that S2CIs may be a beneficial therapy that can reduce liver injury and pre-neoplastic changes in the choline-deficient, ethionine supplemented (CDE) murine model of hepatocarcinogenesis. Administration of a SC2I (SC-236) significantly inhibited a variety of hepatic cell populations that expand during the first month of the CDE mouse model of hepatocarcinogenesis (a choline deficient, ethionine supplemented diet). Numbers of M2PK-positive LPCs (which are more hepatocytic in morphology and are also COX-2 positive) and inflammatory cells were all significantly reduced by SC-236. In contrast, numbers of A6-positive LPCs (which are more biliary cell-like in morphology and do not express COX-2) were unchanged. ... In summary, these data suggest that COX-2 inhibitors such as SC-236 inhibit LPC activation and a variety of pre-neoplastic liver pathologies as a result of COX-2 dependent and independent mechanisms that may be mediated through inhibition of Akt phosphorylation and induction of apoptosis. Moreover, SC2Is may be useful as preventative treatment strategies for HCC in patients with chronic liver disease.

Abstract: Abstract of thesis entitled The effect of celecoxib on hepatocellular carcinoma Submitted by Terence Tang Chi Man For the degree of Doctor of Philosophy at the University of Hong Kong in August 2005 Hepatocellular carcinoma (HCC) is the second leading cause of cancer death in males in Hong Kong in the year of 2004. Previous studies have shown increased levels of cyclooxygenase-2 (COX-2) in a variety of human malignancies including HCC, suggesting that abnormal COX-2 expression plays an important role in carcinogenesis. In addition, some evidence suggests that selective COX-2 inhibitors suppress the formation of tumors in experimental models. However, the significance of COX-2 in progression of HCC remains unclear, and there are no data in the literature on the use of COX-2 inhibitor in HCC. The main objectives of this project were to evaluate the role of COX-2 in human HCC, and to investigate the in vitro and in vivo effects of celecoxib on HCC. Firstly, we evaluated the significance of COX-2 in HCC by examining the relationship between COX-2 and VEGF 165 levels and clinicopathological features. This study showed that elevated COX-2 levels correlated with elevated VEGF levels and invasiveness in HCC. Our data suggested that COX-2 plays a significant role in the invasiveness of HCC, possibly via its angiogenic effect. viSecondly, we performed in vivo studies to investigate the chemopreventive and therapeutic effects of celecoxib, a selective COX-2 inhibitor, on the development and growth of HCC xenograft in nude mice. Our results showed that celecoxib could prolong the latent period of HCC xenograft development in nude mice and decreased the growth of established HCC xenograft in nude mice significantly. However, there was no significant decrease in MVD and VEGF level in the treatment groups compared with the control groups. In in vitro studies, celecoxib induced apoptosis significantly in HCC cell lines and HUVEC. Interestingly, celecoxib induced G1 arrest in both HCC cell lines but not in HUVEC. We have found that the cyclin D1 decreased as the concentration of celecoxib increased. Celecoxib most likely induced apoptosis through the down-regulation of phosphorylation of NFκB, which reduced the cyclin D1 expression, and hence inhibition of proliferation in celecoxib-treated xenografts and cell lines. Finally, we investigated the gene profiling of celecoxib and COX-2 modulation in HCC in the in vitro study by oligo microarray analysis. We aimed to investigate the COX-2-dependent and -independent pathways caused by celecoxib treatment in HCC. Our microarray data suggested that celecoxib may act on both COX-dependent and -independent pathways. We verified six genes by using RT- PCR and northern blot analysis from our microarray data. Three genes (RRM2, KNG1, and FABP1) from COX-2-dependent pathways and three genes (ATF3, CXCL5, and SERPINC1) from COX-independent pathways were considered worthy of further study. In conclusion, our data suggest that COX-2 expression is significantly associated with progression of HCC, and the selective COX-2 inhibitor, celecoxib, can delay the development of HCC and suppress the growth of established HCC in vii experimental animals, most likely via the growth arrest and apoptosis pathways. COX-2 inhibition appears to be a promising strategy for chemoprevention and treatment of HCC

Download or read book Cox 2 written by Joseph R. Bertino and published by Karger Medical and Scientific Publishers. This book was released on 2003-01-01 with total page 300 pages. Available in PDF, EPUB and Kindle. Book excerpt: Numerous investigators have found increased levels of cyclooxygenase-2 (COX-2) in both premalignant and malignant tissues. Moreover, animals engineered to be COX-2 deficient or treated with a selective COX-2 inhibitor showed reduced tumor formation and growth. The present book reviews these findings suggesting that COX-2 is a valid molecular target for the prevention and treatment of cancer. The first chapters are devoted to the epidemiology of nonsteroidal anti-inflammatory drugs (NSAIDs) and cancer, the pharmacology of COX-2 inhibitors and the regulation of COX-2 expression in human cancers. Besides, the book contains a series of chapters examining the link between COX-2 and cancer in specific organs (e.g. skin, breast, cervix, digestive tract, lung, etc.). Furthermore, several clinical trials assessing the potential of COX-2 inhibitors for preventing and treating cancer are described. The final chapter provides an up-to-date consideration of COX-independent targets of NSAIDs and related compounds in cancer prevention and treatment. This state-of-the-art publication is recommended reading not only for medical and radiation oncologists, but also for pharmacologists, gastroenterologists and other subspecialists.

Book How Tobacco Smoke Causes Disease

Download or read book How Tobacco Smoke Causes Disease written by United States. Public Health Service. Office of the Surgeon General and published by . This book was released on 2010 with total page 728 pages. Available in PDF, EPUB and Kindle. Book excerpt: This report considers the biological and behavioral mechanisms that may underlie the pathogenicity of tobacco smoke. Many Surgeon General's reports have considered research findings on mechanisms in assessing the biological plausibility of associations observed in epidemiologic studies. Mechanisms of disease are important because they may provide plausibility, which is one of the guideline criteria for assessing evidence on causation. This report specifically reviews the evidence on the potential mechanisms by which smoking causes diseases and considers whether a mechanism is likely to be operative in the production of human disease by tobacco smoke. This evidence is relevant to understanding how smoking causes disease, to identifying those who may be particularly susceptible, and to assessing the potential risks of tobacco products.

Book In Vivo Models to Study Angiogenesis

Download or read book In Vivo Models to Study Angiogenesis written by Domenico Ribatti and published by Academic Press. This book was released on 2017-08-18 with total page 130 pages. Available in PDF, EPUB and Kindle. Book excerpt: In Vivo Models to Study Angiogenesis provides the latest information and an overview of the most common assays for studying angiogenesis in vivo. Under physiological conditions, angiogenesis is tightly controlled, whereas increased production of angiogenic stimuli and/or reduced production of angiogenic inhibitors leads to abnormal neovascularization, such as occurs in cancer, chronic inflammatory disease, diabetic retinopathy, macular degeneration and cardiovascular disorders. Several genetic and epigenetic mechanisms are involved in the early development of the vascular system. This book presents the latest information from the extensive literature and research available. Evidence is now emerging that blood vessels themselves have the ability to provide instructive regulatory signals to surrounding non-vascular target cells during organ development. Thus, endothelial cell signaling is currently believed to promote fundamental cues for cell fate specification, embryo patterning, organ differentiation and postnatal tissue remodeling. Provides information on the most common assays to study angiogenesis in vivo Presents an ideal reference for those interested in angiogenesis as a normal and vital process in growth and development Covers wound healing, the formation of granulation tissue, and the transition of tumors from benign to malignant
