The growing approaches to reduce the cost of cellulases production can be achievd viausing of cheap , renewable agriculture wastes which consider an excellent carbon source for microbial enzymes production and also looking for microorganism having the ability to hyper-production of cellulases. Therefore, considering the advantages of SSF, the optimal chemical condition for the production of cellulase enzyme by Apergillus nigerfrom solid -state fermentation of sugarcane bagasse is investigated.

Book excerpt: ABST ACT The use of waste as raw material is important for government economy and natural balance. The purpose of this work is to study the production of xylanase and CMCase enzyme by Aspergillus niger in solid state fermentation (SSF) using agricultural residues sugar cane bagasse as substrates. The study is focus on the effect of substrate concentration which is 1 g/L, 3g/L and 5 g/L. All the enzymes were incubating in temperature 37 oC and pH 5.0 for 30 minutes. The reaction was quenched by cooling on ice and the amount of reducing sugar was measured by dinitrosalicyclic (DNS) acid procedure with slight modification (Miller, 1965). The mixture was then incubated at 100 oC for 5 minutes. Subsequently, the reaction was stopped by cooling on ice. Distilled water was added into the final volume of 16 ml and the color intensity was measured at wavelength of 575 nm. The concentration of SCB affected the xylanase and CMCase activity. Xylanase and carboxymethylcellulase activity was higher at lower concentration hen using 3 g of SCB.

In the present study, optimization for cellulase production by Aspergillus niger was investigated by using paddy straw as substrate.Cellulase enzyme was produced by the fungus extracellularly. Optimization of cellulase production was done by using various physical (Temperature, pH, Salinity and Incubation time) and chemical parameters (Carbon sources and nitrogen sources) which could influence the enzyme activity. Cellulase production was maximum at the temperature 20°C and minimum at 40ºC. The optimal pH for the cellulase production was observed maximum in 6.0 and minimum in 7.0. Cellulase production was maximum at 48 hours and minimum at 24 hours. Cellulase production was maximum with when fructose was used as a carbon source and minimum with sucrose. Cellulase production was maximum when Malt extract was used as a nitrogen source and minimum with yeast extract. The cellulase enzyme extract was partially purified and its protein fraction was stained with its molecular weight determination by using SDS-PAGE. It revealed two protein bands with the molecular weights of about 83 kD and 50kD.

It consists on seeding solid culture medium with bacteria or fungi (filamentous or higher) and on producing, in this medium (solid components and exudates) metabolites and high value products. In fact, this process is very old. In older industries such the food and agricultural, this technique has been extensively used. An example of this is the production of pork sausages and Roquefort cheese. Book excerpt: The production of cellulases from rice straw (RS) by four fungi: Trichoderma harzianum (ScahmT105), Trichoderma spp.(1) (STP101), Trichoderma spp.(3) (STP103) and Phanerochaete chrysosporium was investigated. The microbial treatment using solid state fermentation was conducted in 250 ml Erlenmeyer flasks considering ride straw as a major solid substrate. The highest cellulase activities such as 25.53 U/g rice straw due to filter paper activity and 42.86 U/g of rice straw due to carboxymethyl cellulose activity were obtained at day 4 of cultivation using Phanerochaete chrysosporium for the purpose of selecting the best fungus among these four strains. Glucusoamine for growth and reducing sugar as substrate utilization indicator were observed to evaluate the fermentation of rice straw in the experiment and pH values were recorded as well. Four process parameters of the solid state fermentation namely moisture content, mineral content, co-substrate and inoculum size with three levels of each parameter were used to optimize the production of cellulase by Plackett-Burman technique under factorial design. The results for first phase of optimization showed that the production of cellulase were higher i.e. 26.43 U/g of rice straw due to filter paper of activity and 46.25 U/g of rice straw due to carboxymethyl cellulose activity compared to the production obtained during the earlier study of selecting best strain among four fungi but the optimum regions of the surfaces was not found. Second phase of optimization was conducted to determine the actual optimum condition within the ranges of variables tested. The experimental data were used to develop second order polynomial models considering linear, quadratic and interaction effects of the variables (factors). The optimum values obtained at second phase of optimization for moisture content, inoculum size, co-substrate and mineral content were 50% (v/w), 10% (v/w), 1% (w/w) and 5% (v/w) respectively. Using the final model equations the process factors/variables were tested by increasing or decreasing the values within the ranges of parameters tested and optimum production of cellulase were obtained to be 30.18 U/g of rice straw (FPU) and 53.93 U/g of rice straw (CMCase) for Phanerochaete chrysosporium with the optimum process conditions. A final experiment with these optimum process conditions. A final experiment with these optimum process parameters of SSF was conducted to evaluate the production of cellulases as well as the validation of the models which indicated the production of 29.46 U/g of rice straw due to filter paper activity and 54.83 U/g of rice straw due to carboxymethyl cellulose activity in the laboratory which approved the optimum production obtained with 2.4% and 1.6% error, respectively.
