Book excerpt: The purpose of this investigation was to identify the regulatory elements involved in the transactivation of the human c-myc gene by herpes simplex virus type-1 (HSV-1). This study mainly focused on a major c-myc promoter P2 (Fig. 1). Transient expression assays were carried out with a reporter plasmid, that contained either a wild type (+66wt c-myc) or mutant (+66 mt E2F, +66mt CT -I2) c-myc promoter fused to the chloramphenicol acetyltransferase (CAT) gene, cotransfected with effector plasmids that contained the HSV-1 immediate early (IE) genes ( encoding IE proteins, ICP4, ICP0, ICP27). The transient expression assays revealed that the HSV-1 IE protein, ICP4, was a major effector in transactivation of the human c-myc P2 promoter by HSV-1, and ICPO provided a synergistic effect with ICP4. Cis-acting elements E2F and CT-12, which were potential binding sites for cellular transcription factors E2F and Spl respectively, were shown to be important in HSV-1 induced c-myc transactivation. In addition, these assays indicated that ICPO influences ICP4 mediated activation through these two sites. Electrophoretic mobility shift assays, oligonucleotide competition assays, and footprinting analysis together demonstrated that viral proteins didn't directly interact with P2 promoter region. Three cellular transcription factors, Spl, E2F, and YYl, however, specifically interacted with the P2 promoter region, of which, Spl and its related proteins SpX bound to several other sites in addition of CT-I2 site. Moreover, SpX also appeared to interact with another cellular factor that bound together with it on the P2 promoter region, and a second unknown cellular factor was also observed to interact with P2 promoter region.

Book excerpt: Gammaherpesviruses are clinically important human pathogens associated with multiple types of tumors. The life cycle of gammaherpesviruses consists of two distinct phases, latency and lytic replication, both of which are important for the viral-associated pathogenesis. Murine gammaherpesvirus 68 (MHV-68) has been used as a model to study virus-host interactions of human gammaherpesviruses because of its genomic and biological similarities. During the course of MHV-68 lytic infection, the virus is highly dependent on cellular machinery to facilitate its replication. Host cells, on the other hand, develop mechanisms to sense and restrict virus replication. Here, we present an integrative systems approach to elucidate the interactions between MHV-68 and its host. Our previous yeast-two-hybrid analysis and genome-wide expression profiling implicated hundreds of cellular factors mediating virus-host interactions. Among the cellular factors identified, those involved in kinase- and transcription factor- regulated signaling are the most highly enriched. Therefore, high-throughput screens of cellular kinome and transcription factors cDNA and siRNA libraries were carried out, which uncovered 125 MHV-68 dependency factors and 213 restriction factors. We found that many cellular factors affect MHV-68 replication by regulating RTA transcription. Characterization of this subset of genes showed they also play roles in KSHV latency maintenance and reactivation. Furthermore, cellular factors regulating later steps of viral replication, such as viral late gene expression and egress, were also identified and validated. Finally we compared the viral restriction factors to the results of a kinome siRNA screen designed to identify cellular regulators of innate immune response to cytosolic DNA. Remarkably, 23 genes were found among the top 10% of the hits of both screens. The MHV-68 restriction factor RIOK3 was confirmed to play a role in IRF3 activation and type I interferon induction mediated by both cytosolic DNA and RNA. RIOK3 is required for cellular antiviral response to both DNA virus (MHV-68) and RNA virus (Influenza A), suggesting a central role in innate immunity. Further analysis revealed that RIOK3 complexes with TBK1 and IRF3, bridging the activity between the two. Collectively, this multilayered approach unfolded a comprehensive map of gammaherpesvirus-host interactions which will provide basis for developing new anti-viral therapies.

It has been known for some considerable time that heat shock proteins from prokaryotic organisms are immunodominant molecules that are intimately involved in the induction of potential protective inflammatory responses, and this aspect of HSP biology is updated herein. In addition to regulating heat shock protein gene expression, the transcription factor HSF1 also appears to play an important role in regulating immune responses to infection. Heat shock proteins are now known to influence infectious disease processes in a number of diverse ways: they are involved in the propagation of prions, the replication and morphogenesis of viruses, and the resistance of parasites to chemotherapy. These proteins also appear to be important mediators of bacteria-host interactions and inflammation, the latter via interactions with cell surface molecules and structures such as Toll-like receptors and lipid rafts. Heat shock proteins can be expressed on the surface of infected cells, and this is likely to provide a target for the innate immune response. Elevated levels of circulating HSP are present in infectious diseases and these proteins might therefore regulate inflammatory responses to pathogenic challenge on a systemic basis. Heat shock proteins are also implicated in the impact of genital tract infections on the reproductive outcome, as well as in the local and systemic consequences of periodontal disease. Fever-range temperatures can induce the expression of heat shock proteins, and the final chapter in the book examines the influence of fever-range hyperthermia on a variety of cells and the organization of plasma membranes. This book is an essential read for graduates and postgraduates in Biology, pro- and eukaryotic Biochemistry, Immunology, Microbiology, Inflammatory and Infectious Disease, and Pathology.

Book excerpt: It is now widely acknowledged that at the beginning of this century Claude von Pirquet first pointed out that a viral disease, i. e. , measles, resulted in an anergy or depression of preexisting immune response, namely, delayed continuous hypersensitivity to PPD derived from Mycobacterium tuberculosis. Thereafter ob servations that viral infections may result in immunosuppression have been recorded by many clinicians and infectious disease investigators for six or seven decades. Nevertheless, despite sporadic reports that infectious diseases caused by viruses may result in either transient or prolonged immunodepression, investigation of this phenomenon languished until the mid-1960s, when it was pointed out that a number of experimental retroviral infections of mice with tumor viruses may result in marked immunosuppression. However, it was not until the recognition of the new epidemic of acquired immunodeficiency syn drome (AIDS) caused by the human immunodeficiency virus and related vi ruses that acquired immunodeficiencies associated with virus infection became general knowledge among biomedical investigators as well as the lay public. A number of reviews published during the past decade or so pointed out that numerous viruses may affect humoral and cellular immune responses. Furthermore, expanding knowledge about the nature and mechanisms of both humoral and cellular immunity and pathogenesis of viral infections has pro vided clinical and experimental models for investigating in depth how and why viruses of man and animals profoundly affect immune responses.

Additionally, cell based therapies (adoptive T cell therapy, CAR T cells and TCR transduced T cells) have demonstrated significant efficacy in patients with B cell malignancies and melanoma. Immune checkpoint inhibitors in particular have generated enormous excitement across the entire field of oncology, providing a significant benefit to a minority of patients.
