With this term we usually mean a cell-like structure containing the minimal and sufficient number of components to be defined as alive, or at least capable of displaying some of the fundamental functions of a living cell. In fact, when we look at extant living cells we realize that thousands of molecules are organized spatially and functionally in order to realize what we call cellular life. This fact elicits the question whether such huge complexity is a necessary condition for life, or a simpler molecular system can also be defined as alive. Obviously, the concept of minimal cell encompasses entire families of cells, from totally synthetic cells, to semi-synthetic ones, to primitive cell models, to simple biomimetic cellular systems. Typically, in the experimental approach to the construction of minimal the main ingredient is the compartment. Lipid vesicles (liposomes) are used to host simple and complex molecular transformations, from single or multiple enzymic reactions, to polymerase chain reactions, to gene expression. Today this research is seen as part of the broader scenario of synthetic biology but it is rooted in origins of life studies, because the construction of a minimal cell might provide biophysical insights into the origins of primitive cells, and the emergence of life on earth. The volume provides an overview of physical, biochemical and functional studies on minimal cells, with emphasis to experimental approaches. 15 International experts report on their innovative contributions to the construction of minimal cells.

We hope that the book will be interesting and useful to students of the biological sciences and medicine, and to those in the teaching professions who do not have ready access to research papers. Since space is at a premium, we have denied ourselves the luxury of a philosophical discussion of the problems of defining organelles. Rather we have chosen to include all those intracellular structures which have limiting membranes and definable compartments. The separate chapters consider nuclei, plastids, mitochondria, microbodies, endoplasmic and sarcoplasmic reticulum; Golgi bodies, lysosomes and various secretory vesicles, including chromaffin granules and synaptic vesicles. Nucleoli, ribosomes, and centrioles are included in the chapter on nuclei. New and exciting informatioIi about all these structures has emerged in recent years-for example; the nUcleosome, interrupted genes, signal sequences on proteins destined. for the bioenergetic organelles, mapping and sequencing of organelle genes, and consolidation of chemiosmosis as a unifying principle in energy transduction. We have outlined as many of these developments as possible and pointed out some areas of controversy. The literature on subcellular biology is so extensive that it would have been easier to have written a separate book on each organelle.

On the surface, this question is straightforward-in principle, the smallest cells can be identified and measured. But understanding what factors determine this lower limit, and addressing the host of other questions that follow on from this knowledge, require a fundamental understanding of the chemistry and ecology of cellular life. The recent report of evidence for life in a martian meteorite and the prospect of searching for biological signatures in intelligently chosen samples from Mars and elsewhere bring a new immediacy to such questions. How do we recognize the morphological or chemical remnants of life in rocks deposited 4 billion years ago on another planet? Are the empirical limits on cell size identified by observation on Earth applicable to life wherever it may occur, or is minimum size a function of the particular chemistry of an individual planetary surface? These questions formed the focus of a workshop on the size limits of very small organisms, organized by the Steering .Group for the Workshop on Size Limits of Very Small Microorganisms and held on October 22 and 23, 1998. Eighteen invited panelists, representing fields ranging from cell biology and molecular genetics to paleontology and mineralogy, joined with an almost equal number of other participants in a wide-ranging exploration of minimum cell size and the challenge of interpreting micro- and nano-scale features of sedimentary rocks found on Earth or elsewhere in the solar system. This document contains the proceedings of that workshop. It includes position papers presented by the individual panelists, arranged by panel, along with a summary, for each of the four sessions, of extensive roundtable discussions that involved the panelists as well as other workshop participants.

Both structurally and functionally, there are many peculiarities that differentiate the brain from all other organs. The brain is our connection to the world around us and by governing nervous system and higher function, any disturbance induces severe neurological and psychiatric disorders that can have a devastating effect on quality of life. Our understanding of the physiology and biochemistry of the brain has improved dramatically in the last two decades. In particular, the critical role of cations, including magnesium, has become evident, even if incompletely understood at a mechanistic level. The exact role and regulation of magnesium, in particular, remains elusive, largely because intracellular levels are so difficult to routinely quantify. Nonetheless, the importance of magnesium to normal central nervous system activity is self-evident given the complicated homeostatic mechanisms that maintain the concentration of this cation within strict limits essential for normal physiology and metabolism. There is also considerable accumulating evidence to suggest alterations to some brain functions in both normal and pathological conditions may be linked to alterations in local magnesium concentration. This book, containing chapters written by some of the foremost experts in the field of magnesium research, brings together the latest in experimental and clinical magnesium research as it relates to the central nervous system. However, since their discovery nearly 200 years ago, it became clear that these two organelles are interacting, and that their functions are much more complex and far reaching than previously thought. Recent findings indicate that the Golgi–Centriole relationship may be important for directional protein transport, cell polarization and cell cycle progression. Current studies indicate that Golgi and centriole also participate in development and act as cellular and immunological sensors, and that their abnormalities lead to cell and developmental abnormalities, Alzheimer, cancer, various lipid disorders and neurological and immunological diseases in humans. This volume combines the latest information on the structure, molecular composition, and roles of Golgi and centriole in various cellular functions and diseases. The metabolic capacity of a eukaryotic (plant) cell and the steps leading to it are overwhelmingly an endeavour of a joint genetic cooperation between nucleus/cytosol, plastids, and mitochondria. Alter ation of the genetic material in anyone of these compartments or exchange of organelles between species can seriously affect harmoniously balanced growth of an organism. Although the biological significance of this genetic design has been vividly evident since the discovery of non-Mendelian inheritance by Baur and Correns at the beginning of this century, and became indisputable in principle after Renner's work on interspecific nuclear/plastid hybrids (summarized in his classical article in 1934), studies on the genetics of organelles have long suffered from the lack of respectabil ity. Non-Mendelian inheritance was considered a research sideline~ifnot a freak~by most geneticists, which becomes evident when one consults common textbooks. For instance, these have usually impeccable accounts of photosynthetic and respiratory energy conversion in chloroplasts and mitochondria, of metabolism and global circulation of the biological key elements C, N, and S, as well as of the organization, maintenance, and function of nuclear genetic information. In contrast, the heredity and molecular biology of organelles are generally treated as an adjunct, and neither goes as far as to describe the impact of the integrated genetic system.

Each techno logical advance in detection and visualization has led to the description of different organelles and cell types. Basic biochemical processes in cells were recognized and are now weIl understood. It is only recently however, that research has expanded to include the specific meta bolic function of the specialized cell types and organelles. In some cases metabolic roles were recognized when the organelles were first described, e.g., chloroplasts, mito chondria, etc., in others the metabolic role remains unknown. Chemical and biochemical specialization in plants or their organelles is equally challenging. Although biochemists have laboured intensivelyon many isolated plant organelles, it is only recently that technical advances have permitted the examination of specialization in the metabolism of cell types. This area of research, although under intensive investigation in some areas of plant metabolism, is still in its infancy. Further developments in methodology or in production of specific genetic lines of plants will greatly improve our understanding of the specialization of different tissues and cell types. This volume describes the current status in the dis cipline as presented in a Symposium on the Cellular and Subcellular Specialization in Plant Metabolism during the Annual Meeting of the Phytochemical Society of North America, at Cornell University, Ithaca, N.Y., on August 10-14, 1981.
