Book excerpt: Abstract: The most important role of beta-carotene has long been thought to be the metabolic cleavage at the central double bond to produce two molecules of vitamin A (retinol). There is growing interest in the "eccentric cleavage" of dietary carotenoids at different double bond sites of beta-carotene to produce apocarotenals and their possible oxidation to apocarotenoic acids. However, there are no careful, quantitative analyses of the levels of these compounds in human foods and tissues, and little is known of the enzymology of their formation. This project is part of a larger effort to characterize the full family of putative metabolites of beta-carotenes and lycopene, their occurrence, and their biological significance. We are synthesizing and characterizing all of the possible aldehyde and acid metabolites of beta-carotene for use as analytical standards and for direct assay of their biological activity. Multi-step syntheses were employed to synthesize many of the molecules. Nuclear magnetic resonance spectroscopy, UV-visible spectroscopy, high performance liquid chromatography, and mass spectrometry are being used for characterization. The laboratory of Dr. Earl Harrison (Dept. of Nutrition) is currently studying the interaction of the eccentric cleavage products with the nuclear retinoic acid receptors (RARs), which are responsible for cell differentiation and aspects of the immune response. The interaction of these molecules with other nuclear receptors will be analyzed in the future. The samples will also be used as standards to quantitate the concentrations of eccentric cleavage products in biological samples. The synthesis of the molecules prepared to date will be described as well as aspects of the limited biological activity data gathered to date.

Book excerpt: Carotenoid cleavage dioxygenases (CCDs) are a group of enzymes that participate in carotenoid catabolism, with CCD1 and CCD4 involved in norisoprenoid production. Norisoprenoids, breakdown products of carotenoids, are volatile compounds with low sensory thresholds contributing to the aroma of flowers and fruits, including grape Vitis vinifera. Due to the uneven ripening nature of grape berries, a mean value of pooled samples on a given day may not reflect such variation. A single berry approach was performed to relate the gene expression of VvCCD1, VvCCD4a, and VvCCD4b with the firmness of grape berries, making a direct linkage between gene expressions of one berry to the firmness measurement of that berry. VvCCD1 and VvCCD4b gene expression were found to be negatively correlated with firmness under certain conditions, while VvCCD4a expression did not correlate with firmness. A series of carotenoid-producing Escherichia coli cell lines were used to reveal the substrate specificity of VvCCD1, VvCCD4a, and VvCCD4b in vitro. In this study, VvCCD1 showed cleaving activity at the 9-10 (9'-10') carbon position towards cyclic ends of [beta]-carotene, [delta]-carotene, [epsilon]-carotene, and [alpha]-zeacarotene. VvCCD1 also showed 5'-6' carbon position cleaving activity towards the acyclic end for [delta]-carotene, while for [alpha]-zeacarotene the cleaving activity to the acyclic end was found towards both the 5'-6' and 9'-10' carbon positions. Of the carotenoids tested, VvCCD4b was found only capable of cleaving [alpha]-zeacarotene at the 9-10 carbon position, while VvCCD4a did not utilize any of the substrates tested.
