In Mechanization of the Heart: Harvey and Descartes Thomas Fuchs begins by comparing the views of William Harvey [1578-1657] and Rene Descartes [1596-1650] on the heart and the circulation of the blood through the body. These two seventeenth-century scholars -- one a British medical doctor, the other a French philosopher and mathemetician -- differed substantially in their beliefs: they both accepted the idea of circulation of the blood, but differed on the action of the heart. Fuchs traces the ways the opposing views were received, revised, rejected, or renewed in succeeding generations by medical writers in various parts of Europe. He then examines Harvey's approach to cardiac and circulatory physiology, mainly through an examination of Harvey's book De motu cordis: he follows with a discussion of the background in Aristotelian philosophy that was the requirement for all studies in medicineand how that affected Harvey's beliefs. Fuchs then turns to Descartes's presentation of Harvey's views and shows how his view, rather than Harvey's, was accepted in Europe at that time. Marjorie Grene brings to the translation herdistinguished background in philosophy and her keen insights into medical philosophy. Thomas Fuchs teaches psychiatry at the Rupert-Karls-Universitat, Heidelberg. MarjorieGrene is Professor Emeritus of Philosophy at the University of California at Davis, and Adjunct Professor and Honorary Distinguished Professor of Philosophy at Virginia Tech University.

DJ. ter Keurs, was held in Banff Centre, Banff, Alberta, Canada during June 26 to June 30, 1991. The theme of these symposia has been to recognize the similarities and dissimilarities of excitation-contraction coupling in skeletal, cardiac, and smooth muscle. Cross fertilization of concepts of excitation-contraction coupling in these three types of muscle has occurred since the early studies in the late fifties and early sixties on skeletal muscle. Investigators in each field meet only at specialized symposia which exclude investigators in the other fields. The purpose of the symposia has been to bring together international investigators studying excitation-contraction coupling in skeletal, cardiac, and smooth muscle so that we may learn from each other and hence provide a more global concept of excitation-contraction. The Third International Symposia has accomplished its objective as we recognize that calcium channels of the sarcolemma and the sarcoplasmic reticulum play key essential roles in excitation-contraction coupling in all three types of muscles. In skeletal muscle the recognition that E-C coupling consists of two parallel mechanisms, one dependent upon a dihydropyridine voltage-sensitive sensors coupled to calcium release from the terminal cisternae via the ryanodine sensitive channel in the foot structure of the triad.

Since the discovery that the organelle has its own DNA and specific mutations were found in association with neuromuscular and cardiovascular diseases and with aging, an extraordi-nary number of publications have followed, and the term mitochondrial medicine was coined. Furthermore, our understanding of the multiple roles that mitochondria play in cardiac cell homeostasis opened the door for intensive experimentation to understand the pathogenesis and to find new treatments for cardiovascular diseases. Besides its role in adenosine triphosphate generation, mitochondria regu-late a complex network of cellular interactions, involving (1) generation and detoxification of reactive oxygen species, including superoxide anion, hy-drogen peroxide, and hydroxyl radical; (2) maintenance of the antioxidant glutathione in a reduced state and adequate level of mitochondrial matrix superoxide dismutase; (3) cytoplasmic calcium homeostasis, particularly under conditions of cellular calcium loading; (4) transport of metabolites between cytoplasm and matrix; (5) both programmed (apoptosis) and necrotic cell death; and (6) cell growth and development. It is therefore not surprising that this organelle has come to be the center stage in many current investigations of cardiovascular diseases, aging, and agi- related disease. Concomitant with these advances, an impressive effort is under- way for the development of new tools and methodologies to study mitochondrial structure and function, including powerful ways to visualize, monitor, and alter the organelle function to assess the genetic consequences of these perturbations.
