Book excerpt: Since the publication of my previous monograph in 1974, important progress has been made in the broad area of calcium research, partic ularly as it pertains to secretory phenomena. The significant advance in methodology, while widening the scope of our knowledge, has caused research in this particular area to become more and more specialized. It has, therefore, become increasingly difficult for researchers to consider and evaluate work outside their own areas of specialization and to com prehend the field from a broad perspective. While many valuable re views on the importance of calcium in cellular function are being pro duced, they have not attempted to pull together all of the existing knowledge into a more general narrative. This volume brings together experimental data and theories from diverse sources, and attempts to synthesize into a broad conceptual framework the massive amount of specialized information that presently exists in the general area of cal cium metabolism and the secretory process. It is impossible in a book of this length to discuss specific references in detail and still maintain readability, so generalizations must be made.

Book excerpt: The endothelium, a monolayer of endothelial cells, constitutes the inner cellular lining of the blood vessels (arteries, veins and capillaries) and the lymphatic system, and therefore is in direct contact with the blood/lymph and the circulating cells. The endothelium is a major player in the control of blood fluidity, platelet aggregation and vascular tone, a major actor in the regulation of immunology, inflammation and angiogenesis, and an important metabolizing and an endocrine organ. Endothelial cells controls vascular tone, and thereby blood flow, by synthesizing and releasing relaxing and contracting factors such as nitric oxide, metabolites of arachidonic acid via the cyclooxygenases, lipoxygenases and cytochrome P450 pathways, various peptides (endothelin, urotensin, CNP, adrenomedullin, etc.), adenosine, purines, reactive oxygen species and so on. Additionally, endothelial ectoenzymes are required steps in the generation of vasoactive hormones such as angiotensin II. An endothelial dysfunction linked to an imbalance in the synthesis and/or the release of these various endothelial factors may explain the initiation of cardiovascular pathologies (from hypertension to atherosclerosis) or their development and perpetuation. Book excerpt: An enormous amount of research effort has been directed toward elucidating the mechanism by which substances are extruded from cells; and reviews have been written and symposia held in order to systematize the plethora of evidence made available. However, the approaches employed to study the secretory process have been so diverse that it is difficult, if not impossible, for one individual or even a group of individuals to keep abreast of all aspects of the field and to analyze them critically. Thus I undertook the writing of this volume with a great deal of trepidation. In searching for some starting point, I naturally considered as my primary focus the role of calcium in the secretory process, which has occupied my research interests for the past 13 years. But since so much experimentation has been carried out on this and related topics during the last decade or two, I felt it was still necessary to visualize this venture from two alternative ap proaches: (1) a more general one, which would cover the subject of calcium and the secretory process from a broad perspective, but of course not in great detail, and (2) a more specific one, restricting coverage to carefully defined limits but with comprehensive analysis of limited topics. The final course undertaken appears to lie somewhere between these two extremes.

The American Society for Pharmacology and Experimental Therapeutics had the primary responsibility for organizing the scientific program with the assistance of other member societies, including the American Physiology Society, American Association of Pathologists, and American Institute of Nutrition. The purpose of the Calcium Theme was to review progress in the diverse areas of investigation bearing on the ubiquitous role of calcium in biological systems. In addition to contributions from those participating in the Theme, this volume also includes a number of invited papers that were added to fill certain voids in topics covered. The authors were selected because they are investigators active in the mainstream of their particular research area, possessing the acumen to analyze cogently not only their own recent findings but also to relate these findings to their respective area. New information as well as reviews of current concepts generally highlight the individual contributions.
