The synthesis of these amino acids is often time-consuming, non-trivial and not sustainable. Thus, the central topic of this work was the formation of unusual amino acid structures by biocatalytic and catalytic methods. For the elucidation of the biosynthetic pathway of Streptomyces-produced muraymycins and related natural products, a ‘biosynthetic tool kit’ was compiled and prepared. This does not only comprehend the completed synthesis of deuterium-labeled compounds, such as (3R)- and (3S)-3-hydroxy-[5-2H]-l-arginine and other potential intermediates with a nucleosidic structure, but it also includes the development of fermentation methodology including the possibility to detect the muraymycin derivatives by LC MS. Furthermore, different synthetic routes with catalytic key steps for the preparation of the non-proteinogenic amino acid enduracididine were investigated.

Enzymatic transformations carried out by enzymes or whole-cell catalysts are used for the production of a wide variety of compounds ranging from bulk to fine chemicals. The primary consideration for the incorporation of biotransformation in a synthetic sequence is regio- and stereocontrol that can be achieved with enzyme-catalyzed reactions. Biotransformations are thus becoming accepted as a method for generating optically pure compounds as well as for developing efficient routes to target compounds. This Special Issue aims to address the main applications of biocatalysts, isolated enzymes, and whole microorganisms in the synthesis of bioactive compounds and their precursors.

Book excerpt: Through the course of evolution, Nature has developed a swath of useful enzymes for catalyzing chemical reactions crucial to life. These enzymes are used in complex metabolic cascades to produce a wide variety of useful natural products in a chemo-, regio-, and stereoselective fashion. These enzymes can be adopted in synthetic chemistry contexts to afford powerful tools for synthesizing complex molecules in a green way. Through protein engineering, it is possible to further expand the applicability of these enzymes, and by using computational modelling tools, it is possible to accelerate the engineering process. Here, I discuss my efforts in engineering two useful enzymes pyridoxal 5'-phosphate-dependent enzymes, UstD and RgnTDC, to expand their substrate scopes and applicability. UstD is capable of directly installing an amino acid moiety into electrophilic aldehyde substrates through a decarboxylative aldol-like reaction from L-aspartic acid. Through multiple rounds of directed evolution, including linear regression guided evolution, we were able to improve the activity and expressibility of UstD. Through mechanistic studies and crystallography, we have begun probing the complex catalytic cycle that enables UstD to perform its catalyzed reaction, as well as laying the foundation for structure guided evolution. Complementary to UstD, RgnTDC is a l-tryptophan decarboxylase which is able to decarboxylate amino acids and generate precursors to medicinally relevant compounds. Through the use of Substrate Multiplexed Screening (SUMS) in conjunction with logistic regression guided directed evolution and primer-doped mutagenesis, we have created a variety of generalist biocatalysts with diverse active site composition. These variants serve as potential starting points for future engineering campaigns. To help facilitate these future engineering efforts and explore the utility of SUMS, I have also worked to develop computational tools to process and analyze SUMS library screening data. These tools have been integrated into a single web-based program, titled the SUMS Interactive Processing Software (SIPS), and have been developed with broad applicability and community use in mind.

The first application of these remarkable compounds to produce ethanol by fermentation is lost to antiquity. Payer and Persoz (Ann. Chim. Phys. , 53, 73 (1833ii)) appear to have provided the first step toward understanding this com plex area when they reported the isolation of diastase in 1833. These workers showed that diastase could catalyze the hydrolysis of starches to sugars. Somewhat earlier Kirchhoff (Schwigger's Journal, 4, 108 (1812)) had shown that a small amount of dilute acid could hydrolyze a seemingly endless amount of starch to sugars. The genius of Berzelius recognized the commonality of these two observations in connection with a few other isolated observations and in 1834 coined the term catalysis to describe such actions. Professor Leibig was one of the giants of the chemical world in 1840. In addition to his own work, Liebig was training the world's next generation of chemists in his laboratory in Giessen. This cadre of chemists were very impressed by the master teacher so that is it only natural that Liebig's views should dominate with this next generation of chemists. Leibig was, in the 1830s and 1840s, developing his mastery of agricultural chemistry. The mechanism of putrefication was of great concern to Leibig, and he turned to the newly defined area of catalysis for an explanation.

Book excerpt: Research in to enzyme catalysis has enormously enriched the success of biological organic synthesis; particularly in the last decade, remarkable progress in biological catalysis has taken place leading to the application of enzymes to a wider extent in industrial processes. The benefits of establishing enzymes as industrial catalysts are founded on the need to develop a 'clean' and 'pure' technology which carry out isomer and region selective reactions; generate or transform pure isomers compared with racemic mixtures and produce pure compounds in comparison with mixtures of by-products often arising from conventional chemical synthesis. With the onset of molecular biology and genetics, the manufacturing of high value proteins in recombinant host cells has become commonplace; nevertheless, there needs to be a better understanding of the optimum conditions for the production of every recombinant protein. In this study, the over expression of an industrially important amine oxidase - D-Amino Acid Oxidase (DAAO) - was attempted using two different expression systems: Escherichia coli and Pichia pastoris in order to establish the optimum conditions for industrial production. Amine oxidases exhibit stereoselectivity; ergo they can theoretically be used for the deracemisation of nonopticallypure mixtures of amines leading to optically pure amines of high value.Furthermore, DAAO is involved in the production of 7-aminocephalosporanic acid (7-ACA) from cephalosporin c (CPC), which can be used in the synthesis of novel cephalosporin antibiotic derivatives. In order to accelerate development of these new antibiotics DAAO must be readily available. Overexpression of DAAO in E. coli was studied under varying experimental conditions in order to ascertain the best conditions for enzymatic activity whilst simultaneously investigating the variations between both defined and complex media to understand suitability for scale-up.Although E. coli is still regarded as the 'go to' organisms for initial laboratory scale up the methylotrophic yeast P. pastoris is considered an industrial workhorse for the production of recombinant proteins and thus the expression of DAAO was also investigated to ascertain how enzyme activities are affected by this expression system. Furthermore, the supplementation of media with different nutrient sources of varying complexity was investigated, as was the implementation of co-feeding inorder to improve carbon flux during recombinant protein production. Finally, the oxygen availability was investigated in order to gain important understandings on its effects on microbial physiology and DAAO activity.
