    Book Details:
  • Author : Peter Quinn
  • Publisher : Springer Science & Business Media
  • Release : 2008-08-27
  • ISBN : 1402088310
  • Pages : 603 pages

Download or read book Lipids in Health and Disease written by Peter Quinn and published by Springer Science & Business Media. This book was released on 2008-08-27 with total page 603 pages. Available in PDF, EPUB and Kindle. Book excerpt: Lipids are functionally versatile molecules. They have evolved from relatively simple hydrocarbons that serve as depot storages of metabolites and barriers to the permeation of solutes into complex compounds that perform a variety of signalling functions in higher organisms. This volume is devoted to the polar lipids and their constituents. We have omitted the neutral lipids like fats and oils because their function is generally to act as deposits of metabolizable substrates. The sterols are also outside the scope of the present volume and the reader is referred to volume 28 of this series which is the subject of cholesterol. The polar lipids are comprised of fatty acids attached to either glycerol or sphingosine. The fatty acids themselves constitute an important reservoir of substrates for conversion into families of signalling and modulating molecules including the eicosanoids amongst which are the prostaglandins, thromboxanes and leucotrienes. The way fatty acid metabolism is regulated in the liver and how fatty acids are desaturated are subjects considered in the first part of this volume. This section also deals with the modulation of protein function and inflammation by unsaturated fatty acids and their derivatives. New insights into the role of fatty acid synthesis and eicosenoid function in tumour progression and metastasis are presented.

Book Building a Metabolic Bridge Between Glycolysis and Sphingolipid Biosynthesis

Download or read book Building a Metabolic Bridge Between Glycolysis and Sphingolipid Biosynthesis written by Morgan L. Stathem and published by . This book was released on 2014 with total page 59 pages. Available in PDF, EPUB and Kindle. Book excerpt: Cancer therapeutics has seen an emergence and re-emergence of two metabolic fields in recent years, namely bioactive sphingolipids and glycolytic metabolism. Anaerobic glycolysis and its role in cancer has been recognized in cancer biology over 90 years. In recent decades, the role of sphingolipids in cancer cell metabolism has gained recognition, notably ceramide's essential role in programmed cell death and the role of the glucosylceramide synthase (GCS) in chemotherapeutic resistance. Despite this knowledge, a direct link between these two fields has yet to be concretely drawn. Herein, we show that in a model of highly glycolytic cells, generation of the glycosphingolipid (GSL) glucosylceramide (GlcCer) by GCS was elevated in response to increased glucose availability, while glucose deprivation diminished GSL levels. This effect was likely substrate dependent, independent of both GCS levels and activity. Conversely, leukemia cells with elevated GSLs showed a significant change in GCS activity, but no change in glucose uptake or GCS expression. In a leukemia cell line with elevated GlcCer, treatment with inhibitors of glycolysis or the pentose phosphate pathway significantly decreased GlcCer levels. When combined with pre-clinical inhibitor ABT-263, this effect was augmented and production of proapoptotic sphingolipid ceramide increased. Taken together, we have shown that there exists a definitive link between glucose metabolism and GSL production, laying the groundwork for connecting two distinct yet essential metabolic fields in cancer research. Furthermore, we have proposed a novel combination therapeutic option targeting two metabolic vulnerabilities for the treatment of leukemia.

Download or read book Fumonisins in Food written by Lauren S. Jackson and published by Springer Science & Business Media. This book was released on 2013-11-21 with total page 399 pages. Available in PDF, EPUB and Kindle. Book excerpt: The contents of this book are the proceedings of the ACS symposium, "Fumonisins in Food," which was held April 4-6, 1995, at the American Chemical Society National Meeting in Anaheim, CA. This symposi"Qm, which was international in scope, brought together researchers from diverse backgrounds in academia, government, and industry. Thirty-three speakers discussed topics ranging from the analysis offumonisins to toxicology and regulatory aspects. The fumonisins became the spotlight of mycotoxin research in 1988, when re searchers at the South African Medical Research Council isolated and structurally charac terized the fumonisins. Since 1988, there has been an explosion in the numbers of papers dealing with fumonisin-related topics. The interest in the fumonisins has arisen for several reasons. First, fumonisins are found in measurable concentrations in corn grown throughout the world. Second, these compounds have been implicated as the causative agents in a variety of naturally occurring animal diseases. Finally, there is speCUlation that fumonisins may in part be responsible for the high incidence of esophageal cancer in regions of the world in which corn is the staple grain.
