Book excerpt: "The objective of the present study was to investigate the genetic and migratory differences among White-throated Sparrow populations in western Canada in order to locate the presence of a migratory divide. Deuterium isotopes and molecular markers were used to assess the migratory differences between sparrows west (Central Interior BC) and east (Peace Region BC) of the Continental Divide. Head feather isotopes showed that both populations are overwinter wintering in either the Pacific Coast, New Mexico/Arizona or Colorado/Kansas areas. Microsatellites and mitochondrial markers did not show genetic structure among populations, however tail feather isotopes were significantly different. Analysis of migratory samples is congruent with Peace region birds migrating east of the divide. The Central Interior birds were not detected in any migratory monitoring locations. Data of the present study is congruent with a migratory divide and an east/west migration pattern between Central Interior and the Peace Region populations."--P. ii.

Book excerpt: Effective conservation management of migratory birds can be challenging and requires knowledge about population structure and the strength of migratory connectivity. The latter likely affects the degree to which populations are locally adapted and differentiated, and both may affect their ability to adapt to environmental changes. Western Sandpipers (Calidris mauri) are long-distance migrants that exhibit latitudinal differences in morphology and life-history strategies across their non-breeding range. The differential migration patterns in this small shorebird species could be based on population genetic differences or phenotypic plasticity. I investigated the population structure of Western Sandpipers, and its implications for differential migration and conservation, across their global range using genetic, morphometric and acoustic data. I recorded and analyzed breeding male vocalizations, conducted playback experiments and collected genetic and morphometric data across breeding and non-breeding ranges. Amplified Fragment Length Polymorphisms (AFLP) and mtDNA sequence data indicated low, but significant genetic population structure across breeding and non-breeding ranges. I found evidence for an evolutionarily recent demographic expansion (mtDNA), for latitudinal clines in genetic scores (AFLP) and for isolation by distance (AFLP). A scan of thousands of AFLP markers in a pooled lane approach revealed no fixed genetic differences among juveniles with a postulated difference in life-history strategy or migratory direction. Songs, but not alarm calls, varied geographically, decreasing in length and increasing in fundamental frequencies from southern to northern breeding sites. These geographic differences were sufficiently large to allow males to discriminate between local and non-local songs and hence are potentially biologically relevant. AFLP scores and vocalization frequencies showed correlations with body size. In conjunction, my results suggest that Western Sandpipers underwent a recent range and population expansion that has resulted in divergence patterns that are primarily isolation by distance driven. My results suggest interrelationships among genetic population structure, morphology and latitudinal segregation. While populations were not sufficiently distinct to allow for assignment tests, the latitudinal gradients found across both the breeding and non-breeding grounds are suggestive of a gradual 'chain'-like migration pattern, but do not indicate strong migratory connectivity. Consequently, conservation strategies could focus on the protection of major sites at different latitudes.

Book excerpt: Understanding migratory connectivity is essential for assessing the drivers behind population dynamics and for implementing effective management in migratory species. Genetic markers provide a means to describe migratory connectivity, as well as incorporate population genetic analyses, however genetic markers can be uninformative for species with weak genetic structure. In this study, we evaluate range-wide population genetic structure and migratory connectivity in the prothonotary warbler, Protonotaria citrea, a wetland-dependent neotropical migratory songbird, using high-resolution genetic markers. We reveal regional genetic structure between sampling sites in the Mississippi River Valley and the Atlantic Seaboard with overall weak genetic differentiation among populations (FST = 0.0051). By ranking loci by FST and using subsets of the most differentiated genetic markers (200 -- 3000), we identify a maximum assignment accuracy (89.7% to site, 94.3% to region) using 600 single nucleotide polymorphisms. We assign samples from unknown origin nonbreeding sites to a breeding region, illustrating weak migratory connectivity between prothonotary warbler breeding and nonbreeding grounds. Our results highlight the importance of using high-resolution markers in studies of migratory connectivity with species exhibiting weak genetic structure. Using similar techniques, studies may begin to describe population genetic structure that was previously undocumented, allowing us to infer the migratory patterns of an increasing number of species.

Book excerpt: In this body of work, I examine how testosterone (T) physiology mediates the life-history trade-off between mating effort and parental care in the White-throated Sparrow. This species exhibits a behavioral polymorphism that occurs in both sexes. White-striped (WS) morphs are more territorially aggressive, sing more frequently and seek more extra-pair copulations. Tan-striped (TS) morphs provision nestlings more frequently. Thus this species roughly illustrates the trade-off between mating effort and parental care. I examine T physiology on three levels: plasma titres, binding globulins and response to the social environment. I ask whether levels of T correlate with morph-specific behavior and does this relationship change with stage in the nesting cycle. I found that WS males have significantly higher plasma levels of T than TS males. This difference is small, but it persisted through the parental stage of the nesting cycle. This suggests that T may mediate differences in mating effort and parental behavior in males, but is likely not the only factor. Female morphs did not differ in plasma T, thus T does not appear to play a similar role in females. Next I ask how corticosterone binding globulin (CBG) modulates T action. I found that CBG binds over 90% of T and is an important modulator of T action in this species. However CBG capacity did not differ between morphs, nor did morphs differ in baseline levels of corticosterone (CORT, a stress hormone that competes with T for binding sites on CBG.) Therefore interactions with CBG and CORT do not affect T action differently in the morphs, and patterns of free T (T not bound to CBG) mirror patterns of total T. Finally I investigated how T physiology responds to a change in the social environment- the establishment of a dominance relationship. WS males exhibited aggression more frequently and tended to dominate TS males. Levels of total T, CBG, CORT and free T were not predictive of future dominance status. Nor did these measures show persistent changes once the dominance relationship was established. The response of T physiology to the formation of a dominance relationship did not differ between morphs.
