Book A Basic Study to Assess the Potential Usefulness of Resonance Raman Spectroscopy as a Means of Rapidly Detecting and Identifying Bacteria and Other Microorganisms

Download or read book A Basic Study to Assess the Potential Usefulness of Resonance Raman Spectroscopy as a Means of Rapidly Detecting and Identifying Bacteria and Other Microorganisms written by W. H. Nelson and published by . This book was released on 1985 with total page 15 pages. Available in PDF, EPUB and Kindle. Book excerpt: This past year we have explored the potential uses of Resonance Raman and Fluorescence Lifetime Spectroscopies in the rapid characterization of bacteria. Previously we have shown that chromobacteria can be distinguished on the basis of distinctive resonance Raman spectra. Spectra are excited by low power argon ion laser radiation at 488 nm and are due to the presence of carotenoid pigments. While carotenoid pigments in bacteria generally are not useful for identification purposes, the study of chromobacteria has allowed us to assess the potential sensitivity of resonance Raman spectroscopy in this application. We were able to laser illuminate and count two types of bacteria under a microscope. Because it was possible to see the laser excited bacteria while the resonance Raman spectra were being obtained, it was possible to estimate closely the number of bacteria producing a given spectrum. Such high sensitivity suggests but does not prove that spectra are 'surface enhanced'. It is clear that the resonance Raman technique can be highly sensitive and our experiments suggest that remote detection and detection from mixtures is possible in principle.

Book The Rapid Detection of Bacteria and Other Microorganisms A Basic Study in the Application of Resonance Raman and Time Resolved Fluorescence Spectroscopies

Download or read book The Rapid Detection of Bacteria and Other Microorganisms A Basic Study in the Application of Resonance Raman and Time Resolved Fluorescence Spectroscopies written by Wilfred H. Nelson and published by . This book was released on 1988 with total page 22 pages. Available in PDF, EPUB and Kindle. Book excerpt: Resonance Raman Spectra have been obtained for a variety of chromobacteria using low power 488 nm excitation. Spectra are simple, of high quality, and useful for identification purposes at the species level. Raman microprobe studies show conclusively that spectra can be obtained from single cells in pure cultures or in mixed cultures without need for separation. Extensions of the study have been made to representative colorless gram-negative and gram-positive bacteria. Spora other than bacteria have been studied as well. Pollen, mold spores, bacterial spores, algae and viruses all give spectra but only viruses and bacterial spores appear to give intense UV Resonance Raman spectra. The primary fluorescence of bacteria has been studied in detail to determined its potential in rapid detection. We have determined fluorescence for S. epidermidis, P. fluorescens, E. cloacae, E. coli and B. subtilis. Fluorescence contributions have been assigned in part to tryptophan, pteridines, related flavins and pyridine coenzymes.

Download or read book Raman Spectroscopy written by Gustavo Morari Do Nascimento and published by BoD – Books on Demand. This book was released on 2018-04-18 with total page 326 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book gives a wide overview of the state-of-the-art applications of Raman spectroscopy in characterization of materials and biomaterials. The Raman signal is intrinsically smaller than other vibrational techniques; however, mainly through intensification processes, such as resonance Raman (RR) and surface-enhanced Raman spectroscopy (SERS), the Raman cross section can be strongly amplified. Thoroughly in these signal amplifications, the study of a diversity of chemical systems and the use of Raman technique for in situ and in vivo measurements is possible. The main goal of this book is to open up to an extended audience the possibilities of uses of Raman spectroscopy. In fact, this collective work will be beneficial to students, teachers, and researchers of many areas who are interested to expand their knowledge about Raman spectroscopy applied to nanotechnology, biotechnology, environmental science, inorganic chemistry, and health sciences.

Book Biochemical Applications of Raman and Resonance Raman Spectroscopes

Download or read book Biochemical Applications of Raman and Resonance Raman Spectroscopes written by P Carey and published by Elsevier. This book was released on 2012-12-02 with total page 277 pages. Available in PDF, EPUB and Kindle. Book excerpt: Biochemical Applications of Raman and Resonance Raman Spectroscopies focuses on the application of Raman and resonance Raman spectroscopies to biochemical problems. The book reviews biological systems and details the application of Raman spectroscopy to biological molecules such as proteins, nucleic acids, and lipids. It also looks at codevelopments of lasers, optics, and electronics that drive advances in experimental Raman spectroscopy, along with the important ramifications of these advances for biochemical applications. This volume is organized into eight chapters and begins with an overview of the theoretical and experimental aspects of Raman spectroscopy, including a very brief explanation of what Raman and resonance Raman spectroscopies are and a discussion of their advantages and disadvantages for biochemical studies. The explanation of the Raman and resonance Raman effects is taken up in more detail in the next chapter, which develops the concept of the vibrational motions of molecules by initially considering mechanical ""ball and spring"" models and goes on to use this concept to formulate a classical model for Raman scattering. The resonance Raman effect is then described by another model which emphasizes the discrete or quantized energy levels available to a molecule. The reader is also introduced to the experimental aspects of Raman spectroscopy and the application of Raman spectroscopy across the entire field of biochemistry. Each chapter contains an outline of the basic chemistry and biochemical nomenclature involved. This book will be of interest to chemists, biochemists, and spectroscopists, as well as graduate students and experienced research workers.

Book Application of Raman Spectroscopy for Studying Bacteria of the Family Enterobacteriaceae

Download or read book Application of Raman Spectroscopy for Studying Bacteria of the Family Enterobacteriaceae written by Amir Nakar and published by . This book was released on 2023* with total page 0 pages. Available in PDF, EPUB and Kindle. Book excerpt: This thesis investigated using Raman spectroscopy for identifying Enterobacteriaceae and their antibiotic resistances. The study aimed to develop a rapid, accurate diagnostic tool, as current methods are costly and inefficient, leading to increased antibiotic resistance due to inappropriate antibiotic use. The research involved collecting a dataset of Enterobacteriaceae, encompassing 8 genera. Measurements were taken using Single-Cell Raman Microspectroscopy (SC-RMS) and Fiber Probe-Based Raman Spectroscopy (FP-RS). Machine learning models were trained to classify the bacteria based on spectral data. SC-RMS showed limited success (47% accuracy), while FP-RS achieved 100% accuracy on the training dataset and 94% on a validation dataset, demonstrating its potential in differentiating Enterobacteriaceae. Subsequently, Raman spectra of 24 clinical isolates were analyzed using UV-Resonance Raman Spectroscopy (UVRRS). The goal was to discriminate between E. coli, K. pneumoniae, and K. oxytoca. While SC-RMS had limited effectiveness, UVRRS achieved high accuracy (92% at the genus level and 90% at the species level), marking the first successful differentiation of K. oxytoca using Raman spectroscopy. The study also explored differentiating antibiotic-resistant and sensitive Enterobacteriaceae. Both SC-RMS and UVRRS correctly classified 70-75% of the isolates, with UVRRS demonstrating a 90% sensitivity for resistant strains. This differentiation may be related to a higher DNA/Protein ratio in resistant bacteria. Overall, the findings enhance the application of Raman spectroscopy in microbiology, particularly for Enterobacteriaceae. The research supports its use in identifying pathogens, determining their genus and species, and assessing antibiotic resistance. The study contributes significantly to the development of Raman-based microbial diagnostics, promising to improve infectious disease diagnostics, patient care, and the fight against antimicrobial resistance.

Book Infrared and Raman Spectroscopy

Download or read book Infrared and Raman Spectroscopy written by Günter G. Hoffmann and published by Walter de Gruyter GmbH & Co KG. This book was released on 2023-09-18 with total page 446 pages. Available in PDF, EPUB and Kindle. Book excerpt: Quite a few excellent books about vibrational spectroscopy have already been published. So why write a new one? The last years have seen the birth of new techniques and, first of all, a wealth of new applications. Therefore, a lot of new users need an introduction to these techniques and applications, but, if they are new to vibrational spectroscopy, an introduction to the parent techniques as well. Vibrational spectroscopies can detect and analyze vibrations in molecules. Mainly two different forms are used today: Infrared and Raman spectroscopy. Vibrational spectroscopy is used by chemists to characterize their substances. If the spectra of substances are known, analytical chemists can use them to analyze a mixture of chemicals. Samples may be analyzed even with spatial resolution, on the microscopic as well as on the macroscopic scale. "Infrared and Raman Spectroscopy" is intended for researchers or lecturers in Chemistry, Physics, Materials Science and Life Sciences, who are interested in the composition and properties of their samples. It describes how vibrational spectroscopy will enable them to examine thin layers, surfaces and interfaces, and also improve their knowledge about the properties of composites. Special chapters introduce VCD, ROA, and TERS. The book can serve as a short introduction to vibrational spectroscopy too, so that students at the first graduate level will benefit from it as well.
