Book excerpt: A new in vivo model which gives a more accurate picture of cellular inflammation was developed. It involves a subcutaneously implanted plastic cup which can easily be used to assess quantitatively dynamic exudate cell migration. The use of this model in normal and irradiated rats has provided new insights into the kinetics of exudate cells during acute inflammation. Under the conditions described in this study, it appears that mononuclear cells are the dominant type cells in inflammatory exudates much earlier than has been previously reported in studies using less quantitative models.

Jacobson (9), and (2) by treatment with bone marrow suspensions by E. Lorenz and his col laborators (12). The site chosen for this annual meeting of the International Society for Experi mental Hematology is given a special historic sig nificance by the fact that it was 25 years ago that the first publication on this subject by Lorenz ap peared from his laboratory at the National Insti tutes of Health. Lorenz's discovery marked the beginning of a period which lasted until 1956, during which the protection afforded by hemopoietic cell suspensions was confirmed by many. This soon led to an intensive scientific de bate on the mechanism of this protective effect: was it due to a humoral factor produced and pro vided by the bone marrow-as Lorenz The Appearance of postulated-or to transplantation and subsequent proliferation of hemop- etic cells? This question was defini- 1 the Multipotential tively answered in 1956 by evidence from three different laboratories (7, 15, 26), which demonstrated the origin of the cells Hemopoietic in the repopulated tissues using a variety of cellu lar and immunologic markers. By the same token, these contributions marked the birth of radiation Stem Cell chimeras.

It is suggested that many diseases are initiated and their progress is influenced by inflammatory molecules and a decrease in the production and/or action of anti-inflammatory molecules and this imbalance between pro- and anti-inflammatory molecules seems to have been initiated in the perinatal period. This implies that strategies to prevent and manage various adult diseases should start in the perinatal period. An alteration in the metaolism of essential fatty acids and their anti-inflammatory molecules such as lipoxins, resolvins, protecitns, maresins and nitrolipids seems to play a major role in the pathobiology of several adult diseases. Based on these concepts, novel therapeutic approaches in the management of insulin resistance, obesity, type 2 diabetes mellitus, metabolic syndrome, cancer, lupus, rheumatoid arthritis and other auto-immune diseases are presented. Based on all these evidences, a unified concept that several adult diseases are due to an alteration in the balance between pro- and anti-inflammatory molecules is discussed and novel methods of their management are presented.
