The enormous research efforts in this field have been directed toward understanding the nature of the virus and toward its elimination through preventive vaccin ation and the cure of the disease. Human T-cell leukemia virus (HTLV-l) was the first member of the human retrovirus family to be discovered. It was implicated as the cause of adult T-cell leukemia (ATL) even before the association of HIV and AIDS was established. Research on HTL V -1 has, however, been lagging behind that of HIV because of the importance of AIDS. Today HTLV-1 and possibly closely related HTLV-2 are associated with a variety of human neurologic diseases, and research activities in this field may show that human retroviruses can cause a variety of human diseases in addition to those affecting the nervous system. Available in PDF, EPUB and Kindle. Book excerpt: Human retroviruses, HIV and HTLV have been recognized as important pathogens because of their association with lethal diseases such as AIDS and ATL. Considerable resources and efforts have been directed at understanding the interaction between these retroviruses and their host which may provide clues as to how the infection can be controlled or prevented. Among the key scientific successes is the identification of intracellular “restriction factors” that have evolved as obstacles to the replication of pathogens including infectious retroviruses. The discovery of APOBEC, which are strong mutagens of retroviral genomes and intracellular retroelements, began a new era of intense research activities into the spectrum of intrinsic anti-HIV activity, leading to the identification of TRIM5a, BST2/Tetherin, and SAMHD1. In response, HIV has evolved several accessory genes as weaponries to evade these intracellular restriction activities. The intracellular antiretroviral defenses evolved in response to endogenous retroelements that make up more than 40% of the entire mammalian genome, and which are regarded as ancestors of infectious retroviruses. LTR-type retroelements are present in all higher eukaryotes, representing about 8% of the human genome. Non-LTR retroelements can be found at extremely high copy numbers also, with a significant portion of mammalin genomes consisting of LINEs. Mammalian genomes are modified by LINEs through insertions, but also by the indirect replication of non-autonomous retrotransposons such as SINEs. LINEs insertion was shown to have played, and continue to play important roles in genomic evolution and somatic genome mosaicism-mediated physiology. And, because retrotransposition can confer genetic diversity that is beneficial to the host, the vertebrate intrinsic immunity has evolved to support a balance between retroelement insertions that confer beneficial and those that cause deleterious gene disruptions. The articles published in this Research Topic should serve not only as valuable references for the field, but provide future topics of research for investigators that should further our understanding of the retrovirus, retroelements and their restrictions.

Considerable emphasis on prevention of new infections and treatment of those living with HIV will be needed to make this goal achievable. With nearly 37 million people now living with HIV, it is a communicable disease that behaves like a noncommunicable disease. Nutritional management is integral to comprehensive HIV care and treatment. Improved nutritional status and weight gain can increase recovery and strength of individuals living with HIV/AIDS, improve dietary diversity and caloric intake, and improve quality of life. This book highlights evidence-based research linking nutrition and HIV and identifies research gaps to inform the development of guidelines and policies for the United Nations’ Sustainable Development Goals. A comprehensive approach that includes nutritional interventions is likely to maximize the benefit of antiretroviral therapy in preventing HIV disease progression and other adverse outcomes in HIV-infected men and women. Modification of nutritional status has been shown to enhance the quality of life of those suffering HIV/AIDS, both physically in terms of improved body mass index and immunological markers, and psychologically, by improving symptoms of depression. While the primary focus for those infected should remain on antiretroviral treatment and increasing its availability and coverage, improvement of nutritional status plays a complementary role in the management of HIV infection.

Book excerpt: The acquired immunodeficiency syndrome (AIDS) is caused by two retroviruses of the lentivirus subfamily, human immunodeficiency viruses types 1 (HIV-1) and 2 (HIV-2), which derive from cross-species transmissions of viruses naturally infecting two African primate species, the chimpanzee (Pan troglodytes) and the sooty mangabey (Cercocebus atys), respectively. Eleven transmission events of lentiviruses from these primate species to humans have been inferred from phylogenetic sequence analyses, although only four (which originated HIV-1 groups M and O, and HIV-2 groups A and B) have resulted in epidemic spread. One of these transmissions, corresponding to the common ancestor of the HIV-1 group M viruses, whose initial expansion in Central Africa has been dated by molecular clock analyses to around 1930, eventually originated the AIDS pandemic. How humans acquired the ancestors of the AIDS viruses from simians is uncertain, but natural infection of African non-human primates with lentiviruses is common, and contact with blood of these animals, which are frequently hunted in Africa for human consumption, is a plausible mechanism for virus acquisition. Sociodemograhic developments, including rapid urbanisation, large scale migrations, growth of prostitution, wars, and increase in travel, and, possibly, the widespread use of non-sterile injection material, may have promoted the emergence of the AIDS pandemic in Central Africa in the 20th century. Subsequent global propagation of HIV-1 derives from multiple introductions at different time points, often representing phylogenetically identifiable founder events. In areas in which diverse HIV-1 variants are cocirculating, these frequently recombine to generate mosaic forms, some of provided new insights on the origin and spread of AIDS viruses and may contribute to the efforts aimed at containing the pandemic which have propagated extensively.
