Book excerpt: The National Institute of Allergy and Infectious Diseases (NIAID) gives the highest priority to developing countermeasures against bioterrorism agents that are highly infective when dispersed in aerosol form. Developing drugs to prevent or treat illnesses caused by bioterrorism agents requires testing their effectiveness in animals since human clinical trials would be unethical. At the request of NIAID, the National Academies conducted a study to examine how such testing could be improved. Overcoming Challenges to Develop Countermeasures Against Aerosolized Bioterrorism Agents provides recommendations to researchers on selecting the kinds of animal models, aerosol generators, and bioterrorism agent doses that would produce conditions that most closely mimic the disease process in humans. It also urges researchers to fully document experimental parameters in the literature so that studies can be reproduced and compared. The book recommends that all unclassified data on bioterrorism agent studies-including unclassified, unpublished data from U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID)-be published in the open literature. The book also calls on the U.S. Food and Drug Administration to improve the process by which bioterrorism countermeasures are approved based on the results of animal studies.

Book excerpt: A draft Manual of Recommended Practice for Aerosol Sampling and Evaluation was completed and sent to the Energy Research and Development Administration (ERDA) Division of Safety, Standards, and Compliance (DSSC) for review. The results of the Survey of Sampling Techniques for Defining Respirable Concentration and/or Particle Size Characteristics of Aerosols were published as LA-6087. The need for greater standardization of ERDA aerosol sampling techniques was indicated. The Aerosol Training Course was presented in 11 sessions to 85 persons. General elements of good practice were emphasized, and recommendation of specific sampling devices or procedures was avoided. A system for estimating dissolution rates of plutonium aerosols was developed. Studies indicate that plutonium aerosols found in the field have a rapid initial dissolution phase followed by a slower secondary phase. Three methods of particle sizing air samples collected on membrane filters were investigated. The most promising was a scanning electron microscope electron microprobe (SEM-EMp) method. An operating plutonium handling facility was a model for development of techniques to evaluate aerosol surveillance systems performance. Airborne contamination records were studied. The physicochemical properties of a plutonium aerosol existing in the facility were investigated in relation to plutonium handling operations. The techniques developed have indicated some areas of the aerosol surveillance system that need improvement.

Book excerpt: Alternative Reference Methodologies (ARMS) have been developed for sampling of radionuclide; from stacks and ducts that differ from the methods required by the US EPA. The EPA methods are prescriptive in selection of sampling locations and in design of sampling probes whereas the alternative methods are performance driven. Tests were conducted in a stack at Los Alamos National Laboratory to demonstrate the efficacy of the ARMS. Coefficients of variation of the velocity tracer gas, and aerosol particle profiles were determined at three sampling locations. Results showed numerical criteria placed upon the coefficients of variation by the ARMs were met at sampling stations located 9 and 14 stack diameters from flow entrance, but not at a location that is 1.5 diameters downstream from the inlet. Experiments were conducted to characterize the transmission of 10[mu]m aerodynamic equivalent diameter liquid aerosol particles through three types of sampling probes. The transmission ratio (ratio of aerosol concentration at the probe exit plane to the concentration in the free stream) was 107% for a 113 L/min (4-cfm) an isokinetic shrouded probe, but only 20% for an isokinetic probe that follows the EPA requirements. A specially designed isokinetic probe showed a transmission ratio of 63%. The shrouded probe performance would conform to the ARM criteria; however, the isokinetic probes would not.
