SeV reverse genetics originating from a pure academic need to settle long-held questions in the biology and pathogenicity of nonsegmented negative strand RNA viruses (Mononegavirales) is about to bear the impressive fruit of multipurpose cytoplasmic (non-integrating) RNA vectors. This book brings together in one source the SeV biology revealed by conventional approaches and reverse genetics, the methods to construct the first-generation SeV vector and to generate safer versions, and the applications in medical settings that have left or are about to leave the laboratory bench. The applications, which already are diverse and have high medical impact, include use as vaccine vectors against AIDS and respiratory virus infections, creation of BioKnife to resect malignant tumors, induction of “footprint (transgene) free” pluripotent stem cells, and gene therapy for peripheral arterial disease. These achievements—which are just a few of many examples—were attainable only after rigorously incorporating the rich knowledge of SeV biology that has accumulated during the several decades since the discovery of the virus. Application of SeV vector is certain to expand greatly because of its extremely high performance in transgene expression and its remarkable target cell breadth.

Book excerpt: This book records the papers and discussions at a Work shop which took place in London on the 5th and 6th of April 1979, as part of the programme of the Commission of the European Communities on Medical and Public Health Research. However the views expressed are those of the individuals concerned and not of the EEC or any of its organs. The object was to discuss certain biological aspects of natural and experimental slow virus infections. Because the amount of knowledge and the focus of interest varied in respect of each infection the approach and emphasis varied also. In the case of scrapie, we discussed the nature of the agent and the mode of pathogenesis, in the case of SSPE, the search for unusual features of the virus, and recent detail ed work on the immunology of the disease. As for Visna we reviewed the present understanding of the virus and its pathogenicity and also field epidemiology and methods for its control. There were also general papers, on interferon and oncornaviruses for example. We thank all those who made the meeting possible and enabled us to produce this book quickly, so that those who could not attend the meeting may nevertheless be able to read a great deal of what went on at it. In particular we would thank the Ciba Foundation who allowed us the use of their premises and Mrs. Jean Ashley who dealt with most of the arrangements.

Animal rhabdoviruses infect and often cause disease in insects, fish, and mammals, including man. The prototype rhabdovirus, vesicular stomatitis virus !VSV), has been extensively studied and provides perhaps the best model system for studying negative-strand viruses. The popularity of VSV as a model system is to a considerable extent due to its relative simplicity and to its rapid growth, generally to high titer, in many cell types ranging from yeast to human. The nucleocapsids of these viruses also carry transcriptional and replicative functions that are expressed in cell-free systems. The first RNA-dependent RNA poly merase was described in VSV and its G protein provided an early model system for studying the synthesis, processing, and membrane insertion of mammalian glycoproteins. VSV is also highly cytopathogenic and has been studied quite extensively for its capacity to kill cells and to shut off cellular macromolecular synthesis. Even earlier, VSV was discovered to be highly susceptible to the action of interferons and has served ever since as a means for quantitating the activity of interferons. To my way of thinking, the spark that ignited the explosion of re search in this field was struck at the First International Colloquium on Rhabdoviruses, attended by 30 or so participants in Roscoff, France, in June 1972.
