However, there is no cure for relapsed androgen withdrawal-refractory prostate cancer which is fatal due to metastasis. Many studies have demonstrated the existence of cancer cells with stem cell properties (aka cancer propagating cells (CPCs)) within the cancer cell population. CPCs may cause cancer recurrence due to resistence to conventional therapies. Understanding molecular and cellular mechanisms governing normal prostate stem cells and CPCs may improve our understanding and provide new opportunities for the design of therapeutics acting specifically on CPCs. Two of such mechanisms, p53/microRNA-34/Met network and neuroendocrine (NE) signaling are the topic of my dissertation. microRNA-34 (miR-34) family has been proposed to play a tumor suppressor role in various cancers, including the prostate cancer. However, direct genetic evidence for the tumor suppressor functions of miR-34 under physiologically relevant settings has been lacking. By using newly generated mice carrying conditional alleles of mir-34, we have shown that miR-34 cooperates with p53 in suppression of prostate carcinogenesis by joint control of MET-dependent prostate stem/progenitor cells. Thus, therapeutic targeting of MET is likely to affect CPCs, particularly in p53 and miR-34 deficient patients. NE signaling has been shown to be one of the features in advanced prostate cancer. However, the roles of NE cells in prostate carcinogenesis and development are insufficiently elucidated. By using bacterial artificial chromosome (BAC) we have generated a mouse model for conditional NE cell ablation, and shown that prostate epithelium-specific ablation of NE cells results in prostate hypotrophy, suggesting NE cells play an important role in prostate development. Moreover, we have studied effects of neuropeptide in membrane metallo-endopeptidase (Mme) null mice, and shown that Mme cooperates with Pten to suppress prostate carcinogenesis in the control of prostate stem/progenitor cells. Gastrin-releasing peptide (GRP), a substrate of MME, showed similar effects on mouse prostate stem/progenitor cells and human prostate CPCs. These effects were effectively abrogated by GRP receptor (GRPR) antagonist or knockdown of GRPR expression. Taken together, these findings show critical role of NE signaling in prostate development and carcinogenesis and GRPR is a potential therapeutics targeting prostate CPCs.

With the advent of the prostate-specific antigen (PSA) test, the number of newly diagnosed cases has increased tremendously. The rates of PCa have increased so dramatically over the last decade that the age-adjusted incidence rate of PCa is no greater than that for any other cancer among men in the United States. Although PCa rates have risen steadily since 1973, there has been a dramatic acceleration in the late 1980s which has been associated with the introduction and use of PSA for screening and early detection. There is now some evidence that the rates may be levelling off and even decreasing in some areas. After lung cancer, PCa is the leading cause of death due to cancer in men in the United States. Although PCa can occur in younger men, it is essentially a cancer of elderly men. The highest rates of PCa in the world occur among African-American men in the United States. African-Americans have higher rates than Caucasians at all age levels in the United States, and adjusting for social-economic status does not appear to account for this difference to any appreciable extent. There is no clear reason why PCa rates are so much greater among African-Americans compared with Caucasians in the United States. The reported rates in Africans are substantially lower than those of an African-American, suggesting that environmental factors have an influence on PCa. In spite of this substantial impact on our society. PCa remains a relatively understudied disease, with an essentially unknown etiology. The reviews contained in this book are by no means exhaustive. We have, however, attempted to provide information regarding the pathology of prostate cancer, the status of diagnostic and prognostic markers, as well as a discussion of our understanding of the molecular basis for the disease. The biology of prostate cancer is covered with a discussion on the role of apoptosis in prostate cancer and the suggestion of it being the target for new therapeutic development, as well as the role played by integrins and angiogenesis. Book excerpt: Prostate cancer (PCa) is the most common malignant neoplasm among men in Western countries and the second leading cause of cancer mortalities after lung cancer. It is expected that by 2016, 180,890 men will be diagnosed with prostate cancer and 26,120 will die due to the disease. One of the most frequent complications of prostate cancer in patients is bone metastasis. Osteoblastic bone metastasis is the predominant finding at sites of skeletal metastases. Understanding the pathogenesis of prostate cancer progression and bone metastasis and targeting these processes are very important to control this aggressive disease. The main objectives of this work were to identify the role of signaling pathways that have shown to be upregulated in prostate cancer, evaluate a histone deacetylase inhibitor in treatment of prostate cancer metastasis and develop an osteoblastic PCa model for further understanding the molecular mechanisms of bone metastases. Intensive studies have been performed for better understanding the role of various signaling pathways in PCa progression and metastasis. Importantly, gastrin-releasing peptide receptor (GRPr) signaling was shown to be upregulated in human prostatic intraepithelial neoplasia (PIN), invasive prostatic carcinoma and related skeletal metastases. Our objective in this study was to determine the importance of this signaling in the pathogenesis of PCa progression. By activation of GRPr signaling using bombesin (BBN), there was an increase in proliferation and migration of canine prostate cancer cells. GRPR signaling also modulated the expression level of genes that are implicated in epithelial-mesenchymal transition (EMT), growth, invasion and metastasis of PCa. Histone deacetylase inhibitors (HDACi) are a class of therapeutics that can prevent tumor growth and induce differentiation and apoptosis in a wide group of cancers. AR-42, a HDACi, was shown to be efficient against multiple myeloma, lung cancer and hepatocellular carcinoma. In this study, we evaluated AR-42 as an anti-metastatic drug against canine prostate cancer. AR-42 decreased the mRNA expression level of anoikis resistance and osteomimicry genes in vitro. AR-42 declined the incidence of xenograft bone metastases and tumor growth and increased apoptosis and stemness of metastatic tumors. Osteoblastic bone metastasis is very common finding in PCa patients. During PCa progression, cancer cells acquire osteomimicry properties that facilitate their metastasis and support their proliferation and survival in the bone microenvironment. The pathogenesis of osteoblastic bone metastasis and osteomimicry is still not well understood. For this reason, we developed and characterized a novel canine prostatic cancer cell line (LuMa) that had osteomimicry properties and induced osteoblastic bone metastasis in vivo. The predominant sites of LuMa cells metastasis were bone, brain and adrenal glands.
