Book excerpt: Plants, fungi, and viruses were among the first biological objects studied with an electron microscope. One of the two first instruments built by Siemens was used by Helmut Ruska, a brother of Ernst Ruska, the pioneer in constructing electron microscopes. H. Ruska published numerous papers on different biological objects in 1939. In one of these, the pictures by G. A. Kausche, E. Pfankuch, and H. Ruska of tobacco mosaic virus opened a new age in microscopy. The main problem was then as it still is today, to obtain an appropriate preparation of the specimen for observation in the electron microscope. Beam damage and specimen thickness were the first obstacles to be met. L. Marton in Brussels not only built his own instrument, but also made considerable progress in specimen preparation by introducing the impregnation of samples with heavy metals to obtain useful contrast. His pictures of the bird nest orchid root impregnated with osmium were revolutionary when published in 1934. It is not the place here to recall the different techniques which were developed in the subsequent years to attain the modern knowledge on the fine structure of plant cells and of different plant pathogens. The tremendous progress obtained with tobacco mosaic virus is reflected in the chapter by M. Wurtz on the fine structure of viruses in this Volume. New cytochemical and immunological techniques considerably surpass the morphological information obtained from the pathogens, especially at the host-parasite interface.

Recently several bioengineering and molecular manipulation technologies have been developed to activate the ‘sleeping’ plant innate immune system, which has potential to detect and suppress the development of a wide range of plant pathogens in economically important crop plants. Enhancing disease resistance through altered regulation of plant immunity signaling systems would be durable and publicly acceptable. Strategies for activation and improvement of plant immunity aim at enhancing host’s capability of recognizing invading pathogens, boosting the executive arsenal of plant immunity, and interfering with virulence strategies employed by microbial pathogens. Major advances in our understanding of the molecular basis of plant immunity and of microbial infection strategies have opened new ways for engineering durable resistance in crop plants.

Molecular techniques have accelerated the formulation of short- and long-term strategies of disease management. Exclusion and eradication of plant pathogens by rapid and precise detection and identification of microbial pathogens in symptomatic and asymptomatic plants and planting materials by employing molecular methods has been practiced extensively by quarantines and certification programs with a decisive advantage. Identification of sources of resistance genes, cloning and characterization of desired resistance genes and incorporation of resistance gene(s) into cultivars and transformation of plants with selected gene(s) have been successfully performed by applying appropriate molecular techniques. Induction of resistance in susceptible cultivars by using biotic and abiotic inducers of resistance is a practical proposition for several crops whose resistance levels could not be improved by breeding or transformation procedures. The risks of emergence of pathogen strains less sensitive or resistant to chemicals have been reduced appreciably by rapid identification of resistant strains and monitoring the occurrence of such strains in different geographical locations.

For example, the initial events of adh«sion of fungal spores to the exposed surface tissues of the host are essential for subsequent invasion of the plant or animal and establishment of pathogenesis. A point of consensus among investigators who have directed their attention to such events in plants, insects, and vertebrates isthat spore adhesion to the host cuticle or epithelium is more than a simple binding event. lt is a complex and potentially pivotal process in fungal-plant interactions which "may involve the secretion of ftuids that prepare the infection court for the development of morphological stages of the germling" and subsequent invasion of the host (Nicholson and Epstein, Chapter 1). The attachment of the fungal propagule to the arthropod cuticle is also "mediated by the chemical components present on the outer layer of the spore wall and the epicuticle . . . . Initial attachment may be reinforced further by either the active secretion of adhesive materials or the modification of spore wall materiallocated at the [fungal spore arthropod] cuticle interface (Boucias and Pendland, Chapter 5).

Transgenic plants with engineered genes show promise for effective exploitation of this approach for practical application. Research efforts during the recent years to sequence the whole genomes of the pathogens and plants may lead to development of better ways of manipulating disease resistance mechanisms enabling the grower to achieve higher production levels and the consumer to enjoy safer food and agricultural products. Experimental protocols included in appropriate chapters will be useful for researchers and graduate students.
