Book Conducting Substance Use Research

Download or read book Conducting Substance Use Research written by Audrey L. Begun and published by Oxford University Press. This book was released on 2014-03-05 with total page 210 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book assists new and experienced scholars in planning and conducting high quality, contemporary studies for knowledge building about substance use. The target audience is individuals new to substance use as a field of study, either as novice researchers or as experienced researchers in other areas who find themselves lacking experience to address overlapping issues of substance use. Organized around a translational science framework, the contents address substance use research about epidemiology, etiology, intervention efficacy and effectiveness, and implementation of evidence-informed interventions. In addition, examples and issues are drawn from social work traditions involving multiple levels of study (organisms to large social systems), integrating biopsychosocial aspects, and adopting a lifespan perspective. The authors examine the implications for research of current "great debates" in the field and present readers with a variety of specific substance use research resources and tools. This practical "how to" guide takes the reader step-by-step through issues specific to substance use research in study design, participant recruitment and retention, measurement and analysis, and the processes involved in the dissemination, diffusion, and implementation of evidence-informed innovations. A variety of technical resources and measurement tools are provided, as are references to journals for scholars to consider both as knowledge resources and as outlets for disseminating their work. In sum, this book offers a fresh approach to conducting substance use research that is not readily available in other texts.

Book Behavioral Neurobiology of Alcohol Addiction

Download or read book Behavioral Neurobiology of Alcohol Addiction written by Wolfgang Sommer and published by Springer Science & Business Media. This book was released on 2012-07-28 with total page 721 pages. Available in PDF, EPUB and Kindle. Book excerpt: The question how alcohol alters mood states and why this may end up becoming an addiction is puzzling alcohol researchers since decades. In this volume, an assembly of highly distinguished experts and leaders in alcohol addiction research provides lucid presentations of the current knowledge and research challenges as well as interesting viewpoints on future research directions aimed to stimulate communication and convergence between clinical and preclinical researchers, and to renew interest in the vibrant field of alcohol addiction research among a wide scientifically minded audience. Five Current Topics are discussed in this volume: Neurobiological mechanisms of alcoholism, Genetics, Clinical phenotypes and their preclinical models, Brain imaging, and Translational approaches for treatment development, both pharmacological and non-pharmacological. These areas have in our opinion brought alcohol research substantially forward and influenced our thinking about how to reach our common paramount goal, namely to offer effective treatment solutions for an extensive group of patients with largely unmet medical needs.

Book Translational Examination of Risk related Decision making as an Endophenotype for Alcohol Use Disorders

Download or read book Translational Examination of Risk related Decision making as an Endophenotype for Alcohol Use Disorders written by James Rowan Ashenhurst and published by . This book was released on 2014 with total page 174 pages. Available in PDF, EPUB and Kindle. Book excerpt: Alcohol use disorders, a cluster of symptoms centered around a loss of control over alcohol use resulting in negative consequences on health and well-being, continue to cause worldwide suffering and economic loss. While decades of research have advanced our understanding of the genetic and brain-based causes of alcohol use disorders, the complexity of the symptom cluster and the corresponding intricacy of the implicated neural circuitry complicates the development of interventions. Thus, instead of focusing on the disorder as a whole, psychiatric research has placed research emphasis on intermediate phenotypes, or `endophenotypes' that lie between the genetic basis and the disorder as a whole (Ducci & Goldman, 2008, 2012; D. Goldman, Oroszi, & Ducci, 2005; Manji, Gottesman, & Gould, 2003). A number of endophentypes have been explored, most notably craving or incentive sensitization (Robinson & Berridge, 2001, 1993), affect or mood (Ahmed & Koob, 2005; G.F. Koob & Le Moal, 1997), and aspects of executive function (R. Goldstein & Volkow, 2011) includ- ing impulse control (Jentsch & Taylor, 1999). Related most closely to executive function, this dissertation examines biases in risk-related decision-making as having explanatory value for understanding alcohol dependence at its various stages: from alcohol use initiation, to acquisition of the clinical syndrome, to recovery and abstinence. Data presented herein was drawn from both human clinical populations, and animal models, leveraging a translational approach. First, the neurobiology and genetics of addiction are examined, as is the methods used to measure risk-related decision-making, and how this phenotype might conceptually relate to alcohol use disorders. Next, two analyses of risk-taking data from a clinical and sub- clinical sample demonstrate that, counter to initial predictions, risk-taking as assessed by the Balloon Analogue Risk Task [BART; (C. Lejuez et al., 2002)] is negatively related to clinical symptomatology. The nature of this relationship is further probed, revealing that differences in alcohol problem severity predict differences in loss reactivity, such that those with greater severity take less risk after a big loss than those with lesser severity. Next, the effects of acute and sub-chronic alcohol dosing regimens are explored in an rodent analogue of the BART. Results indicated that acute alcohol dose dependently de- creased risk-taking in the rat-BART, an effect partially consistent with the human literature where alcohol and other drugs are shown to have few effects on behavior in the human task (Peacock, Bruno, Martin, & Carr, 2013). Furthermore, six weeks of sub-chronic admin- istration of alcohol (versus a saline control) did not significantly alter performance of the rat-BART. Suggestions for future studies are explored. Finally, one component of the definition of an endophenotype is that it is heritable, and thus regulated by genes (Ducci & Goldman, 2008, 2012; D. Goldman et al., 2005; Manji et al., 2003). In order to assess the heritability of risk-related decision-making, a panel of inbred strains was phenotyped, revealing that about 55% of the variability in performance is attributable to genetic effects. An attempt is made to identify regions of the rat chromosomes that are linked with rat-BART performance using an F2 intercross strategy and quantitative trait loci (QTL) analysis. Preliminary results indicated a candidate region on Chromosome 1 (between approximately 90.99 Mb and 129.99 Mb). However, these results should be interpreted with caution as higher density trait mapping is required to more narrowly define the QTL region, and this significant result was only found in one of six variants of the rat-BART subject to analyses. In sum, the data presented here, in the context of the broader literature, indicated that future research must address differences in behavioral metrics of risk-related decision-making, as results obtained can be contradictory depending on the task used. Additionally, there are areas where future research is more likely to be clinically influential than others. In particular, more work should be done to examine risk-taking during adolescence, at recovery and during treatment. Interventions at those stages appear most likely to improve outcomes for alcohol dependence.

Book Translational Research Methods in Diabetes Obesity and Nonalcoholic Fatty Liver Disease

Download or read book Translational Research Methods in Diabetes Obesity and Nonalcoholic Fatty Liver Disease written by Andrew J. Krentz and published by Springer. This book was released on 2019-04-02 with total page 556 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book aims to aid the selection of the most appropriate methods for use in early phase (1 and 2) clinical studies of new drugs for diabetes, obesity, non-alcoholic fatty liver disease (NAFLD) and related cardiometabolic disorders. Clinical research methods to assess the pharmacokinetics and pharmacodynamics of new diabetes drugs, e.g. the euglycemic clamp technique, have become well-established in proof-of-mechanism studies. However, selection of the most appropriate techniques is by no means straightforward. Moreover, the application of such methods must conform to the regulatory requirements for new drugs. This book discusses the need for new pharmacotherapies for diabetes, obesity and NAFLD and the molecular targets of drugs currently in development. Emerging technologies including functional imaging, circulating biomarkers and omics are considered together with practical and ethical issues pertaining to early phase clinical trials in subjects with cardiometabolic disorders. Translational Research Methods in Diabetes, Obesity, and Non-Alcoholic Fatty Liver Disease is of interest to biomedical scientists, pharmacologists, academics involved in metabolic research and clinicians practicing in these specialties.

Book Substance Use Disorders Treatment in Therapeutic Communities

Download or read book Substance Use Disorders Treatment in Therapeutic Communities written by Miroslav Horák and published by Springer Nature. This book was released on 2022-10-17 with total page 474 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book presents an in-depth qualitative study carried out with inpatients under treatment for substance use disorders (SUDs) in seven therapeutic communities (TCs) located in three countries: Peru, Nicaragua and Czech Republic. By comparing the experiences in these different cultural contexts, the book presents a grounded theory of SUDs treatment in TCs from a cross-cultural perspective, identifying factors that influence the efficacy of SUDs treatment in TCs based on interviews carried out with inpatients. Based on rigorous qualitative research methods, this book presents not only a comparative analysis of TCs located in different cultural contexts, but also analyzes the cross-cultural nature of the therapeutic programs adopted in these communities, such as the combination of traditional Amazonian medicine based on the therapeutic use of ayahuasca with conventional psychotherapy and occupational therapy, among other approaches. Departing from the interviews carried out with inpatients, the authors present a comparative analysis of how the different TCs address important issues related to SUDs treatment, and complement this analysis with machine-generated summaries of relevant scientific papers. These summaries contain results of similar research projects conducted in other cultural contexts. Substance Use Disorders Treatment in Therapeutic Communities: A Cross-Cultural Approach presents the results of a unique comparative study with great translational potential which will be of interest to both researchers and practitioners working in TCs. This unique comparative study identifies factors affecting the efficacy of therapeutic programs and proposes a grounded theory which aims to serve as an important source of information for therapists and other professionals working with SUDs treatment and for the replication of applied therapeutic methods in other TCs.

Book Genetic Influences on Addiction

Download or read book Genetic Influences on Addiction written by James MacKillop and published by MIT Press. This book was released on 2013-12-13 with total page 397 pages. Available in PDF, EPUB and Kindle. Book excerpt: A comprehensive review of research examining intermediary mechanisms to understand the link between genetic variation and addiction liability. Although there is scientific consensus that genetic factors play a substantial role in an individual's vulnerability to drug or alcohol addiction, specific genetic variables linked to risk or resilience remain elusive. Understanding how genetic factors contribute to addiction may require focusing on intermediary mechanisms, or intermediate phenotypes, that connect genetic variation and risk for addiction. This book offers a comprehensive review of this mechanistic-centered approach and the most promising intermediate phenotypes identified in empirical research. The contributors first consider the most established findings in the field, including variability in drug metabolism, brain electrophysiological profiles, and subjective reactions to direct drug effects; they go on to review highly promising areas such as expectancies, attentional processing, and behavioral economic variables; and finally, they investigate more exploratory approaches, including the differential susceptibility hypothesis and epigenetic modifications. Taken together, the chapters offer a macro-level testing of the hypothesis that these alternative, mechanistic phenotypes can advance the understanding of genetic influences on addiction. The book will be of interest to researchers and practitioners in a range of disciplines, including behavioral genetics, psychology, pharmacology, neuroscience, and sociology.
