Book excerpt: Surface enhanced Raman scattering (SERS) spectroscopy is a sensitive analytical technique, which extends its applications to the detection of substances at low concentration levels. SERS spectroscopy measurements were performed on adenine and rhodamine 6G on silver and gold colloids to evaluate the linearity at low analyte concentrations and at different time intervals. The maximum intensity for adenine was observed at Raman shifts of 728 cm-1 and 1318 cm-1 and the maximum intensity for R6G was observed at Raman shifts of 1352 cm-1 and 1519 cm-1. These SERS measurements were compared with solution absorbance measurements at 650 nm and 900 nm. Raman enhancement factors have also been evaluated for each compound. The results of these studies demonstrate that SERS techniques provide high sensitivity and have the potential to detect a range of compounds in biological samples.

Raman scattering (RS) is a widely used vibrational technique providing highly specific molecular spectral patterns. A severe limitation for the application of this spectroscopic technique lies in the low cross section of RS. Surface-enhanced Raman scattering (SERS) spectroscopy overcomes this problem by 6-11 orders of magnitude enhancement compared with the standard RS for molecules in the close vicinity of certain rough metal surfaces. Thus, SERS combines molecular fingerprint specificity with potential single-molecule sensitivity. Due to the recent development of new SERS-active substrates, labeling and derivatization chemistry as well as new instrumentations, SERS became a very promising tool for many varied applications, including bioanalytical studies and sensing. Both intrinsic and extrinsic SERS biosensing schemes have been employed to detect and identify small molecules, nucleic acids and proteins, and also for cellular and in vivo sensing.

Book excerpt: This volume contains the proceedings of the NATO-Advanced Study Institute on the "Spectroscopy of Biological Molecules", which took place on July 4-15, 1983 in Acquafredda di Maratea, Italy. The institute concentrated on three main subiects: the structure and dymanics of DNA, proteins, and visual and plant pigments. Its timeliness has been linked to rapid advances in certain spectroscopic techniques which yielded a consider able amount of new information on the structure and inter actions of biologically important molecules. Among these techniques Fourier transform infrared, resonance and surface enhanced 'Raman spectroscopies, Raman microscopy and micro probing, time resolved techniques, two photon and ultrafast electronic, and C-13, N-15 and P-31 NMR spectroscopies and kinetic and static IR difference spectroscopy receiced a great deal of attention at the Institute. In addition, an entirely new technique, near-millimeter-wave spectroscopy has been presented and discussed. Two introductory quantum chemical lectures, one on the structure of water in DNA, and another pn the energy bands in DNA and proteins set the stage for the experimentally oriented lectures that followed. Fundamental knowledge on hydrogen bonding was the topic of two other lectures. Panel discussions were held on the structure and confor mations of DNA, metal-DNA adducts and proteins and on visual pigments. Many scientists who normally attend different conferences and never meet, met at Aquafredda di Maratea. We feel, that at the end of our Institute a synthetic vi~w emerged on the powerful spectroscopic and theoretical methods which are now available for the study of biological molecules.

Book excerpt: Herein analysis of dye molecules has been carried out by means of surface enhanced Raman spectroscopy (SERS) and/or surface enhanced resonance Raman spectroscopy (SERRS) using citrate- and/or borohydride-reduced silver colloids employing laser exciting wavelengths equal to 514.5 and/or 632.8 nm. SERS and/or SERRS spectra are reported using, as model system probes, eight dye molecules which belong to several distinct chemical structural classes. Experimental protocols were developed and subsequently modified, as required, for each dye molecule examined. Vibrational spectroscopic profiles were obtained, where possible, with respect to concentration and pH dependence. SERS and/or SERRS vibrational bands provided unique fingerprint spectra for each dye molecule. In an attempt to develop novel applications of SERRS the technique has been used, in a kinetic investigation, to monitor and analyse the synthesis of the dye indigo carmine from indigo using a silver sol as the SERRS substrate/medium. In another study it was possible to differentiate between two structurally similar anthraquinones, alizarin and purpurin, using SERRS. It was also possible to demonstrate the existence of multiple molecular species of certain dye molecules, as a function of pH e.g. nuclear fast red, metanil yellow, purpurin and alizarin. For some dye molecules e.g. alcian blue it was possible to combine the linear regions of normal (non-resonance/non-enhanced) Raman and SERS/SERRS plots, thereby extending the dynamic range available for semi-quantitative analysis. The sensitivity of the SERS/SERRS technique for semi-quantitative trace analysis of eight dye molecules has been successfully demonstrated.
