Book excerpt: The multipotent differentiation potential of human mesenchymal stem cells (hMSCs) have been vastly characterized and investigated. These cells are capable of forming somatic adult tissues such as bone, cartilage and fat making these cells a potential source for regenerative therapies. Tissue engineering utilizes these cells and biocompatible scaffolds to create such tissues including bone, cartilage, and fat. A series of experiments were conducted to optimize cytocompatibility of biomaterial scaffolds, osteogenesis of hSMCs following chemical stimulus, and chondrogenesis in monolayer in efforts to create a bone and cartilage, condylar construct shaped as the human proximal tibia. This construct was seeded with pre-differentiated hMSCs and subsequently implanted in vivo. Furthermore, adipogenesis of hMSCs was investigated and tissue engineered constructs were created then subjected to a novel magnetic resonance technique to obtain mechanical properties. Conventional MR imaging cannot measure shear stiffness of adipose tissue and monitor development of engineered constructs, however, microscopic magnetic resonance elastography (muMRE) has this capability. Results indicate a decrease in measured shear stiffness as constructs were cultured in vitro suggesting adipose tissue development and lipid synthesis.
