Book excerpt: Saccharomyces cerevisiae strains null for snf3 are unable to grow on low concentrations of glucose. Several suppressors of the snf3 null have been identified that allow growth on low glucose. Suppression occurs upon overexpression of SKS1 kinase, a protein of unknown function. Previous evidence indicates that SKS1 overexpression restores glucose transport by activating specifically the transporter HXT2. The mechanism developed here proposes that this relief of repression of HXT2 due to the loss of the positive signal generated by Snf3 is accomplished via the titration of the negative regulatory protein Mth1. Due to the composition of its promoter region, HXT2 is the transporter most susceptible to loss of repression. This study also further elucidated the role of SKS1 in suppression. SKS1 was able to suppress a dominant mutation of MTH1 that is not suppressible by loss of the co-repressor. This observation suggests a direct action of Sks1 on Mth1. This work helps to build our understanding of how individual HXT genes are regulated.
