Book excerpt: Plasmodium falciparum virulence proteins and molecules required for assembly of the knobs used for cytoadherence are exported through the Maurer's clefts which are formed in the erythrocyte cytoplasm immediately following merozoite invasion into the erythrocyte host cell. Proteins participating in knob formation, cytoadherence or immune evasion include variant-surface antigens (VSAs) that are trafficked alone or chaperoned by other parasite proteins through the Maurer's clefts. In this study, recombinant PfMC-2TM was expressed using HeLa based in vitro human cell free expression system. Further, the stage specific expression, trafficking, solubility and topology of endogenous PfMC-2TM in the parasite and erythrocyte cytoplasm was investigated in 3D7 and FC-27 strains of P. falciparum. Detailed cell biological and biochemical characterizations were performed to identify the mode of PfMC-2TM export from the parasite to erythrocyte cytoplasm. Following Brefeldin A (BFA) treatment of parasites, PfMC-2TM traffic was evaluated using immunofluorescence and western blotting with antibodies reactive with PfMC-2TM. Our data shows that PfMC-2TM is sensitive to BFA treatment in the ring and trophozoite stage. Permeabilization of infected erythrocytes with streptolysin O (SLO) and saponin, showed that the N and C-termini of PfMC-2TM are exposed to the erythrocyte cytoplasm with the central portion of the protein protected in the MC membranes. PfMC-2TM was expressed as early as 4 h post invasion (hpi), was tightly colocalized with Maurer's cleft resident protein REX-1 and trafficked to the erythrocyte membrane without a change in solubility. PfMC-2TM associates with the infected erythrocyte membrane as an integral membrane protein and is seen to be transiently exposed on the infected red blood cell surface. PfMC-2TM remains insoluble upon association with the MC and erythrocyte membrane, suggestive of protein-lipid interactions with membranes of the MC and erythrocyte. PfMC-2TM is an additional marker of the nascent MCs, that can be used as a clinical marker of early blood stage infection.

Book excerpt: Rhoptry proteins participate in the invasion of red blood cells by merozoites during the malaria parasite's asexual-stage cycle. Interference with the rhoptry protein function has been shown to prevent invasion, and three rhoptry proteins have been suggested as potential components of a vaccine against malaria. Rhoptry associated membrane antigen (RAMA) is a 170-kDa protein of Plasmodium falciparum which is processed to a 60-kDa mature form in the rhoptries. p60/RAMA is discharged from rhoptries of free merozoites and binds to the red-cell membrane before being internalized to form part of the parasitophorous vacuole of the newly developing ring. We examined the range of anti-RAMA responses in individuals living in an area of endemicity for malaria and determined its association with clinical immunity. RAMA is immunogenic during infections, and at least three epitopes within RAMA are recognized by hyperimmune sera in immunoblots. Sera from individuals living in a region of Vietnam where malaria is endemic possessed strong antibody responses toward two C-terminal regions of RAMA. Cytophilic antibody isotypes (immunoglobulin G1 [IgG1] and IgG3) predominated in humoral responses to both C-terminal epitopes. Acute episodes of P. falciparum infection result in significant boosting of levels of antibody to an epitope at the extreme C terminus of RAMA that harbors the red-cell-binding domain. Immunity to P. falciparum infection was linked to elevated levels of IgG3 responses to this functional domain of RAMA, suggesting that the region may contain a protective epitope useful for inclusion in a multiepitope vaccine against malaria.

Book excerpt: A method for fixation of Plasmodium falciparum infected erythrocytes and solubilization of the erythrocyte membrane with detergent was developed. This method was applied to two-color flow cytometric analysis of both intraerythrocytic (IE) malaria DNA and parasite-derived antigen on the erythrocyte surface membrane. Infected erythrocytes were fixed with 0.025% glutaraldehyde followed by treatment with 1% saponin to gain access to intramembranous components and allow antibody to interact with antigen. DNA of IE parasite was subsequently stained with propidium iodide. Using this procedure cell morphology was well preserved with excellent parasite DNA staining. Using anti-malaria antibodies which recognize ring-infected erythrocyte sur antigen (Pfl55/RESA), we observed that glutaraldehyde-fixed saponin treated infected erythrocytes exhibited a variable immunofluorescence intensity as assessed by both flow cytometry and fluorescence microscopy. Ring-infected cells displayed strong immunofluorescence staining, whereas a weak signal was detected on cells containing schizonts. Simultaneous measurement of parasite DNA and antigen in the infected erythrocyte membrane can facilitate the study of antigen expression in the cell membrane in association with development of IE parasites.
