It begins with an introduction to the organization and physiology of the nervous system, especially of the peripheral nervous system with its high degree of plasticity. Subsequent chapters discuss the interaction between peripheral nerves and tumor cells, the so-called neuro-neoplastic synapse, with regard to carcinogenesis, predictive tumor markers, tumor growth and tumor progression leading to metastasis. In this part, the concept of neoneurogenesis is postulated as a process by which the tumor regulates its own innervation through the release of neurotrophic factors, in analogy to the process of neoangiogenesis discovered three decades ago. Once nerve endings have grown into the tumor, these can release neurotransmitters which promote tumor cell migration and metastasis development. The final chapters reflect on the role of stem cells in neoneurogenesis and consider pharmacological approaches for the inhibition of neuro-neoplastic interactions as a basis for new anticancer therapies. Targeted primarily at investigators in experimental and clinical oncology, this book is also of special interest to neurobiologists as well as developmental and cell biologists.

Somehow, I never forgot the professor’s words. When I started working in Dr. Irv Weissman’s labo- tory at Stanford as a postdoctoral fellow, I realized that the findings mentioned by the professor were from Weissman’s laboratory and had been published in a 1988 edition of the journal Science. It has been over 20 years since the publication of that seminal paper, and since then tremendous advances in understanding the biology and maturation of HSCs, namely the process of hematopoiesis, which includes lymphocyte development, have been made. These discoveries were made possible in part by advancements in technology. For example, recent availability of user friendly fluorescence activated cell sorting (FACS) machines and monoclonal an- bodies with a variety of fluorescent labels has allowed more scientists to sort and analyze rare populations in the bone marrow, such as HSCs. All classes of hematopoietic cells are derived from HSCs. Stem cell biology draws enormous attention not only from scientists, but also from ordinary people because of the tremendous potential for development of new therapeutic application to diseases that currently lack any type of effective therapy. Thus, this type of “regenerative medicine” is a relatively new and attractive field in both basic science and clinical medicine.

It has been studied extensively for several decades, yet many basic questions about its regulation, composition and functions remain open. Thus, neither the full impact of AChE’s activities on neuronal development, maintenance and functioning nor the reasons for why and how different AChE molecular forms are being synthesized and degraded are fully known yet, although recent evidence suggests the involvement of micro-RNA regulators in both of these aspects. The identification of the anchoring proteins, PRiMA and ColQ, stimulates studies on AChE’s assembly and its specific localization in the brain and muscle. Additionally, non-cholinergic function(s) of AChE has been proposed as relevant both for neuronal differentiation and cognitive functioning, whereas transgenic engineering, recombinant production and structure-function analyses of this enzyme’s variants with various inhibitors paved new avenues for the development of novel Alzheimer’s disease therapeutics, agricultural pesticides and prophylactics in anticipation of warfare agent exposure. The aims of this Research Topic is to provide a forum for experts in the field to critically discuss recent developments in AChE studies, initiate global critiques of the current views for resolving the fundamental questions in AChE research and promote the development of novel versions and uses of AChE-targeted agents. Topics of interest include but are not limited to: (i) post-transcriptional regulation of AChE; (ii) the non-cholinergic functions of AChE; and (iii) anti-AChE inhibitors and diseases.
