Its content is based in part on the lecture outlines and selected histopathology laboratory components of an advanced course entitled The Pathobiology of Experimental Animal and Human Neoplasia, developed by me for the Experimental Pathology Curriculum of the Department of Pathology at the Medical College of Virginia. In this regard, an effort has been made to integrate pathology with carcinogenesis, genetics, biochemistry, and cellular and molecular biology in order to present a comprehensive and current view of the neoplastic process. For focus, emphasis is mainly being placed on the neoplastic cells themselves, and not on associated host-mediated responses. It ishoped that this book will accomplish its purpose of providing students and researchers who already possess strong but diverse basic science backgrounds with unifying concepts in tumor pathobiology, so as to stimulate new research aimed at furthering our understanding of neoplastic disease. He was a versatile, internationally renowned biochemist, gifted with bound less energy and a great breadth of view. After early work on respiratory enzymes, he developed a lifelong in te rest in complex carbohydrates and, particularly, glycoproteins. He early recognized the important role of the latter substances in membrane function and developed the now widely accepted model for the structure of major erythrocyte membrane glyco pro teins. He expanded this model to include the possible general roles of membrane glycoproteins in specific membrane functions, particularly cell recognition phenomena. Richard Winzler possessed a broad and critical perception of the ftux of biomedical progress. He thus appreciated very early that general recognition of the complexity of membrane-associated phenomena would foster a new, rapidly expanding, interdisciplinary membrane technology. He also recognized the need for means to bridge the communication barriers inevitably genera ted by such an evolution, and this conviction largely stimulated us to write this book. Membrane technology is a vast field and we cannot treat all its aspects. Instead, we concentrate on those areas that, in our view, have evolved most rapidly. In this, we pay primary but not exclusive attention to the plasma membrane, but where relevant, consider other biomem branes and diverse model systems. Moreover, we focus on animal cells, but include information derived from microbial and artificial membranes where germane.

It includes chapters that address i) the theoretical basis of investigations of transport processes across biological membranes, ii) some of the experimental operations often used by scientists in this field, iii) chemical and biological properties common to most biological membranes, and iv) planar thin lipid bilayers as models for biological membranes. The themes developed in these chapters recur frequently throughout the entire series. Transport of molecules across biological membranes is a special case of diffu sion and convection in liquids. The conceptual frame of reference used by investigators in this field derives, in large part, from theories of such processes in homogeneous phases. Examples of the application of such theories to transport across biological membranes are found in Chapters 2 and 4 of this Volume. In Chapter 2, Sten-Knudsen emphasizes a statistical and molecular approach while, in Chapter 4 Sauer makes heavy use of the thermodynamics of irreversi ble processes. Taken together, these contributions introduce the reader to the two sets of ideas which have dominated the thinking of scientists working in this field. Theoretical consideration of a more special character are also included in several other Chapters in Volume I. For example, Ussing (Chapter 3) re-works the flux ratio equation which he introduced into the field of transport across biological membranes in 1949.

The bridge was enormous and was made all of metal, and the painter's job was to keep it from rusting. He would start at one end and slowly proceed, day by day, month by month, toward the other end, painting the bridge. But no sooner would he finish with the painting than the bridge would begin to rust again. The rust, too, would start at one end and slowly proceed toward the other end, systematically destroying the painter's endeavor. And so the painter would return to where he had started, and begin painting again, slowly proceeding toward the other end of the bridge, always just one step ahead of the rust. And if the story is true, the painter might still be painting that bridge-a modern Sisyphus! During the writing of this book, the story of the painter and his bridge kept coming to mind. The field the book covers has been developing so rapidly that, like the painter, I too had to return to where I had started and fight the rust of obsolescence. But unlike the painter, I had a deadline to meet, which constituted a point of no return. And so, sending off this manuscript, I have no choice but to watch the fruits of my endeavor be overtaken by the rust.
