Book The Discovery of the Cellular Sodium Pump in the 20th Century

Download or read book The Discovery of the Cellular Sodium Pump in the 20th Century written by Beatrice Maier Anner and published by GRIN Verlag. This book was released on 2024-02-26 with total page 49 pages. Available in PDF, EPUB and Kindle. Book excerpt: Document from the year 2024 in the subject Chemistry - Bio-chemistry, , language: English, abstract: During her doctoral and postdoctoral studies, Beatrice Maier Anner became familiar with a fascinating biological system, called sodium pump by the physiologists and Na,K-ATPase by the biochemists; it is a sort of biomotor located in the cell membrane, which constantly expulses sodium (Na) ions from the cell interior and replaces them by potassium (K) ions, at the expense of chemical energy provided by adenosine-triphosphate (ATP) which is cleaved to adenosine-diphosphate (ADP) by the Na,K-ATPase, which gets its pump energy from the cleavage of this chemical bond. There would be no life without this fundamental biological system. Interestingly, it is also a pharmacological receptor, i.e. compounds extracted from the Digitalis plant of from the toad bind to the pump and block it, hence the toxicity of e.g. digitoxin or the toad's bufotoxins. The author has acquired extensive practical and theoretical knowledge of this biological system, acquired at the Baylor College of Medicine, Houston, TX, USA consisting in inserting purified functional Na,K-ATPase into artificial membranes (liposomes) in order to study the pump activity on a molecular level. The present book is a unique historical overview from the discovery and purification of the sodium pump to its reconstitution in artificial membranes in the 20th century. This short book permits to become rapidly familiar with this fascinating biomotor and to find unique historical sources.

Book Reconstituted Sodium Pump

Download or read book Reconstituted Sodium Pump written by Beatrice Maier Anner and published by GRIN Verlag. This book was released on 2024-04-24 with total page 62 pages. Available in PDF, EPUB and Kindle. Book excerpt: Document from the year 2024 in the subject Chemistry - Bio-chemistry, , language: English, abstract: The sodium pump, also called Na,K-ATPase (NKA) is a biomotor inserted in the membrane of all living cells; it extrudes sodium ions from the cell interior and replaces them with potassium ions. This is a fundamental and vital process. Furthermore, the sodium pump is also a receptor for pharmacological and toxicological compounds, such as the heart drug digitoxin and the arrow poison ouabain. Around 1974 it became possible to extract the sodium pump from the cell membrane and insert it into artificial phospholipid vesicles where it worked in the same way as in the cell. Now, the pump could be studied in its pure form on a molecular level. The author has been involved in this pioneering research and tells some fascinating stories concerning the first ultrastructural description of the reconstituted sodium pump in collaboration with specialists at Arhus University in Denmark and at Duke University in the US, and about the use of the reconstituted sodium pump for testing an unknown inhibitory compound in collaboration with Harvard Medical School.

Available in PDF, EPUB and Kindle. Book excerpt: The sodium of animal cell membranes converts the chemical energy obtained from the hydrolysis of adenosine 5' -triphosphate into a movement of the cations Na + and K + against an electrochemical gradient. The gradient is used subse quently as an energy source to drive the uptake of metabolic substrates in polar epithelial cells and to use it for purposes of communications in excitable cells. The biological importance of the sodium pump is evident from the fact that be tween 20-70% of the cell's metabolic energy is consumed for the pumping pro cess. Moreover, the sodium pump is an important biological system involved in regulatory processes like the maintenance of the cells' and organism's water me tabolism. It is therefore understandable that special cellular demands are han dled better by special isoforms of the sodium pump, that the expression of the sodium pump and their isoforms is regulated by hormones as is the activity of the sodium pump via hormone-regulated protein kinases. Additionally, the sodium pump itself seems to be a receptor for a putative new group of hormones, the endogenous digitalis-like substances, which still have to be defined in most cases in their structure. This group of substances has its chemically well known coun terpart in steroids from plant and toad origin which are generally known as "car diac glycosides". They are in medical use since at least 200 years in medicine in the treatment of heart diseases.

Book Transport And Diffusion Across Cell Membranes

Download or read book Transport And Diffusion Across Cell Membranes written by Wilfred Stein and published by Elsevier. This book was released on 2012-12-02 with total page 704 pages. Available in PDF, EPUB and Kindle. Book excerpt: Transport and Diffusion across Cell Membranes is a comprehensive treatment of the transport and diffusion of molecules and ions across cell membranes. This book shows that the same kinetic equations (with appropriate modification) can describe all the specialized membrane transport systems: the pores, the carriers, and the two classes of pumps. The kinetic formalism is developed step by step and the features that make a system effective in carrying out its biological role are highlighted. This book is organized into six chapters and begins with an introduction to the structure and dynamics of cell membranes, followed by a discussion on how the membrane acts as a barrier to the transmembrane diffusion of molecules and ions. The following chapters focus on the role of the membrane's protein components in facilitating transmembrane diffusion of specific molecules and ions, measurements of diffusion through pores and the kinetics of diffusion, and the structure of such pores and their biological regulation. This book methodically introduces the reader to the carriers of cell membranes, the kinetics of facilitated diffusion, and cotransport systems. The primary active transport systems are considered, emphasizing the pumping of an ion (sodium, potassium, calcium, or proton) against its electrochemical gradient during the coupled progress of a chemical reaction while a conformational change of the pump enzyme takes place. This book is of interest to advanced undergraduate students, as well as to graduate students and researchers in biochemistry, physiology, pharmacology, and biophysics.

Book Liposomes as Tools in Basic Research and Industry 1994

Download or read book Liposomes as Tools in Basic Research and Industry 1994 written by Jean R. Philippot and published by CRC Press. This book was released on 2017-11-22 with total page 286 pages. Available in PDF, EPUB and Kindle. Book excerpt: This book is devoted to a broader understanding of liposomes as a versatile tool used in many domains, including basic research and applied technology, focusing on less common applications and recent developments. Over the past few years, new types of liposomes made of nonphospholipid molecules have opened new perspectives in applications. These lipid vesicles, already used in cosmetology, are being manufactured for industrial and agricultural uses. However, "Stealth" liposomes, pH-sensitive liposomes, and cationic liposomes have enlarged and improved the application field of liposomes in clinical research. The book covers these different uses of liposomes with particular attention to new formulations and new applications.

Book Regulation of Potassium Transport Across Biological Membranes

Download or read book Regulation of Potassium Transport Across Biological Membranes written by Luis Reuss and published by . This book was released on 1990 with total page 520 pages. Available in PDF, EPUB and Kindle. Book excerpt: Potassium is quantitatively the main cation of the intracellular fluid compartment and, therefore, plays major roles in the maintenance and regulation of cell volume. In addition, potassium is thought to participate in the regulation of a number of functions common to most or all cells, including differentiation and growth. Finally, transport of potassium ions across the cell membranes influences not only the intracellular content (and chemically activity) of the cation but also the extracellular concentration, particularly in the case of tissues possessing extracellular compartments in which ionic diffusion and blood-flow mediated extraction are restricted. This is of paramount importance in excitable tissues, in which the potassium concentration gradient across the cell membrane is the main determinant of the membrane voltage and, hence, of automatism and/or excitability.
