Book excerpt: Genetic Control of Immune Responsiveness: Relationship to Disease Susceptibility documents the proceedings of an international conference held at Brook Lodge, Augusta, Michigan, May 8-10, 1972. It brings together the detailed experimental evidence on the mechanism of action of specific immune response genes, and shows how the two major classes of immune response genes affect our understanding of basic immunology and antibody formation on the one hand, and of host factors determining disease susceptibility on the other. The book is organized into seven parts that correspond to the seven sessions of the conference. Part I presents a session on how the study of the genetic control of immune responses in guinea pigs and mice has contributed to the development of T cell immunology. Part II presents a session on genetic fine structure of the major (H-2) histocompatibility complex in the mouse. Part III presents a session dealing with the association between the production of homogeneous antibody, or of idiotypic antibody, and immunoglobulin allotypes. Part IV presents a session on the relationship between histocompatibility-linked and allotype-linked immune response genes and antigen-specific receptors on T and B cells. Part V presents a session on animal models of multigenic control of susceptibility to disease. Part VI presents a session on the associations between HL-A type and specific disease entities. Finally, Part VII provides a commentary on the ideas and concepts emerging from the three days of free-ranging discussion.

Book excerpt: The major histocompatibility complex (MHC) is a highly polymorphic and diverse multigene locus in all jawed vertebrate species that has an integral role in adaptive/innate immune systems, transplantation, and infectious and autoimmune diseases. The MHC supra-locus in mammalian vertebrates is usually partitioned into three distinct regions, known as classes I, II, and III, which, to varying extents, can be found conserved in nonmammalian jawed vertebrates, such as bony fish, amphibians, and bird lineages. The MHC gene region is characterized particularly by the expression of class I and class II glycoproteins that bind peptides derived from intracellular or extracellular antigens to circulating T-cells. While this expressed antigenic specificity remains the predominant interest with respect to MHC function and polymorphism in a population, a broader concept has emerged that examines the MHC as a multifunctional polymorphic controller that facilitates and regulates genome diversity with a much greater array of functions and effects than just MHC-restricted antigen recognition. This volume of 19 reprints presented by various experts and collected from the Special Issue of Cells on “MHC in Health and Disease” covers a broad range of topics on the genomic diversity of the MHC regulatory system in various vertebrate species, including MHC class I, II, and III genes; innate and adaptive immunity; neurology; transplantation; haplotypes; infectious and autoimmune diseases; fecundity; conservation; allelic lineages; and evolution. Taken together, these articles demonstrate the immense complexity and diversity of the MHC structure and function within and between different vertebrate species.

Thus, the serologic information summarized in this volume is essential for optimal patient care. At the same time, recombinant DNA technology has led to a revolution in our understanding of many aspects of basic biology. Among the advances has been the initial characterization of the structure of some HLA loci. While this will ultimately improve clinical services, constant reference to serologic data is essential so that the powerful new techniques can be applied in the most effective ways. The title of the seminar was "Reducing the costs of disease by improving resistance through genetics". The seminar was held as an activity of the Community Programme for the Coordination of Agricultural Research, 1984-1988. Costs of disease depend on losses caused by morbidity, mortality and production decreases and on the costs of preventive measures including vaccination and medication. Production losses often contribute a major portion to the total costs. To reduce costs of disease preventive measures like vaccination, preventive medication and hygienic procedures are applied. Genetic resistance is an attractive preventive measure because of its consistent nature in the next generations, because it precludes veterinary services and because there are no side-effects. Constraints are the long term investment, relatively slow progress per generation (in combination with production traits) and the considerable lack of knowledge about inheritance of resistance mechanisms in farm animals.
