Both his father's and mother's ancestors came over to join the Massachusetts Colony of the Pilgrims about 1650-1670. Among their descendants eighteen were ministers or ministers' wives. His father, Jacob Chamberlain, "a county surveyor, justice of the peace and well-to-do farmer, belonged to a family of religious men, church deacons and leading men in local affairs." He is described by his sou Jacob as"a man of strong faith and active Christian character, a consistent member of the Church for seventy years, thirty-one in Sharon and thirty-nine at Hudson, Ohio. He was always one of the active working members and, according to his means, one of the most liberal supporters of the Gospel at home and abroad. He died in 1878, at the age of eighty-six."
