Book excerpt: Excerpt from The Journal of Hygiene, 1903, Vol. 3 In the course of a bacteriological examination of river waters during the autumn of 1901, an opportunity occurred for studying an unusually large series of microorganisms. The material derived from comparing a considerable number of cultures - 543 - is so suggestive that, although the work is not yet complete, it may be of advantage to other workers if the general character of the results be indicated at this time. The cultures were isolated from three different rivers, the Illinois, the Mississippi, and the Missouri. The nature of the collecting stations may be briefly described. 1. The Illinois River at Averyville. As is well known the Illinois River receives a large part of the sewage of Chicago, but by the time the flow reaches Averyville (130 miles below Lockport, the point where the Chicago drainage canal discharges into the Desplaines River), oxidation of the organic matter is practically complete and the bacterial content is not high. The mean monthly discharge of the river at this point was about 7000 cubic feet per second during Oct. - Dec., 1900. Book excerpt: Excerpt from The American Journal of Hygiene, 1922, Vol. 2 In our routine procedure the soil was thoroughly broken up be fore being emptied into the sieves. Precautions should be taken not to carry over a little of the soil from one sample to another, and to keep the bottom of the sieve from contact with any dirt. The water used in the funnels was usually heated to a temperature of about 115° F. A funnel, after being set up, was filled within an inch and a half of the rim, so that when the sieve was placed in position there would be no contact between the water and the bottom of the sieve. Water of the same temperature was then poured over the rim of the funnel, until it was practically full and the level of the water almost an inch above the bottom of the sieve. The samples were usually set up in the isolating apparatus some time during the day and left stand ing over night before the examinations were made. One method of drawing off the samples of water for microscopical examination was to attach a second clamp higher on the tube and to allow the water between the two clamps to run Off into a 50 cc. Centrifuge tube (see Fig. A simpler method is to slightly Open the clamp at the bottom of the rubber tube and let the water run out until the centri fuge tube is filled. After centrifugation the supernatant water was drawn Off with a long pipette and the residue spread out on a two-by three-inch slide, for microscopical examination: In some instances, when the soil sample was of saturated clay or very finely divided sand, a considerable amount of sediment would fall into the funnel. This could, if necessary, be reduced in amount by running it through a smaller isolating appartus before microscopical examination. Book excerpt: Excerpt from The Journal of Hygiene, 1912, Vol. 11 The present paper contains the results of an enquiry into the differentiation of certain organisms of varying source, which present characters more or less closely allied to those of the mannite or Flexner type of the dysentery bacillus. The non-mannite type, first discovered by Shiga in 1898, as the cause of certain forms of bacillary dysentery in Japan, and later by Kruse in 1901, as the cause of epidemic dysentery in Rhenish West phalia, has been shown to remain remarkably true to type, and our present cultural and serological methods amply suffice to separate it off clearly from the mannite or Flexner type. We are, in fact, justified in speaking of the Shiga bacillus of Dysentery. It is other wise with the mannite-fermenting type. The work of the last ten years in various countries has elicited the fact that the bacillus discovered by Flexner in Manila in 1901 must be regarded merely as a type of a group comprising an ever-increasing number of strains differing in certain properties from the type-strain, and from each other. The members, or sub-types, of the mannite-fermenting group, which have been most thoroughly investigated are Flexner's bacillus, the Y bacillus of Hiss and Russell discovered in 1904, and the bacillus of Strong. I-X (1901-1910) After this work was continued in Cambridge on normal birds which reached us alive, and on sickly birds caught alive, immediately killed and packed in ice and sent to us from time to time from various moors as occasion offered. During the visits to Scotland it was, of course, not possible to carry out all the precautionary measures which are described later as having been taken when working in our own laboratories at Cambridge, but the most important of these precautions were observed, such, for example, as plucking the birds before they were brought into the laboratory, the free use of the flame for singeing, and of the actual cautery for destroying any stray particles of feather and for burning the skin through which the incisions were made. In the preliminary experiments also the method of getting at the lungs from behind, which is described later, was adopted; and emulsions of the organs were made between sterilised plates, but the glass frame, in which this was done at Cambridge, had not been adopted at the time of the earlier experiments. One of the first objects of the experiments was to seek for Klein's bacillus and to compare it with other members of the colon group in the light of the great advances in bacteriology since Klein's observations were made 18 years ago.
